Panaxdiol Saponins Component Promotes Hematopoiesis and Modulates t Lymphocyte Dysregulation in Aplastic Anemia Model Mice

Objective: to investigate the potential efficacy of panaxadiol saponins component(PDS-C) in the treatment of aplastic anemia(AA) model mice. Methods: totally 70 mice were divided into 7 groups as follows: normal, model, low-, medium-, high-dose PDS-C(20, 40, 80 mg/kg, namely L-, M-, H-PDS-C), cyclosporine(40 mg/kg), and andriol(25 mg/kg) groups, respectively. An immune-mediated AA mouse model was established in BALB/c mice by exposing to 5.0 Gy total body irradiation at 1.0 Gy/min, and injecting with lymphocytes from DBA mice. On day 4 after establishment of AA model, all drugs were intragastrically administered daily for 15 days, respectively, while the mice in the normal and model groups were administered with saline solution. After treatment, the peripheral blood counts, bone marrow pathological examination, colony forming assay of bone marrow culture, t lymphocyte subpopulation analysis, as well as t-bet, GAtA-3 and Fox P3 proteins were detected by flow cytometry and Western blot. Results: the peripheral blood of white blood cell(WBC), platelet, neutrophil counts and hemoglobin(Hb) concentration were significantly decreased in the model group compared with the normal group(all Pto 3 dose PDS-C treatment, the WBC, platelet, neutrophil counts were significantly increased at a dose-dependent manner compared with the model group(all Pthe myelosuppression status of AA was significantly reduced in M-, H-PDS-C groups, and hematopoietic cell quantity of bone marrow was more abundant than the model group. the colony numbers of myeloid, erythroid and megakaryocytic progenitor cells in the model group were less than those of the normal mice in bone marrow culture, while, PDS-C therapy enhanced proliferation of hematopoietic progenitor cells by significantly increasing colony numbers(all Pthermore,PDS-C therapy increased peripheral blood CD3^+ and CD3^+CD4^+ cells and reduced CD3^+CD8^+ cells(Ptreatme

Pyridocarbazole alkaloids from Ochrosia borbonica: lipid-lowering agents inhibit the cell proliferation and adipogenesis of 3t3-L1 adipocyte via intercalating into supercoiled DNA

Bioassay-guided fractionation of an ethanolic extract of Ochrosia borbonica led to the isolation of two known pyridocarbazole alkaloids,ellipticine(1)and 9-methoxyellipticine(2),and six known monoterpenoid indole alkaloids(3-8).Lipid-lowering assay in 3 t3-L1 cell model revealed that 1 and 2 could significantly inhibit the lipid droplet formation(EC50(28)0.41 and 0.92μmol·L^–1,respectively)and lower triglyceride levels by 50%-60%at the concentration of 1μmol·L^–1,being more potent than the positive drug luteolin(EC50(28)2.63μmol·L–^1).A mechanistic study indicated that 1 and 2 could intercalate into supercoiled DNA,which consequently inhibited the mitotic clonal expansion of 3 t3-L1 cells at the early differentiation phase,leading to the retardance of following adipogenesis and *** findings suggest that 1 and 2 may serve as promising leads for further d evelopment of anti-obesity drugs.

the roles of thyroid hormone receptor and t3 in metamorphosis of Haliotis diversicolor

thyroid hormone is a kind of important hormone which regulates metamorphosis. Its role is well described in amphibian metamorphosis. thyroid hormones (t3 and t4) have also been demonstrated to play a role in metamorphosis of marine invertebrates. However, the mechanism of thyroid hormone in metamorphosis of marine invertebrates remains unknown. A homolog of vertebrate thyroid hormone receptor (tR) was cloned and identified in abalone Haliotis diversicolor and was named HdtR . the mRNA expressions of HdtR , thyroid peroxidase ( tPO ), thyroid peroxidase 1 ( tPO1 ), idothyronine deiodinase Ⅲ( IDⅢ) and integrin alpha-V ( ItGAV ) had significant diff erence in metamorphosis of H . diversicolor . Metamorphosis rate and mortality rate were significantly diff erent in HdtR RNAi experiment and t3 inducing experiment. In RNAi experiment, ItGAV and CCND1 (cyclin D1) expression of dsRNA HdtR exposing group were significantly lower than those of blank control and negative control. But CtNNB (catenin beta) expression of dsRNA HdtR exposing group was higher than that those of blank control and negative control. ERK (extracellular signal regulated kinases) and PI3K (phosphoinositide-3-kinase) had no significant diff erence in RNAi experiment. Moreover, ItGAV of 1 μmol/L t3 group was significantly lower than that of 0 μmol/L t3 group, PI3K expression of 10 μmol/L t3 group was higher than that of 0 μmol/L t3 group, and the other genes expression had no significant diff erence in t3 inducing experiment. the data of genes expression suggested that CCND1 might be an eff ector gene of tR genomic action, while CtNNB might be regulated by unliganded tR. CCND1 and CtNNB may be involved in cell proliferation of metamorphosis. t3 might regulate the expression level of PI3K via nongenomic way. these results shed light on the mechanism of thyroid hormone in abalone metamorphosis.

Changes of 5＇ terminal Nucleotides of PCR Primers Causing Variable t-A Cloning Efficiency

t-A cloning takes advantage of the unpaired adenosyl residue added to the 3＇ terminus of amplified DNAs by taq and other thermostable DNA polymerase and uses a Ilnearlzed plasmld vector with a protruding 3＇ thymldylate residue at each of Its 3＇ termini to clone polymerase chain reaction （PCR）-derived DNA fragments. It Is a simple, reliable, and efficient Ilgatlon-dependent cloning method for PCR products, but the drawback of variable cloning efficiency occurs during application. In the present work, the relationship between variable t-A cloning efficiency and the different 5＇ end nucleotlde base of primers used In PCR amplification was studied. the results showed that different cloning efficiency was obtained with different primer pairs containing A, t, C and G at the 5＇ terminus respectively. the data shows that when the 5＇ end base of primer pair was adenosyl, more white colonies could be obtained In cloning the corresponding PCR product In comparison with other bases. And the least white colonies were formed when using the primer pair with 5＇ cytldylate end. the gluanylate end primers resulted In almost the same cloning efficiency In the white colonies amount as the thymldylate end primer did, and this efficiency was much lower than that of adenosyl end primers. this presumably is a consequence of variability In 3＇dA addition to PCR products mediated by taq polymerase. Our results offer instructions for primer design for researchers who choose t-A cloning to clone PCR products.

A new method to increase the luminance and quantum efficiency of polymer light emitting diodes with a lower threshold voltage has been *** threshold voltagef luminajice and quantum efficiency have been significantly improved by doping certain dopants with a lower highest occupied molecular orbital(HOMO)level into the hole transporting layer.A high performance device has been achieved by addition of the perylene and tri ph enylamin e as a dopant into poly(N-vinylcarbazole).the luminance and quantum efficiency increase by 2-3 times in comparison with the undoped device,reaching 10000cd/m^(2) in luminance and 0.58%in quantum efficiency,while threshold voltage is reduced to one half va/*** energy diagram has been obtained by measuring the HOMO levels and band gap *** on this,the carriers injection and balance between electrons and holes as well as the action of dopant are discussed.

As electrodes,two-dimensio nal materials show special advantages including the infinite planar lengths,broad electrochemical window,large surface-volume ratio,and much exposed active *** theory,the two-dimensional materials consist of the elements with high electronegativity may absorb more Na atoms,resulting in a high battery storage *** on the above idea,we selected the two dimensional metallic PS2 with 1 t-type structure as an anode material,and explored its potential applications as an electrode material for Na-ion battery through first-principle *** we expected,when two dimensional PS2 is used as an anode in Na-ion battery,it can adsorb maximum three layers of sodium atoms on both sides of the monolayer,resulting in a maximum theoretical capacity of 1692 mAh/***,it also possesses a rather small sodium diffusion barrier of 0.17 eV,a low average open-circuit voltage of 0.18 V,and a relatively small lattice changes within 13% during the intercalation of *** results suggested that the two dimensional PS2 is a potentially excellent Na-ion battery *** idea of designing two-dimensional anode materials with high storage capacity may provide some references for designing the next generation anode materials of metal-ion batteries.

Risk Management for International Portfolios with Basket Options:A Multi-Stage Stochastic Programming Approach

the authors consider the problem of active international portfolio management with basket options to achieve optimal asset allocation and combined market risk and currency risk management via multi-stage stochastic programming(MSSP). the authors note particularly the novel consideration and signi?cant bene?t of basket options in the context of portfolio optimization and risk *** empirical tests strongly demonstrate that basket options consistently have more clearly improvement on portfolio performances than a portfolio of vanilla options written on the same underlying assets. the authors further show that the MSSP model provides as a supportive tool for asset allocation,and a suitable test bed to empirically investigate the performance of alternative strategies.

Fusing t5 exonuclease with Cas9 and Cas12a increases the frequency and size of deletion at target sites

CRISPR/Cas systems, especially CRISPR/Cas9, generally result in small insertions/deletions, which are unlikely to eliminate the functions of regulatory and other non-coding sequences. to generate larger genomic deletions usually requires the use of pairs of guide RNAs. Here we show that it is possible to create such deletions with a single guide RNA by fusing Cas9 or Cas12a with t5 exonuclease(t5exo). these fusion constructs were found to increase both the frequency and size of deletions at target loci in rice protoplasts and seedlings. Moreover, the genome editing efficiencies of Cas9 and Cas12a were also enhanced by fusion with t5 exonuclease. these t5exo-Cas fusions expand the CRISPR toolbox, and facilitate knockout of regulatory and non-coding DNA sequences. From a wider standpoint, our results suggest a general strategy for producing larger deletions using other Cas nucleases.

Constrained predictive control of nonlinear stochastic systems

the receding horizon control（RHC） problem is considered for nonlinear Markov jump systems which can be represented by takagi-Sugeno fuzzy models subject to constraints both on control inputs and on observe *** the given receding horizon,for each mode sequence of the t-S modeled nonlinear system with Markov jump parameter,the cost function is optimized by constraints on state trajectories,so that the optimization control input sequences are obtained in order to make the state into a terminal invariant *** of the receding horizon,the stability is guaranteed by searching a state feedback control *** on such stability analysis,a linear matrix inequality approach for designing receding horizon predictive controller for nonlinear systems subject to constraints both on the inputs and on the outputs is *** simulation shows the validity of this method.

Free vibration and transient response of double curved beams connected by intermediate straight beams

this paper investigates the free vibration and transient response of interconnected structures including double curved beams and intermediate straight beams,which are all constructed from functionally graded porous(FGP) materials. the strain potential and kinetic energies of each beam along with the work done by the external force are calculated. Additionally, a higher-order beam element is introduced to derive stiffness and mass matrices, along with the force vector. the curved and straight beams are discretized, and their assembled stiffness, mass matrices, and force vectors, are *** conditions at the joints are used to derive the total matrices of the entire structure. Subsequently, the natural frequencies and transient response of the system are determined. the accuracy of the mathematical model and the self-developed computer program is validated through the comparison of the obtained results with those of the existing literature and commercial software ANSYS, demonstrating excellent ***, a comprehensive study is conducted to investigate the effects of various parameters on the free vibration and transient response of the considered structure.
