High hydrostatic pressure is a non-thermal processing technique for extracting functional compounds from herbal *** study aims to investigate the effectiveness of the combination of high hydrostatic pressure(HHP),enzymatic hydrolysis,and sonication for extracting wild-simulated ginseng(WSG)to improve yields of major and minor *** using LC-MS/MS analysis,the total ginsenoside yield was enhanced by 25.62%by increasing the pressure of HHP and sonication treatment(HHP-soni)extraction from 100 to 300 ***,compared to HHP-soni at 100 MPa,adding two commercial food-grade enzymes(Viscozyme and Pectinex)resulted in a 1.23-1.43-fold increase in total ginsenoside contents,as well as a 1.09-1.95-fold increase in the contents of seven major ***,HHP-soni with enzymes increased the yields of five minor ginsenosides(Ro,Rh1,F2,CO,and Rg3)by 1.76-30.24 times compared to extraction with Pectinex at 0.1 *** compound K and protopanaxadiol were detected after extraction using Pectinex with HHP-soni,but not after extraction using Pectinex and sonication at 0.1 *** the contents of the stereoisomers 20(S)-Rg3 and 20(R)-Rg3 were similar after extraction at pressures of 100 and 200 MPa,the 20(S)-Rg3 content was 1.87 times higher than the 20(R)-Rg3 content after HHP treatment at 300 *** the analytical data,we inferred the hydrolytic pathways of the major to minor ginsenosides,and specifically the ginsenoside Rb1 standard,when using Pectinex under experimental *** findings suggest that the combination process of HHP,sonication,and Pectinex treatment could improve the quality of WSG.

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

Objective： To investigate whether the dried root of Phellodendron amurense Ruprecht（Phellodendri cortex; PC） extract improves arthritic symptoms through anti-inflammatory and immune-modulatory effects in collagen-induced arthritis in mice. Methods： Rheumatoid arthritis（RA） was induced in male DBA/1 mice by immunization with type Ⅱ collagen（ColⅡ）. CIA mice were divided into 5 groups（n=10 per a group） with normal, CIA control, PC extract（50 mg/kg and 100 mg/kg）-treated, and meloxicam（50 mg/kg）-treated as the reference drug. The PC extract or meloxicam were administered orally in CIA mice once a day for 14 days after arthritis induction. Arthritic score, levels of anti-ColⅡ IgG2a antibody, prostaglandin E2（PGE2）, tumor necrosis factor（TNF）-α, and interleukin（IL）-17 in the sera of CIA mice were measured. Histopathological changes in the ankle joints of CIA mice were also analyzed by staining with hematoxylin and eosin（H and E）, safranin-O and immunohistochemistry using anti-TNF-α and anti-IL-17 antibodies. Results： The arthritic score was increased in CIA mice in a time-dependent manner, as were the serum levels of anti-ColⅡ IgG2a antibody, PGE2, TNF-α, and IL-17. however, the oral administration of PC extract at 50 and 100 mg/kg in CIA mice significantly decreased the arthritic scores, and the serum levels of anti-ColⅡ IgG2a, PGE2, TNF-α, and IL-17 compared with those in the CIA group（P〈0.05 or P〈0.01）. Furthermore, histopathological improvement of the joint architecture in CIA mice was observed after administration of PC extract. PC extract also significantly inhibited the expression of TNF-α and IL-17 in the joints of CIA mice by suppressing the expression of their m RNA and proteins. Conclusion： PC extract may improve the pathological progression of RA through the inhibition of joint destruction by synovial inflammation and immune-stimulation, therefore, it would be a potential anti-arthritic agent in RA.

AIM: To investigate the incidence and distribution of second primary cancers(SPCs) in early gastric cancer(EGC) patients who underwent endoscopic resection(ER), compared to advanced gastric cancer(AGC) patients who underwent ***: The medical records of 1021 gastric cancer(GC) patients were retrospectively reviewed from January 2006 to December 2010. The characteristics and incidence of SPCs were investigated in those with EGC that underwent curative ER(the EGC group) and those with AGC who underwent curative surgical resection(the AGC group).RESULTS: We ultimately enrolled 184 patients in the EGC group and 229 patients in the AGC group. A total of 38 of the 413(9.2%) GC patients had SPCs; the rate was identical in both groups. Of these 38 patients, 18 had synchronous and 20 had metachronous cancers. The most common SPC was lung cancer(18.4%), followed by colorectal cancer(13.2%) and esophageal cancer(13.2%). No significant risk factors were identified for the development of ***: Endoscopists should provide close surveillance and establish follow-up programs to ensure SPC detection in GC patients undergoing curativeresection regardless of their clinical characteristics.

Cu catalysts can convert CO_(2) through an electrochemical reduction reaction into a variety of useful carbon-based ***,this capability provides an obstacle to increasing the selectivity for a single ***,we report a simple fabrication method for a Cu-Pd alloy catalyst for use in a membrane electrode assembly(MEA)-based CO_(2) electrolyzer for the electrochemical CO_(2) reduction reaction(ECRR)with high selectivity for CO *** the composition of the Cu-Pd alloy catalyst was fabricated at 6:4,the selectivity for CO increased and the production of multi-carbon compounds and hydrogen is *** a Cu-Pd alloy catalyst with 6:4 ratio as the cathode of the MEAbased CO_(2) electrolyzer showed a CO faradaic efficiency of 92.8%at 2.4 V_(cell).We assumed that these results contributed from the crystal planes on the surface of the Cu-Pd *** phases of the Cu-Pd alloy catalyst were partially separated through annealing to fabricate a catalyst with high selectivity for CO at low voltage and C_(2)H_4 at high *** results of CO-stripping testing confirmed that when Cu partially separates from the lattice of the Cu-Pd alloy,the desorption of~*CO is suppressed,suggesting that C-C coupling reaction is favored.
