Anterior gradient-2 （AGR2） promotes tumor growth, cell migration and cellular transformation and its enhanced expression is almost completely restricted to malignant tissues, thus making AGR2 an interesting target for the development of immunotherapeutic strategies. We investigated whether the AGR2 molecule comprises human leukocyte antigen （HLA）-A 0201-binding epitopes recognized by human cytotoxic T lymphocytes （CTLs）, which could be targeted in dendritic cell （DC）-based cancer immunotherapy against colorectal cancer （CRC）. We reviewed the sequence of AGR2 for peptides that could potentially bind to HLA-A 0201 with the aid of a computer-based program. Five candidate peptides with different binding scores were synthesized and tested. These peptides were then assessed for their immunogenicity to elicit specific immune responses mediated by CTLs in vitro by means of enzyme-linked immunospot assays and CTL assays. AGR2 was highly expressed in several CRC cell lines, including DK01, DLD1, KM 12C, HCT-8 and HT-29. DCs pulsed with AGR2-P2 （aa 11-19; LLVALSYTL） or AGR2-P4 （aa 127-135; RIMFVDPSL） generated potent CTLs that could lyse T2 cells pulsed with AGR2-P2 or AGR2-P4 and HLA-A0201＋ AGR2-positive CRC cell lines in a strong dose-dependent and HLA-A 0201-restricted manner. In conclusion, these novel epitopes derived from AGR2 protein may be attractive candidates for DC-based immunotherapy for CRC.

Value Function Mechanism in WSNs-Based Mango Plantation Monitoring System

Mango fruit is one of the main fruit commodities that contributes to Taiwan’s *** implementation of technology is an alternative to increasing the quality and quantity of mango plantation product *** this study,a Wireless Sensor Networks(“WSNs”)-based intelligent mango plantation monitoring system will be developed that implements deep reinforcement learning(DRL)technology in carrying out prediction tasks based on three classifications:“optimal,”“sub-optimal,”or“not-optimal”conditions based on three parameters including humidity,temperature,and soil *** key idea is how to provide a precise decision-making mechanism in the real-time monitoring system.A value function-based will be employed to perform DRL model called deep Q-network(DQN)which contributes in optimizing the future reward and performing the precise decision recommendation to the agent and system *** WSNs experiment result indicates the system’s accuracy by capturing the real-time environment parameters is 98.39%.Meanwhile,the results of comparative accuracy model experiments of the proposed DQN,individual Q-learning,uniform coverage(UC),and NaÏe Bayes classifier(NBC)are 97.60%,95.30%,96.50%,and 92.30%,*** the results of the comparative experiment,it can be seen that the proposed DQN used in the study has themost optimal *** with 22 test scenarios for“optimal,”“sub-optimal,”and“not-optimal”conditions was carried out to ensure the system runs well in the real-world *** accuracy percentage which is generated from the real-world data reaches 95.45%.Fromthe resultsof the cost analysis,the systemcanprovide a low-cost systemcomparedtothe conventional system.

Application of Fuzzy Control to Improve Flow Balance of Multi-Cavity Hot Runner System

In this study, we propose a new temperature compensation control strategy for a multi-cavity hot runner injection molding system, At first, the melt filling time of each cavity can be measured by installing temperature sensors on the position around end filling area, and filling time difference between the various cavities can be calculated. Then the melt temperature of each hot nozzle can be adjusted automatically by a control strategy established based on the Fuzzy Theory and a program compiled with LABVIEW software. Temperature changes the melt mobility, so the adjustment of temperature can equalize the filling time of the melt in each cavity, which can reduced the mass deviation between each cavity and make product properties of each cavity consistent. The conclusion of the experiment is as follows： For this contact lens box of a four-cavity Hot Runner mold, by applying hot runner temperature compensation control system, time difference can be reduced from 0.05 s to 0.01 s at each cavity, and the mass Standard deviation of the four cavity can be improved from 0.006 to 0.002. The ratio of imbalance can be reduced from 20% to 4%. Hence, the hot runner temperature compensation control system has significant feasibility and high potential in improving melt flow balance of multi-cavity molding application.

DEHP,Di-（2-ethylhexyl）phthalate,the most common phthalate plasticizer used in the production of polyvinyl chloride（PVC）.Several in vivo studies have shown that DEHP absorbed in the body is metabolized to produce MEHP,mono-ethylhexyl phthalate and other *** metabolite can be absorbed through the villi and travel in the circulation system then causes series damages in endocrine and reproductive *** addition,investigations also demonstrated an association with elevated induction of rat hepatic cancer,testicular cancer,and developmental toxicity in reproductive system under MEHP ***,the mechanism regarding to carcinogenicity induced by DEHP or its metabolite MEHP remains unclear up to *** therefore are investigating the cell viability and mutagenicity of DEHP and MEHP in Chinese hamster ovary（CHO）*** and mutagenicity induced by these chemicals in AS52 cell which inserted gpt *** show that the parental chemical,DEHP,as well as MEHP derivatives caused dose-dependent decreases in cell survival,but with very different potencies:MEHP has the high potency;and DEHP *** to these compounds induced dose-dependent production of ROS,as determined by Cm-H2DCF-DA *** was noted however that simultaneous treatment of ROS scavenger N-acetylcysteine（NAC）shows the protection against DEHP and MEHP induced DNA *** 6-TG selection assay,data indicate that exposure MEHP cause dose-dependent mutant frequency as compare to the *** summary,DEHP and MEHP are cytotoxic to the cells and require induction of ROS to exert their genotoxicity effects.

Activation of Ethanol Production by Combination of Recombinant <i>Ralstonia eutropha</i>and Electrochemical Reducing Power

Ralstonia eutropha was genetically modified to induce ethanol production from glucose. An electrochemical bioreactor was prepared to generate electrochemical reducing power coupled to regeneration of NADH. Growing cells of recombinant R. eutropha produced about 29 mM of ethanol in conventional conditions and 56 mM of ethanol in electrochemically reduced conditions from 100 mM glucose. Grown cells of the recombinant produced about 52 mM of ethanol in conventional conditions and 142 mM of ethanol in electrochemically reduced condition from 100 mM glucose. These results are a clue that electrochemical reducing power can induce the recombinant R. eutropha to produce more ethanol coupled to increase of NADH/NAD+ ratio.

As the COVID-19 epidemic spread across the globe,people around the world were advised or mandated to wear masks in public places to prevent its spreading *** some cases,not wearing a mask could result in a *** monitor mask wearing,and to prevent the spread of future epidemics,this study proposes an image recognition system consisting of a camera,an infrared thermal array sensor,and a convolutional neural network trained in mask *** infrared sensor monitors body temperature and displays the results in real-time on a liquid crystal display *** proposed system reduces the inefficiency of traditional object detection by providing training data according to the specific needs of the user and by applying You Only Look Once Version 4(YOLOv4)object detection technology,which experiments show has more efficient training parameters and a higher level of accuracy in object *** datasets are uploaded to the cloud for storage using Google Colaboratory,saving human resources and achieving a high level of efficiency at a low cost.
