Transglutaminase 2 serves as a pathogenic hub gene of KRAS mutant colon cancer based on integrated analysis

BACKGROUND Colon cancer is acknowledged as one of the most common malignancies worldwide,ranking third in United States regarding incidence and mortality.Notably,approximately 40%of colon cancer cases harbor oncogenic KRAS mutations,resulting in the continuous activation of epidermal growth factor receptor signaling.AIM To investigate the key pathogenic genes in KRAS mutant colon cancer holds considerable importance.METHODS Weighted gene co-expression network analysis,in combination with additional bioinformatics analysis,were conducted to screen the key factors driving the progression of KRAS mutant colon cancer.Meanwhile,various in vitro experiments were also conducted to explore the biological function of transglutaminase 2(TGM2).RESULTS Integrated analysis demonstrated that TGM2 acted as an independent prognostic factor for progression-free survival.Immunohistochemical analysis on tissue microarrays revealed that TGM2 was associated with an elevated probability of perineural invasion in patients with KRAS mutant colon cancer.Additionally,biological roles of the key gene TGM2 was also assessed,suggesting that the downregulation of TGM2 attenuated the proliferation,invasion,and migration of the KRAS mutant colon cancer cell line.CONCLUSION This study underscores the potential significance of TGM2 in the progression of KRAS mutant colon cancer.This insight not only offers a theoretical foundation for therapeutic approaches but also highlights the need for additional clinical trials and fundamental research to support our preliminary findings.

Dear Editor,Genome-wide association studies(GWASs)have reported a genetic association between certain populations and the severity of COVID-19.In severe patients,changes in the locus 3p21.31(including SLC6A20,LZTFL1,CCR9,FYCO1,CXCR6,and XCR1)are associated with COVID-19 hospitalization(Ellinghaus et al.,2020).Moreover,variations in 9p34.2 region(including genes related to the ABO blood grouping)can affect both susceptibility and severity of COVID-19;in particular,individuals with blood group A may have an increased risk of COVID-19 infection,while those with blood group O may have a protective effect(Ellinghaus et al.,2020;Shelton et al.,2021).

Emerging evidence suggested that zinc finger protein 831(ZNF831)was associated with immune activity and stem cell regulation in breast cancer.Whereas,the roles and molec-ular mechanisms of ZNF831 in oncogenesis remain unclear.ZNF831 expression was significantly diminished in breast cancer which was associated with promoter CpG methylation but not mu-tation.Ectopic over-expression of ZNF831 suppressed breast cancer cell proliferation and col-ony formation and promoted apoptosis in vitro,while knockdown of ZNF831 resulted in an opposite phenotype.Anti-proliferation effect of ZNF831 was verified in vivo.Bioinformatic analysis of public databases and transcriptome sequencing both showed that ZNF831 could enhance apoptosis through transcriptional regulation of the JAK/STAT pathway.ChiP and luciferase report assays demonstrated that ZNF831 could directly bind to one specific region of STAT3 promoter and induce the transcriptional inhibition of STAT3.As a result,the attenuation of STAT3 led to a restraint of the transcription of Bcl2 and thus accelerated the apoptotic progression.Augmentation of STAT3 diminished the apoptosis-promoting effect of ZNF831 in breast cancer cell lines.Furthermore,ZNF831 could ameliorate the anti-proliferation effect of capecitabine and gemcitabine in breast cancer cell lines.Our findings demonstrate for the first time that ZNF831 is a novel transcriptional suppressor through inhibiting the expression of STAT3/Bcl2 and promoting the apoptosis process in breast cancer,suggesting ZNF831 as a novel biomarker and potential therapeutic target for breast cancer patients.

Impact wear properties of dissimilar joints between bainitic frogs and pearlite rails

The impact wear behavior and damage mechanism of dissimilar welded joints between U26Mn frog and U75 V rail before and after normalizing treatment were studied by cyclic impact tests.The experiment indicated that the impact wear volume of the joints increased with the increasing number of impact cycles.The main wear mechanisms include pitting wear,mild fatigue wear,delamination wear,and fatigue wear,and plastic deformation was the primary impact wear mechanism.Among them,fatigue wear had the greatest influence on wear volume,while other wear mechanisms had limited effect.The impact wear resistance of the base material was better than that of the heat-affected zone.Normalizing treatment was beneficial to improving the impact wear resistance of welded joints owing to its effect to promote pearlite recovery,grain refinement,and uniform distribution of grains.The martensite generated in the rail welded joints aggravated the impact wear damage to the materials,which should be avoided.

Construction of Naphthalenediimide Lanthanide(Ⅲ)-MOFs and Composites Incorporated Electron-Rich Pyrene Derivative as Multifunctional Fluorescence Sensing for Nitro Aromatic Compounds and Aldehydes

The development of host-guest MOF luminescent composites has attracted considerable attention. However,it is still a challenge to reasonably design large scale conjugated polycarboxylic acids metal-organic framework (MOF) and modulate donor-acceptor interaction. Herein,a series of isostructural 3D porous lanthanide MOF [Ln_(2)(BINDI)0.5(NO_(3))(DMA)(H_(2)O)]∙DMA∙2.5H_(2)O (Ln = La (1),Ce (2),Pr (3) and Nd (4);H_(4)BINDI (N,N’-bis(5-isophthalic acid)-1,4,5,8-naphthalenediimide) were synthesized. Considering the electron-deficient performances of Ln-BINDI MOFs,D-A type composites Ln-MOFs (1—4@H_(4)TBAPy) were prepared via the incorporation of electron-rich H_(4)TBAPy (1,3,6,8-tetrakis(p-benzoic acid) pyrene),which more improved the luminescence performance of complexes 1—4 and can be used as fluorescence sensors for the detection of nitro compounds and aldehydes. High sensitivity of 1@H_(4)TBAPy towards pNBA,pNA PNP,DNP,and TNP could be achieved through hydrogen bond interactions between MOF and analytes,as well as the π-π interaction between H_(4)TBAPy and the naphthalene ring of BINDI,thus the fluorescence quenching efficiency of 1@H_(4)TBAPy was better than that of the complex 1. In addition,it is found that 1@H_(4)TBAPy has high selectivity and sensitivity to aromatic aldehyde SA,5-Mesal and HMBA. Such strategy to enhance the emission of NDI based Ln-BINDI MOFs,will open up an avenue to obtain more fluorescent MOFs for sensing.

An overview on integrated localization and communication towards 6G

While the fifth generation(5 G) cellular system is being deployed worldwide, researchers have started the investigation of the sixth generation(6 G) mobile communication networks. Although the essential requirements and key usage scenarios of 6 G are yet to be defined, it is believed that 6 G should be able to provide intelligent and ubiquitous wireless connectivity with Terabits per second(Tbps) data rate and submillisecond(sub-ms) latency over three-dimensional(3 D) network coverage. To achieve such goals, acquiring accurate location information of the mobile terminals is becoming extremely useful, not only for locationbased services but also for improving wireless communication performance in various ways such as channel estimation, beam alignment, medium access control, routing, and network optimization. On the other hand, the advancement of communication technologies also brings new opportunities to greatly improve the localization performance, as exemplified by the anticipated centimeter-level localization accuracy in 6 G by extremely large-scale multiple-input multiple-output(MIMO) and millimeter wave(mm Wave) technologies.In this regard, a unified study on integrated localization and communication(ILAC) is necessary to unlock the full potential of wireless networks for dual purposes. While there are extensive studies on wireless localization or communications separately, the research on ILAC is still in its infancy. Therefore, this article aims to give a tutorial overview on ILAC towards 6 G wireless networks. After a holistic survey on wireless localization basics, we present the state-of-the-art results on how wireless localization and communication inter-play with each other in various network layers, together with the main architectures and techniques for localization and communication co-design in current two-dimensional(2 D) and future 3 D networks with aerial-ground integration. Finally, we outline some promising future research directions for ILAC.

Interface Design Strategy for GNS/AZ91 Composites with Semi-Coherent Structure

The interfacial structure plays an important role in the mechanical properties of magnesium matrix composite(MMCs)reinforced with graphene nanosheet(GNS)due to their poor wettability with the Mg matrix.An interface design strategy was proposed to form the semi-coherent interfacial structure with superior bonding strength.The lattice mismatch and interfacial bonding strength between Mg/rare earth oxide/carbon were utilized as key characteristics to evaluate the interfacial structure.Lanthanum oxide(La2O3)was selected as the intermediate candidate due to its low lattice mismatch and high interfacial bonding strength.To identify the interfacial structure of Mg/La2O3/graphene,first-principles calculations were conducted to calculate the ideal work of separation and electronic structure of the interfaces.Results demonstrated the presence of strong ionic and covalent interactions at the interface,which theoretically verified the strong interfacial bonding strength among Mg/La2O3/graphene interfaces.To experimentally validate the interface strength,MMCs with the interface structure of Mg/La2O3/GNS were developed.The formation of in-situ La2O3 led to the successful attainment of semi-coherent structures between Mg/La2O3 and La2O3/GNS,resulting in high strength and good ductility of the composite.Overall,this work proposes a new approach to interface design in MMCs with an enhancement of mechanical properties.

In recent years,lithium-sulfur batteries（LSBs）are considered as one of the most promising new generation energies with the advantages of high theoretical speeific capacity of sulfur(1675 mAh·g-1),abundant sulfur resources,and environmental friendliness storage technologies,and they are receiving wide attention from the industry.However,the problems of the shuttle effect and lithium dendrite growth in LSBs have limited their practical application,so there is a need to find ways to solve these problems.It is an excellent choice to use novel materials to modify battery materials.Among those novel materials,the metal-organic framework（MOF） has the properties of regular pores and controllable structure.When applied as a positive electrode and diaphragm,it can restrain the shuttle effect and lithium dendrite growth,especially since it shows excellent performance in diaphragm modification.Therefore,various design strategies and synthesis methods of MOF-modified separators are reviewed in this paper,and the applications of MOF in LSBs separators in different forms are introduced,including the composite of MOF and carbon-based materials,the compounding of MOF and polymer,self-carbonization to form MOF-derived materials.At the same time,different characterization techniques are systematically reviewed to obtain the physical and chemical properties of MOF particles and the working mechanism of MOF-modified diaphragm,which provides a basis for further research in this field.Finally,some future research trends and directions are put forward to fully tap the future commercial potential of MOF-modified diaphragm in LSBs.

What is already known about this topic?The inclusion of meningococcal vaccines in the National Immunization Program(NIP)over several years has significantly reduced the incidence of meningococcal meningitis in China to historic lows.Worldwide,there has been a diversification of meningococcal serogroups,leading to a shift in dominant serogroups in China from serogroup A to serogroups C and B,accompanied by a rise in reports of serogroups Y and W.What is added by this report?An outbreak of serogroup Y Neisseria meningitidis(Nm)in a secondary vocational school involved a single confirmed severe case and 24 individuals with laboratory-confirmed Nm carriage.Epidemiological investigation revealed that the outbreak was localized to the classroom of the confirmed case.Prolonged close contact within a confined space was identified as a significant risk factor for Nm transmission.The genotype sequence identified was type 1655(ST-1655),which is categorized under clonal complex 23(CC-23)and bears resemblance to 8 previously confirmed cases of serogroup Y meningococcal meningitis within Guangdong Province.This suggests that serogroup Y infections continue to sporadically emerge and have become prevalent strains.What are the implications for public health practice?This outbreak underscores the critical need to enhance surveillance of meningococcal serogroups and population carrier,and advocate for vaccination with MenY-containing vaccines.

Lipase and photodecarboxylase coexpression: A potential strategy for alkane-based biodiesel production from natural triglycerides

Alkane-based biodiesel is considered the next generation of biodiesel owing to its potential environmental benefits and the fact that it exhibits much higher specific caloric values than traditional biodiesel.However,the formidable obstacle impeding the commercialization of this cutting-edge fuel alternative lies in the cost associated with its production.In this study,an engineered strain Escherichia coli(E.coli)showcasing harmonized coexpression of a lipase(from Thermomyces lanuginosus lipase,TLL)and a fatty acid photodecarboxylase(from Chlorella variabilis,CvFAP)was first constructed to transform triglycerides into alkanes.The potential of E.coli BL21(DE3)/pRSFDuet-1-TLL-CvFAP for alkane synthesis was evaluated with tripalmitin as a model substrate under various process conditions.Following a comprehensive examination of the reaction parameters,the scope of the biotransformation was expanded to‘real’substrates(vegetable oils).The results showed that bioderived oils can be transformed into alkanes with high yields(0.80-10.20 mmol·L^(-1))under mild conditions(35℃,pH 8.0,and 36 h)and blue light illumination.The selected processes were performed on an increased lab scale(up to 100 ml)with up to 24.77 mmol·L^(-1) tripalmitin,leading to a yield of 18.89 mmol·L^(-1) pentadecane.With the employment of a method for efficiently producing alkanes under mild conditions and a simple procedure to isolate alkanes from the reaction system,the utilization of sustainable biomass as a fundamental feedstock emerges as the primary solution to lower the cost of alkane-based biodiesel.Thus,this study proposes a readily implementable and highly effective approach for alkane-based biodiesel production.
