TOPK Inhibition Enhances the Sensitivity of Colorectal Cancer Cells to Radiotherapy by Reducing the DNA Damage Response

Objective Abnormal expression of T-lymphokine-activated killer cell-originated protein kinase(TOPK)was reported to be closely related to the resistance of prostate cancer to radiotherapy and to targeted drug resistance in lung ***,the role of TOPK inhibition in enhancing radiosensitivity of colorectal cancer(CRC)cells is *** study aimed to evaluate the radiosensitization of TOPK knockdown in CRC *** The expression of TOPK was detected in CRC tissues by immunohistochemistry,and the effect of TOPK knockdown was detected in CRC cells by Western ***-8 and clonogenic assays were used to detect the growth and clonogenic ability of CRC cells after TOPK knockdown combined with radiotherapy in CRC ***,proteomic analysis showed that the phosphorylation of TOPK downstream proteins changed after *** damage was detected by the comet *** in the DNA damage response signaling pathway were analyzed by Western blotting,and apoptosis was detected by flow *** The expression of TOPK was significantly greater in CRC tissues at grades 2–4 than in those at grade *** irradiation,CRC cells with genetically silenced TOPK had shorter comet tails and reduced expression levels of DNA damage response-associated proteins,including phospho-cyclin-dependent kinase 1(p-CDK1),phospho-ataxia telangiectasia-mutated(p-ATM),poly ADP-ribose polymerase(PARP),and meiotic recombination 11 homolog 1(MRE11).Conclusions TOPK was overexpressed in patients with moderately to poorly differentiated ***,TOPK knockdown significantly enhanced the radiosensitivity of CRC cells by reducing the DNA damage response.

High-precision simulation and experimental verification of residual stress and surface topography of cylindrical surface shot peening

Shot peening is commonly employed for surface deformation strengthening of cylindrical surface part. Therefore, it is critical to understand the effects of shot peening on residual stress and surface topography. Compared to flat surface, cylindrical surface shot peening has two significant features: (i) the curvature of the cylindrical surface and the scattering of the shot stream cause distributed impact velocities; (ii) the rotation of the part results in a periodic variation of the impact velocity component. Therefore, it is a challenge to quickly and accurately predict the shot peening residual stress and surface topography of cylindrical surface. This paper developed a high-precision model which considers the more realistic shot peening process. Firstly, a kinematic analysis model was developed to simulate the relative movement of numerous shots and cylindrical surface. Then, the spatial distribution and time-varying impact information was calculated. Subsequently, the impact information was used for finite element modeling to predict residual stress and surface topography. The proposed kinematic analysis method was validated by comparison with the discrete element method. Meanwhile, 9310 high strength steel rollers shot peening test verified the effectiveness of the model in predicting the residual stress and surface topography. In addition, the effects of air pressure and attack angle on the residual stress and surface topography were investigated. This work could provide a functional package for efficient prediction of the surface integrity and guide industrial application in cylindrical surface shot peening.

Synergistic enhancement on mechanical properties and corrosion resistance of biodegradable Mg-Zn-Y alloy via V-microalloying

For the sake of improving the mechanical properties and corrosion resistance of biodegradable Mg alloy synergistically,various content of element V(0,0.05,0.10,0.15,0.20 wt.%)are introduced into an Mg-Zn-Y alloy with long-period stacking ordered(LPSO)structure,and the effects of V on its microstructure,mechanical properties and corrosion resistance are investigated *** results indicate that the grains are effectively refined by V addition,and the primaryα-Mg in Mg-Zn-Y-V0.1 alloy is most significantly refined,with grain size being decreased by 62%.The amount of 18R LPSO structure is increased owing to the V *** growth mode of the second phase(W-phase and 18R LPSO structure)is transformed to divorced growth pattern,which ascribes to the thermodynamic drive force of V to promote the nucleation of LPSO ***,18R LPSO structure presents a continuous *** to grains refinement and modification of second phase,the tensile strength and strain of alloys are both enhanced ***,the ultimate tensile strength and the elongation of V0.1 alloy are 254 MPa and 15.26%,which are 41%and 61%higher than those of V-free alloy,*** to the continuously distributed 18R LPSO structure with refined grains and stable product film,the weight loss and hydrogen evolution corrosion rates of V0.1 alloy are 7.1 and 6.2 mmy^(-1),respectively,which are 42.6%and 45.4%lower than those of V-free alloy.

In order to fully utilize the regenerative braking energy of metro trains and stabilize the metro DC traction busbar voltage,a hybrid regenerative braking energy recovery system with a dual-mode power management strategy is ***,the construction of the hybrid regenerative braking energy recovery system is ***,based on the power demand of low-voltage load in metro stations,a dual-mode power management strategy is proposed to allocate the reference power of each system according to the different working conditions,and the control methods of each system are ***,the correctness and effectiveness of the dual-mode strategy are verified through simulation,and the proposed braking energy utilization scheme is compared with other singleform utilization *** results illustrate that the hybrid system with the dual-mode strategy can effectively recycle the regenerative braking energy of metro train and inhibit the busbar voltage fluctuation;the proposed braking energy utilization scheme has certain advantages on energy recovery and DC bus voltage stabilization compared with other single-form schemes;the proposed power management strategy can correctly allocate the reference power of each system with a lower construction cost.

An Adaptive Control Strategy for Energy Storage Interface Converter Based on Analogous Virtual Synchronous Generator

In the DC microgrid,the lack of inertia and damping in power electronic converters results in poor stability of DC bus voltage and low inertia of the DC microgrid during fluctuations in load and photovoltaic *** address this issue,the application of a virtual synchronous generator(VSG)in grid-connected inverters control is referenced and proposes a control strategy called the analogous virtual synchronous generator(AVSG)control strategy for the interface DC/DC converter of the battery in the ***,a flexible parameter adaptive control method is introduced to further enhance the inertial behavior of the AVSG ***,a theoretical analysis is conducted on the various components of the DC microgrid,the structure of analogous virtual synchronous generator,and the control structure’s main parameters related to the DC microgrid’s inertial ***,the voltage change rate tracking coefficient is introduced to adjust the change of the virtual capacitance and damping coefficient flexibility,which further strengthens the inertia trend of the DC ***,a small-signal modeling approach is used to analyze the approximate range of the AVSG’s main parameters ensuring system ***,conduct a simulation analysis by building the model of the DC microgrid system with photovoltaic(PV)and battery energy storage(BES)in MATLAB/*** results from different scenarios have verified that the AVSG control introduces fixed inertia and damping into the droop control of the battery,resulting in a certain level of inertia ***,the additional adaptive control strategy built upon the AVSG control provides better and flexible inertial support for the DC microgrid,further enhances the stability of the DC bus voltage,and has a more positive impact on the battery performance.

A Disulfide-Based Poly(ether-b-amide)Copolymer with Rapid Self-healing Ability under Moderate Conditions

Comprehensive Summary A disulfide-based poly(ether-b-amide)copolymer with rapid self-healing capability under moderate conditions was synthesized from 4,4’-dithiodibutyric acid(DTDBA),isophorondiamine(IPDA)and poly(tetramethylene oxide)(PTMO)based on a two-step *** incorporation of IPDA with asymmetric structure and substantial steric hindrance not only effectively decreased the number of regular H-bonds,but also inhibited the crystallization of polyamide segments,imparting the synthesized poly(ether-b-amide)copolymer with an amorphous structural *** on the coordination of segmental diffusion and recombination of disulfide bonds and hydrogen bonds,the scratches and damaged mechanical properties of PEBADS-I611 could be completely self-healed after healing at 40°C for only 11 min and 3 h,respectively.
