Let G be a nontrivial connected and vertex-colored graph. A subset X of the vertex set of G is called rainbow if any two vertices in X have distinct colors. The graph G is called rainbow vertex-disconnected if for any two vertices x and y of G, there exists a vertex subset S of G such that when x and y are nonadjacent, S is rainbow and x and y belong to different components of G-S;whereas when x and y are adjacent, S + x or S + y is rainbow and x and y belong to different components of(G-xy)-S. For a connected graph G, the rainbow vertex-disconnection number of G, denoted by rvd(G), is the minimum number of colors that are needed to make G rainbow vertexdisconnected. In this paper, we characterize all graphs of order n with rainbow vertex-disconnection number k for k ∈ {1, 2, n}, and determine the rainbow vertex-disconnection numbers of some special graphs. Moreover, we study the extremal problems on the number of edges of a connected graph G with order n and rvd(G) = k for given integers k and n with 1 ≤ k ≤ n.

The wake induced vibration(WIV)of a one-and two-degree-of-freedom(1DOF,2DOF)downstream wave-cone cylinder(WCC)behind a stationary equal-size upstream wave-cone cylinder in the staggered arrangement is numerically investigated at subcritical Reynolds number of 3900 by using shear stressed transfer(SST)k-ωturbulence *** streamwise pitch ratios(P/Dm)vary from 4 to 6 with a fixed incident angleα=8°.Experimental measurements were also performed for the validation of the present numerical *** is found that the largest vibration amplitude in crossflow direction occurred at P/Dm=4,Ur=8 with small difference of streamwise vibration at P/Dm=4,*** from single wavy-cone cylinder(SWCC),the downstream flexible one of a pair staggered WCCs got larger vibration amplitude during phase switching stage instead of in-phase *** upstream wake will suppress the triple frequency of main frequency in the power spectra density(PSD)functions of Cl but stimulate the double one of *** intriguing vibration mechanism happened in all 2DOF cases where the trajectory of downstream WCC is a significant ellipse rather than a figure of *** transformation of phase switching and the variation of the main frequency of drag coefficient(Cd)can be explained by the vortex-shedding modes of downstream WCC.

The most widely adopted method for diagnosing respiratory infectious diseases is to conduct polymerase chain reaction(PCR)assays on patients’respiratory specimens,which are collected through either nasal or oropharyngeal *** manual swab sampling process poses a high risk to the examiner and may cause false-negative results owing to improper *** this paper,we propose a pneumatically actuated soft end-effector specifically designed to achieve all of the tasks involved in swab *** soft end-effector utilizes circumferential instability to ensure grasping stability,and exhibits several key properties,including high load-to-weight ratio,error tolerance,and variable swab-tip stiffness,leading to successful automatic robotic oropharyngeal swab sampling,from loosening and tightening the transport medium tube cap,holding the swab,and conducting sampling,to snapping off the swab tail and sterilizing *** an industrial collaborative robotic arm,we integrated the soft end-effector,force sensor,camera,lights,and remote-control stick,and developed a robotic oropharyngeal swab sampling *** this swab sampling system,we conducted oropharyngeal swab-sampling tests on 20 *** digital PCR assay results(RNase P RNA gene absolute copy numbers for the samples)revealed that our system successfully collected sufficient numbers of cells from the pharyngeal wall for respiratory disease *** summary,we have developed a pharyngeal swab-sampling system based on an“enveloping”soft actuator,studied the sampling process,and imple-mented whole-process robotic oropharyngeal swab-sampling.

OBJECTIVE: To reveal the potential underlying mechanism of the Shenqi Tiaoshen formula(参芪调肾方, SQTS) in the treatment of chronic obstructive pulmonary disease(COPD) by utilizing network pharmacology, molecular docking, and experimental verification. METHODS: Multiple open-source databases and research related to Traditional Chinese Medicine or compounds were employed to screen active ingredients and corresponding potential targets of the SQTS. The protein-protein interaction network screened hub genes, the relevant molecular mechanism and gene regulation were initially identified through the Kyoto Encyclopedia of Genes and Genomes(KEGG) and Gene Ontology(GO) analysis, and molecular docking was used to confirm further the interaction of the main components bound to the core targets. In vivo experiments on the COPD combined Qi-deficiency syndrome rat model were performed to verify the intervention effects and predicted potential molecular mechanisms of the SQTS. RESULTS: This study selected 156 active compounds and 326 candidate targets for treating COPD. Quercetin, Nobiletin, Kaempferol, Luteolin, Ginsenoside Rh2 and Formononetin were probably the main active compounds of SQTS in COPD treatment as they affected the most COPD-related targets, and interleukin-1(IL-6), signal transducing activator of transcription 3(STAT3), matrix metalloproteinase-9(MMP9), vascular endothelial growth factor A(VEGFA), protein kinase B(AKT1), hypoxiainducible factor-1α(HIF-1α), and forkhead box O3(FoxO3) were identified as the hub genes associated with its therapeutic effect. KEGG analysis was mainly enriched in the signaling pathways closely related to inflammation, immunity and oxidative stress, such as HIF-1, tumor necrosis factor(TNF), phosphatidylinositol 3 kinase(PI3K)-AKT, FoxO, apoptosis, IL-17, and toll-like receptor. Background: No other effects of atropine other than as an antagonist of muscarinic acetylcholine receptor (mAChR) have been found. Methods: In this study, human kidneyepithelial cells were treated with different physiological regulators. Results: Subsequently, it was found that atropine could significantly induce autophagy as demonstrated by the appearance of autophagosome-like double- or single-membrane vesicles in the cytoplasm ofhost cells and the number of GFP-LC3 dots. In addition, increased conversion of the autophagy marker protein LC3-I and LC3-II and increased p62/SQSTM1 indicatedincomplete autophagy. In addition, atropine induced autophagosome levels in a dose-dependent manner within a certain concentration range in human kidney epithelial cells. In atropine-treated mouse skeletal muscle cells containing nicotinic acetylcholinereceptors and rat cardiac muscle cells containing mAchR, atropine induced autophagy in mouse skeletal muscle cells but not in rat cardiac muscle cells. Furthermore, atropine did not induce autophagy in tissue cells containing mAchR in vivo but did in tissue cells not containing mAchR. Conclusion: This study expands the application and understanding of atropine’s action mechanism in the field of medicine.

To improve the nitriding resistance of Zr_(2)Fe alloy,the Ce-added Zr_(2)(Fe_(1-x)Ni_(x))(x=0,0.15,0.3,0.5)alloys were prepared by magnetic levitation melting *** effects of Ni substitution for Fe on the phase structure,hydrogen and nitrogen absorption properties of the alloys were investigated by X-ray diffraction,scanning electron microscopy and p-c isotherm *** experimental results show that Ni substitution can effectively inhibit the formation of both α-Zr and ZrFe_(2) phases and promote the formation of C16 structure Zr_(2)(Fe,Ni)phase,causing Ni-substituted alloys to exhibit low nitrogen absorption rate and *** 623 K under 0.5 MPa nitrogen pressure,the nitrogen absorption capacity of Ce-added Zr_(2)(Fe_(0.5)Ni_(0.5))alloy reaches to 0.8 mL/g,much lower than that of Zr_(2)Fe alloy(1.5 mL/g).Ni substitution decreases the crystal cell volume of the C_(16)Zr_(2)(Fe,Ni)phase,resulting in an increase in the hydrogen absorption equilibrium *** 623 K under 0.05 MPa hydrogen pressure,the hydrogen absorption capacity decreases from 1.46 wt%of Zr_(2)Fe alloy to 1.41 wt%of Ce-added Zr_(2)(Fe_(0.5)Ni_(0.5))alloy.

Rational surface charge engineering of haloalkane dehalogenase for boosting the enzymatic performance in organic solvent solutions

Biocatalysis in organic solvents(OSs)has numerous important applications,but native enzymes in OSs often exhibit limited catalytic ***,we proposed a computation-aided surface charge engineering strategy to improve the catalytic performance of haloalkane dehalogenase DhaA in OSs based on the energetic analysis of substrate binding to the DhaA *** variants with enhanced OS resistance were obtained by replacing negative charged residues on the surface with positive charged residue(Arg).Particularly,a four-substitution variant E16R/E93R/E121R/E257R exhibited the best catalytic performance(five-fold improvement in OS resistance and seven-fold half-life increase in 40%(vol)dimethylsulfoxide).As a result,the overall catalytic performance of the variant could be at least 26 times higher than the wild-type *** spectroscopy and molecular dynamics simulation studies revealed that the residue substitution mainly enhanced OS resistance from four aspects:(a)improved the overall structural stability,(b)increased the hydrophobicity of the local microenvironment around the catalytic triad,(c)enriched the hydrophobic substrate around the enzyme molecule,and(d)lowered the contact frequency between OS molecules and the catalytic *** findings validate that computationaided surface charge engineering is an effective and ingenious rational strategy for tailoring enzyme performance in OSs.
