AIM To investigate the effect of metformin on activated hepatic stellate cells(HSCs) and the possible signaling pathways involved. METHODS A fibrotic mouse model was generated by intraperitoneal injection of carbon tetrachloride(CCl_4) and subsequent treatment with or without metformin. The level of fibrosis was detected by hematoxylin-eosin staining, Sirius Red staining, and immunohistochemistry. The HSC cell line LX-2 was used for in vitro studies. The effect of metformin on cell proliferation(CCK8 assay),motility(scratch test and Transwell assay), contraction(collagen gel contraction assay), extracellular matrix(ECM) secretion(Western blot), and angiogenesis(ELISA and tube formation assay) was investigated. We also analyzed the possible signaling pathways involved by Western blot *** Mice developed marked liver fibrosis after intraperitoneal injection with CCl_4 for 6 wk. Metformin decreased the activation of HSCs, reduced the deposition of ECM, and inhibited angiogenesis in CCl_4-treated mice. Platelet-derived growth factor(PDGF) promoted the fibrogenic response of HSCs in vitro, while metformin inhibited the activation, proliferation, migration, and contraction of HSCs, and reduced the secretion of ECM. Metformin decreased the expression of vascular endothelial growth factor(VEGF) in HSCs through inhibition of hypoxia inducible factor(HIF)-1α in both PDGF-BB treatment and hypoxic conditions, and it down-regulated VEGF secretion by HSCs and inhibited HSC-based angiogenesis in hypoxic conditions in vitro. The inhibitory effects of metformin on activated HSCs were mediated by inhibiting the Akt/mammalian target of rapamycin(m TOR) and extracellular signal-regulated kinase(ERK) pathways via the activation of adenosine monophosphate-activated protein kinase(AMPK).CONCLUSION Metformin attenuates the fibrogenic response of HSCs in vivo and in vitro, and may therefore be useful for the treatment of chronic liver diseases.

Impacts of Climate Change on Forest Ecosystems in Northeast China

This paper reviews the studies and research on climate change impacts on the forest ecosystems in Northeast China. The results show that in the context of global and regional warming, the growing season of coniferous forests has been increasing at an average rate of 3.9 d per decade. Regional warming favors the growth of temperate broad-leaved forests and has a detrimental effect on the growth of boreal coniferous forests. Over the past hundred years, the forest edge of the cool temperate zone in the southern Daxing＇anling region has retreated 140 km northward. From 1896 to 1986, the northern boundary of broad-leaved forests in Heilongjiang province has extended northwestward about 290 km. Future climatic changes （until 2060） may lead to the northern deciduous needle forests moving out of China＇s territory altogether. The occurrence cycles of pests and diseases have shortened; their distribution ranges have expanded. The life cycle of tent caterpillars （Malacosoma neustria testacea Motschulsky） has shortened from 14-15 years in the past to 8-10 years now. The pine caterpillar （Dendrolimus tabulaeformis Tsai et Liu）, which has spread within western Liaoning province and the nearby areas, can now be found in the north and west. Lightning fires in the Daxing＇anling region have significantly increased since 1987, and August has become the month when lightning fires occur most frequently. Overall, the net primary productivity （NPP） of forest in Northeast China has increased. The NPP in 1981 was around 0.27 Pg C, and increased to approximately 0.40 Pg C in 2002. With the current climate, the broad-leaved Korean pine forest ecosystem acts as a carbon sink, with a carbon sink capacity of 2.7 Mg C hm-2. Although the carbon sink capacity of the forest ecosystems in Northeast China has been weakened since 2003, the total carbon absorption will still increase. Although iron oxide(Fe_(3)O_(4)) nanoparticles have broad application prospects as magnetic resonance imaging(MRI) contrast agent, their biocompatibility and biotoxicity still need to be improved. In this study, we prepared Fe_(3)O_(4)@Angelica sinensis polysaccharide nanoparticles(Fe_(3)O_(4)@ASP NPs) with a 9 nm Fe_(3)O_(4) core and ASP as the coating material. The Fe_(3)O_(4)@ASP NPs are superparamagnetic, can be taken up by liver and spleen macrophages in the circulatory system in vivo, and are a good-biocompatibility and low-toxicity transverse relaxation time(T_(2)) and T_(2)-star(T_(2)^(*)) magnetic resonance imaging(MRI) contrast agent for the liver. The cytotoxicity assessment using HeLa cells and the pathological tests in mice validate that Fe_(3)O_(4)@ASP NPs have low toxicity and good biocompatibility in vivo, which can be attributed to the ASP as a natural polysaccharide with good biocompatibility and its function of protecting the liver. Fe_(3)O_(4)@ASP NPs are a potential new MRI contrast agent with high signal intensity in vivo.

Microstructure,texture,and mechanical properties of the extruded Mg-2.49Nd-1.82Gd-0.2Zn-0.2Zr alloy were investigated at different extrusion temperatures(260 and 320℃),extrusion ratios(10:1,15:1,and 30:1),and extrusion speeds(3 and 6 mm/s).The experimental results exhibited that the grain sizes after extrusion were much finer than that of the homogenized alloy,and the second phase showed streamline distribution along the extrusion direction(ED).With extrusion temperature increased from 260 to 320℃,the microstructure,texture,and mechanical properties of alloys changed *** dynamic recrystallization(DRX)degree and grain sizes enhanced as the extrusion ratio increased from 10:1 to 30:1,and the strength gradually decreased but elongation(EL)*** the extrusion speed increased from 3 to 6 mm/s,the grain sizes and DRX degree increased significantly,and the samples presented the typical-rare-earth(RE)*** alloy extruded at 260℃ with extrusion ratio of 10:1 and extrusion speed of 3 mm/s showed the tensile yield strength(TYS)of 213 MPa and EL of 30.6%.After quantitatively analyzing the contribution of strengthening mechanisms,it was found that the grain boundary strengthening and dislocation strengthening played major roles among strengthening *** results provide some guidelines for enlarging the industrial application of extruded Mg-RE alloy.

Three main phytotoxic compounds including lunatoic acid A （1）, 5Z-7-oxozeaenol （2） and zeaenol （3） were isolated from the fermentation broth of Cladosporium oxysporum DH14, a fungus residing in the locust （Oxya chinensis ） gut. Two additional derivative compounds, compound la and lb, were synthesized by methylation and chlorination of compound 1, respectively. The structures of such compounds were identified on the basis of spectroscopic analysis and by comparison of the corresponding data to those previously reported in the literature. Compounds 1-3 exhibited significantly phytotoxic activities against the radicle growth of Amaranthus retroflexus L. with the 50% inhibitory concentrations （ICso values） of 4.51, 4.80 and 8.16 μg· mL^-1, respectively, which is comparable to that positive control 2,4-dichlorophenoxyacetic acid （IC50=1.95 μg· mL^-1）. Furthermore, the compound 1 showed selective phytotoxic activity with the inhibition rate of less than 22% against the crops of Brassica rapa L., Sorghum durra, Brassica campestris L,, Capsicum annucm and Raphanus safivus L. under the concentration of 100 μg· mL^-1 Both derivatives of compound 1 had moderate phytotoxic activity against the radicle growth of A. retroflexus L. The findings of our present study suggest that these compounds provide new promising candidates for the potential management strategies of weeds.

This paper considers the dynamical behavior of solutions for non-autonomous stochastic fractional Ginzburg–Landau equations driven by additive noise with α∈(0, 1). First, we give some conditions for bounding the fractal dimension of a random invariant set of non-autonomous random dynamical system. Second, we derive uniform estimates of solutions and establish the existence and uniqueness of tempered pullback random attractors for the equation in H. At last, we prove the finiteness of fractal dimension of random attractors.

xiao-Xu-Ming decoction has been widely used to treat stroke and sequelae of stroke. We have previously shown that the active fractions of xiao-Xu-Ming decoction attenuate cerebral ischemic injury. However, the global protein profile and signaling conduction pathways regulated by xiao-Xu-Ming decoction are still unclear. This study established a two-vessel occlusion rat model by bilateral common carotid artery occlusion. Rats were intragastrically administered 50 or 150 mg/kg xiao-Xu-Ming decoction for 4 consecutive weeks. Learning and memory abilities were measured with Morris water maze. Motor ability was detected with prehensile test. Coordination ability was examined using the inclined screen test. Neuronal plasticity was observed by immunofluorescent staining. Differentially expressed proteins of rat hippocampus were analyzed by label-free quantitative proteomics. Real time-polymerase chain reaction and western blot assay were used to identify the changes in proteins. Results showed that xiao-Xu-Ming decoction dramatically alleviated learning and memory deficits, and motor and coordination dysfunction, and increased the expression of microtubule-associated protein 2. xiao-Xu-Ming decoction extract remarkably decreased 13 upregulated proteins and increased 39 downregulated proteins. The regulated proteins were mainly involved in oxidation reduction process, intracellular signaling cascade process, and protein catabolic process. The signaling pathways were mainly involved in ubiquitin mediated proteolysis and the phosphatidylinositol signaling system. Furthermore, there was an interaction among Rab2 a, Ptpn1, Ppm1 e, Cdk18, Gorasp2, Eps15, Capza2, Syngap1 and Mt-nd1. Protein analyses confirmed the changes in expression of MTND1. The current findings provide new insights into the molecular mechanisms of xiao-Xu-Ming decoction extract's effects on chronic cerebral hypoperfusion.
