Effects of low molecular weight heparin on clot rate and activated clotting time： an in vitro study

Background Due to lack of point-of-care testing, the use of low-molecular-weight heparin （LMWH） therapy in some special patients is restricted. This study was designed to explore the effects of LMWH on clot rate （CR） and activated clotting time （ACT）, and to search for an appropriate method for bedside monitoring of anticoagulant activity of LMWH. Methods Thirty-two healthy volunteers were selected from the staff of Beijing Tongren Hospital. CR and ACT were measured with different reagents （glass beads, diatomite, kaolin and magnetic bar） on blood samples spiked with increasing concentrations of LMWH （dalteparin, 0.2-1.8 IU/ml）. Correlations between concentrations of LMWH and values of CR and ACT were analysed based on the data obtained and regression analysis was performed to establish a regression equation. Results With the increase in doses of dalteparin, CR values reduced gradually. The values of CR of four reagents （glass beads, diatomite, kaolin and magnetic bar） were 20.4-4.5 IU/min, 27.4-6.9 IU/min, 27.5-7.9 IU/min and 7.8-0.1 IU/min respectively and an linear relationship was observed between the CR values and dalteparin concentrations （P〈0.05）. The values of ACT were 173-615 seconds, 130-270 seconds, 123-226 seconds, 337-1411 seconds respectively, which showed a linear regression between the ACT values and dalteparin concentrations （P〈0.01）. Differences in slope of the regression curves of ACT were observed with all the reagents tested （glass beads 248.2 s/IU, diatomite 74.8 s/IU, kaolin 58.2 s/IU and magnetic bar 1112.2 s/IU, P〈0.01）. While the minimum concentration of dalteparin was 0.2 IU/ml, 0.4 IU/ml, 1.4 IU/ml and 0.2 IU/ml separately, the ACT values of the four coagulants （glass beads, diatomite, kaolin and magnetic bar） were beyond the normal limit and showed a noticeable increase respectively （P〈0.01）. The corrosion behavior and microstructure of Fe78Si9B13 glassy alloy in NaOH and NaCl solutions under a 0.02-T magnetic field were investigated through electrochemical testing and scanning electron microscopy （SEM）. The current-density prepeak （PP） in the anodic polarization curves in low-concentration NaOH solutions （classified as type I） tends to disappear when the NaOH concentration is increased to 0.4 mol/L and the magnetic field is applied. Under the magnetic field, the height of the second current-density peak is increased in low-concentration NaOH solutions （type I） but decreased in high-concentration NaOH solutions （type Ⅱ）. The non-monotonic effect of the magnetic field was similarly observed in the case of polarization curves of samples measured in NaCl solutions, Ring-like corroded patterns and round pits are easily formed under the magnetic field in NaOH and NaC1 solutions. These experimental results were discussed in terms of the magnetohydrodynamic （MHD） effect.

The anisotropy of the microstructure, thermal expansion behavior, corrosion resistance and magnetic properties of AISI 1045 steel was investigated. The distribution of Fe3C lameUae in the investigation plane parallel to the radial directions of molds was observed to differ from that in the investigation plane perpendicular to the radial directions by transmission electron microscopy. The lattice constants a0 of α-Fe deduced from the XRD patterns of samples prepared using a sand （S）-mold and cut parallel to the radial direction of the mold （S//） and using a metal （M）-mold and cut parallel to the radial direction （M//）, the corrosion resistance measured using an electrochemical work- station, and the magnetic permeability obtained by vibrating sample magnetometry also indicated the existence of anisotropy in the tested samples. The anisotropic change of corrosion potential （Eeorr）, pitting potential （Epit） and magnetic permeability （μ） of the samples was observed to depend on the orientation factorF200 of α-Fe in the measured samples, which is controlled by the distribution of Fe3C lamellae in the eutectoid structure.

Background For patients with moderate to high-risk acute coronary syndromes （ACS） who undergo early, invasive treatment strategies, current guidelines recommend the usage of glycoprotein （GP） lib/Ilia inhibitors as an upstream treatment for a coronary care unit or as an downstream provisional treatment for selected patients who are undergoing percutaneous coronary intervention （PCI）. The relative advantage of either strategy is unknown. The purpose of this study was to evaluate the effects of upstream tirofiban versus the effects of downstream tirofiban on myocardial damage and 180-day major adverse cardiovascular events （MACE） after PCI in high-risk non-ST-segment elevation ACS （NSTE-ACS） undergoing PCI. Methods From July 2006 to July 2007, 160 high-risk NSTE-ACS undergoing PCI were randomized to receive upstream （within 4-6 hours before coronary angiography） tirofiban or downstream （the guidewire crossing the lesion） tirofiban, to evaluate the extent of myocardial damage after PCI by quantitatively and qualitatively analyzing the value of cardiac troponin I （cTnl） as well as MB isoenzyme of creatine kinase （CK-MB） before and after PCI. The incidences of 24-hour, 3-day, 7-day, 30-day and 180-day MACE after PCI were followed up and the rates of bleeding complications and thrombocytopenia during tirofiban administration were recorded. Results The peak release and cumulative release of cTnl levels within 48 hours after PCI were significantly lower with upstream tirofiban than downstream tirofiban （0.45 vs 0.63 and 0.32 vs 0.43, respectively; P 〈0.05）. Post-procedural cTnl elevation within 48 hours was significantly less frequent among patients who received the upstream tirofiban than those who received the downstream tirofiban （66.3% vs 87.5%, P 〈0.05）. HLA Class I Expressions on Peripheral Blood Mononuclear Cells in Colorectal Cancer Patients

Objective： To investigate the expression change of human leukocyte antigen （HLA） class I on human peripheral blood mononuclear ceils （PBMCs） at both mRNA and protein levels, and to evaluate its roles in the development of colorectal cancer （CRC）. Methods： In the present study, 50 patients with CRC, 35 patients with benign colorectal lesion and 42 healthy volunteers were enrolled. Expression levels of HLA class I mRNA and protein were determined using real-time quantitative reverse transcription PCR （RT-PCR） and flow cytometry analysis, respectively. Results： The expression levels of HLA class I mRNA and proteins were not influenced by age and gender. The relative ratios of HLA class I mRNA were 0.99±0.27 in healthy controls, 0.76±0.19 in benign patients, and 0.48±0.21 in CRC patients. Mean fluorescence intensities of HLA class I were 145.58±38.14 in healthy controls, 102.05±35.98 in benign patients and 87.44±34.01 in CRC patients. HLA class I on PBMCs was significantly down-regulated at both mRNA and protein levels in patients with stage III and IV CRC. CRC patients with lymph node metastasis also showed a decreased HLA class I expression at protein level. Conclusion： HLA class I expressions on PBMCs are associated with staging of CRC and lymph node metastasis. Monitoring the expression of HLA class I on PBMCs may provide useful information for diagnosis and metastasis judgement of CRC.

Alkene hydrosilylation is one of the most concise and atom-economical methods to synthesize organosilicon ***,we reported the precise immobilization of metal single atoms(M-SAs;M=Ru,Rh,Ir,Pd,Pt,and Au)into a porphyrinic metalorganic framework(MOF)of PCN-222(PCN=porous coordination network),and then applied the resultant MOF composites of M-SAs@PCN-222 to alkene *** solvent-free conditions,Pt-SAs@PCN-222 displayed an especially high catalytic efficiency with the turnover frequency up to 119 s−1 and the maximum turnover number of 906,250 at room *** and theoretical studies revealed that there existed strong interactions between Pt-SAs@PCN-222 and the substrates,which helped to condense the substrates in the cavities of the porous *** density functional theory calculations and molecular dynamics simulations disclosed that PCN-222 could transfer electrons to Pt-SAs to enhance the silane oxidative addition and drive the reaction to proceed smoothly via Chalk–Harrod pathway.
