Enhancer RNAs(eRNAs)are a class of non-coding RNA,which play a critical role in tumor progression.1 Previous studies have indicated abundant CpG methylation,somatic mutation,and copy number variation in eRNA regions in cancers.2,3 We constructed a landscape of genetic alteration-driven eRNAs and provided an integrative pipeline to identify drug candidates that affect eRNA ***,we explored the prognostic value of genetic alteration-driven eRNAs.

This paper constructs a coupled aero-hydro-elastic-servo simulation framework for a monopile offshore wind turbine(OWT).In this framework,a detailed multi-body dynamics model of the monopile OWT including the gearbox,blades,tower and other components(nacelle,hub,bedplate,etc.)has been explicitly *** effects of pile−soil interaction,controller and operational conditions on the turbine dynamic responses are studied systematically in time domain and frequency *** results show that(1)a comprehensive drivetrain model has the capability to provide a more precise representation of the complex dynamic characteristics exhibited by drivetrain components,which can be used as the basis for further study on the dynamic characteristics of the drivetrain.(2)The pile−soil interaction can influence the wind turbine dynamic responses,particularly under the parked condition.(3)The effect of the pile−soil interaction on tower responses is more significant than that on blade responses.(4)The use of the controller can substantially affect the rotor characteristics,which in turn influences the turbine dynamic responses.(5)The tower and blade displacements under the operational condition are much larger than those under the parked *** model and methodology presented in this study demonstrate potential for examining complex dynamic behaviors of the monopile *** ensure accuracy and precision,it is imperative to construct a detailed model of the wind turbine system,while also taking into account simulation efficiency.

The evolution of the microstructure and toughness of APL5L X80 pipeline steel after thermal welding simulation was investigated by X-ray diffraction,electron backscatter diffraction,and transmission electron *** results indicated that primary heat-affected zones can be divided into weld,coarse-grained,fine-grained,intercritical,and sub-critical *** microstructure of the weld zone is mainly composed of bainitic ferrite and a small amount of granular bainite;however,the original austenite grains are distributed in the columnar *** structure of the coarse-grained zone is similar to that of the weld zone,but the original austenite grains are *** contrast,the microstructure in the fine-grained zone is dominated by fine granular bainite,and the effective grain size is only 8.15μm,thus providing the highest toughness in the entire heat-affected *** intercritical and subcritical zones were brittle valley regions,and the microstructure was dominated by granular ***,the martensite-austenite(M/A)constituents are present in island chains along the grain boundaries,and the coarse size of the M/A constituents seriously reduces the *** results of the crack propagation analyzes revealed that high-angle grain boundaries can significantly slow down crack growth and change the crack direction,thereby increasing the material *** impact toughness of the low-temperature tempering zone was equivalent to that of the columnar grain zone,and the impact toughness was between those of the critical and fine-grained zones.

It is a major challenge for the airframe-inlet design of modern combat aircrafts,as the flow and electromagnetic wave propagation in the inlet of stealth aircraft are very *** this study,an aerodynamic/stealth optimization design method for an S-duct inlet is *** upwind scheme is introduced to the aerodynamic adjoint equation to resolve the shock wave and flow *** multilevel fast multipole algorithm(MLFMA)is utilized for the stealth adjoint equation.A dorsal S-duct inlet of flying wing layout is optimized to improve the aerodynamic and stealth *** the aerodynamic and stealth characteristics of the inlet are effectively ***,the optimization results are analyzed,and it shows that the main contradiction between aerodynamic characteristics and stealth characteristics is the centerline and crosssectional *** S-duct is smoothed,and the cross-sectional area is increased to improve the aerodynamic characteristics,while it is completely opposite for the stealth *** radar cross section(RCS)is reduced by phase cancelation for low frequency *** method is suitable for the aerodynamic/stealth design of the aircraft airframe-inlet system.

Clinical experiments have proven that the pedunculopontine nucleus(PPN)plays a crucial role in the modulation of beta oscillations in Parkinson’s disease(PD).here,we propose a new computational framework by introducing the PPN and related synaptic connections to the classic basal ganglia-thalamo-cortical ***,the improved model can not only simulate the basic saturated and beta activities mentioned in previous studies but also produce the normal alpha rhythm that is much closer to physiological ***,the results show that Parkinsonian oscillation activities can be controlled and modulated by the connection strength between the PPN and the globus pallidus internal nucleus(GPi)and the subthalamic nucleus(STN),supporting the fact that PPN is overinhibited in ***,the internal mechanism underlying these state transitions is further explained from the perspective of ***,both deep brain stimulation(DBS)and optogenetic technology are considered effective in terms of abnormal *** when a low-frequency DBS is added to the PPN,beta oscillations can be suppressed,but it is excited again as the DBS’s frequency gradually increases to a larger *** results coincide with the experimental results that low-frequency stimulation of the PPN is effective,and verify the rationality of the ***,we show that optogenetic stimulation of the globus pallidus external(GPe)expressing excitatory channelrhodopsin(ChR2)can effectively inhibit beta oscillations,whereas exciting the STN and PPN has a limited *** results are consistent with experimental reports suggesting that the symptoms of PD’s movement disorder can be alleviated under the GPe-ChR2,but not STN-ChR2,*** the functional role of the PPN and the feasibility of optogenetic stimulation remain to be clinically explored,the results obtained help us understand the mechanisms of beta oscillations in PD.

Proliferation and differentiation of intestinal stem cell(ISC)to replace damaged gut mucosal epithelial cells in inflammatory states is a critical step in ameliorating gut ***,when this disordered proliferation continues,it induces the ISC to enter a cancerous *** gut microbiota on the free surface of the gut mucosal barrier is able to interact with ISC on a sustained ***-biota metabolites are able to regulate the proliferation of gut stem and progenitor cells through transcription factors,while in steady state,differentiated colono-cytes are able to break down such metabolites,thereby protecting stem cells at the gut *** the future,the gut flora and its metabolites mediating the regulation of ISC differentiation will be a potential treatment for enteropathies.

Uncertainty quantification of inverse analysis for geomaterials using probabilistic programming

Uncertainty is an essentially challenging for safe construction and long-term stability of geotechnical *** inverse analysis is commonly utilized to determine the physico-mechanical ***,conventional inverse analysis cannot deal with uncertainty in geotechnical and geological *** this study,a framework was developed to evaluate and quantify uncertainty in inverse analysis based on the reduced-order model(ROM)and probabilistic *** ROM was utilized to capture the mechanical and deformation properties of surrounding rock mass in geomechanical *** programming was employed to evaluate uncertainty during construction in geotechnical engineering.A circular tunnel was then used to illustrate the proposed framework using analytical and numerical *** results show that the geomechanical parameters and associated uncertainty can be properly obtained and the proposed framework can capture the mechanical behaviors under ***,a slope case was employed to demonstrate the performance of the developed *** results prove that the proposed framework provides a scientific,feasible,and effective tool to characterize the properties and physical mechanism of geomaterials under uncertainty in geotechnical engineering problems.
