Evolution of the R2R3-MYB gene family in six Rosaceae species and expression in woodland strawberry

R2R3-MYB gene family play important roles in plants development, metabolism, and responses to various biotic and abiotic stresses. In this study, 838 R2 R3-MYB genes were identified from six Rosaceae species, including 105 in woodland strawberry(Fragaria vesca), 173 in European pear(Pyrus communis), 219 in apple(Malus domestica), 121 in peach(Prunus persica), 121 in Chinese rose(Rosa chinensis), and 99 in black raspberry(Rubus occidentalis). All R2 R3-MYB genes in the six Rosaceae species were clustered into 51 species-specific duplicated clades with 109 genes and 50 lineage-specific duplicated clades with 242 genes according to phylogenetic analysis. R2 R3-MYB genes were distributed on all chromosomes in each of the six species, with a small amount of tandem duplication events. The proportion of tandem repeat genes ranged from 0 to 25.1%. The R2 R3-MYB protein was conserved in a clade and likely to share similar functions. The distribution of Ks showed the duplication times of R2 R3-MYB genes in six Rosaceae species. Furthermore, most of the R2 R3-MYB genes had Ka/Ks values less than 1, which indicated they were driven by purifying selection during the evolutionary processes. The GO term enrichment analysis revealed that R2 R3-MYB genes in strawberry and black raspberry were more divergent than in other Rosaceae species. Analysis of transcriptomes of 42 different tissues and development stages of woodland strawberry showed that high expression levels of R2 R3-MYB suggested that the R2 R3-MYB genes in strawberry played a key role in growth and development of both vegetative tissues and fruits. The strawberry R2 R3-MYB genes in sub-group of S1, S2, S11, S20, and S22 had high expression levels both in young leaves(YL) and old leaves(OL) strawberry tissues under drought treatments.

Metal-organic chemical vapor deposition was applied to fabricate YBa2Cu3OT-a （YBCO） films on singlecrystal LaA103 （001） substrates for its high deposition rate, easy adjustment on film composition, and low requirement on vacuum apparatus. The effects of Cu（tmhd）2 concentra- tion in the precursor on the properties of YBCO films were systematically investigated. X-ray diffraction （XRD） reveals that the mole ratio of Cu/Ba in the precursor from 0.77 to 0.97 is helpful to improve the crystallization and out-of-plane orientation of YBCO films; however, it hardly affects the inplane texture. Scanning electron microscope （SEM） shows the dense, crack-free but rough surface, on which there are Cu-O and Ba-Cu-O outgrowths identified by energy-dispersive spectrometer （EDS）. As the mole ratio of Cu/Ba increasing, the average size of Ba-Cu-O precipitates keeps increasing and the film composition becomes inhomoge- neous at the mole ratio of Cu/Ba of 0.97. The 250 nm thick YBCO film prepared at the mole ratio of Cu/Ba of 0.91 shows the critical current density （Jc） of 4.0 ***-2 （77 K, 0 T）.

Early bacterial colonization and succession within the gastrointestinal tract have been suggested to be crucial in the development of host *** this study,we have investigated the changes in live weight and concentrations of selected serum parameters in relation to their fecal bacterial communities as determined by high throughput sequencing of the 16S r RNA gene over the same period in *** results showed that lambs’growth performance,the serum parameters,fecal bacterial community and fecal bacterial functions were all affected(P<0.05)by age of the *** within age groups of fecal microbiota was lower in the preweaning period and increased sharply(P<0.05)after weaning at 60 *** similarity between the samples collected from birth to 90 days of age and those collected at 120 days of age,increased(P<0.05)sharply after 30 days of *** age-associated changes in microbial genera were correlated with the changes in concentrations of immune indicators,including negative(P<0.05)correlations between the relative abundance of Lachnospiraceae UCG-010,Eubacterium coprostanoligenes group,Ruminococcaceae UCG-005,Ruminococcaceae UCG-009,Ruminococcaceae UCG-013,Ruminiclostridium 6,Ruminococcaceae UCG-008,and Oscillibacter with serum concentrations of lipopolysaccharide(LPS),D-lactate dehydrogenase(DLA),immunoglobulin(IgA,IgM,and IgG),and cytokines(interleukin-1β(IL-1β),IL-6,IL-12,and IL-17),tumor necrosis factor-α(TNF-α),and the relative abundance of these genera increased from 45 days of *** conclusion,these results suggested that the age-related abundances of particular genera were correlated with serum markers of immunity in lambs,and there might be a critical window in the period from birth to 45 days of age which provide an opportunity for potential manipulation of the fecal microbial ecosystems to enhance immune function.

Micro RNA-124（mi R-124） is abundantly expressed in neurons in the mammalian central nervous system, and plays critical roles in the regulation of gene expression during embryonic neurogenesis and postnatal neural differentiation. However, the expression profile of mi R-124 after spinal cord injury and the underlying regulatory mechanisms are not well understood. In the present study, we examined the expression of mi R-124 in mouse brain and spinal cord after spinal cord injury using in situ hybridization. Furthermore, the expression of mi R-124 was examined with quantitative RT-PCR at 1, 3 and 7 days after spinal cord injury. The mi R-124 expression in neurons at the site of injury was evaluated by in situ hybridization combined with Neu N immunohistochemical staining. The mi R-124 was mainly expressed in neurons throughout the brain and spinal cord. The expression of mi R-124 in neurons significantly decreased within 7 days after spinal cord injury. Some of the neurons in the peri-lesion area were Neu N＋/mi R-124-. Moreover, the neurons distal to the peri-lesion site were Neu N＋/mi R-124＋. These findings indicate that mi R-124 expression in neurons is reduced after spinal cord injury, and may reflect the severity of spinal cord injury.

Determination of risperidone in human plasma by HPLC-MS/MS and its application to a pharmacokinetic study in Chinese volunteers

This study presents a rapid, specific and sensitive liquid chromatography/tandem mass spectrometry (LC-MS/MS) assay for determination of risperidone (RIS) in human serum using paroxetine as an internal standard (IS). An Alltima-C18 column (2.1 mm×100 mm, 3 μm) and a mobile phase consisting of 0.1% formic acid-acetonitrile (40:60, v/v) were used for separation. The analysis was performed by selected reaction monitoring (SRM) method, and the peak area of the m/z 411.3→191.1 transition for RIS was measured versus that of the m/z 330.1→192.1 transition for IS to generate the standard curves. The assay linearity of RIS was confirmed over the range 0.25～50.00 ng/ml and the limit of quantitation was 0.05 ng/ml. The linear range corresponds well with the serum concentrations of the analytes obtained in clinical pharmacokinetic studies. Intraday and interday relative standard deviations were 1.85%～9.09% and 1.56%～4.38%, respectively. The recovery of RIS from serum was in the range of 70.20%～84.50%. The method was successfully applied to investigate the bioequivalence between two kinds of tablets (test versus reference products) in 18 healthy male Chinese volunteers. The result suggests that two formulations are bioequivalent.

Biogenic synthesis of silver nanoparticles using ginger(Zingiber officinale)extract and their antibacterial properties against aquatic pathogens

With the development of aquaculture, there is an urgent demand for an alternative antibacterial agent to reduce the drug resistance and environmental pollution caused by the abuse of antibiotics. Recently, silver nanoparticles（Ag NPs） have been viewed as a novel type of antimicrobial agents due to their unique advantages. In this study,Ag NPs were biosynthesized with the ginger rhizomes extract. The biosynthesized Ag NPs were characterised by UV–visible spectroscopy, transmission electron microscopy, X-ray diffraction and fourier transform infrared spectroscopy. Furthermore, the antimicrobial activities of the Ag NPs were fully analyzed against six typical aquatic pathogens. The results indicated that the components in ginger extract could function as the chemical reductant to synthesize Ag NPs. Moreover, compared with the Ag NPs synthesized by chemical methods, the biosynthesized Ag NPs were smaller, and had higher stability and antibacterial activity. Therefore, the biosynthesized Ag NPs using ginger extract may have prospective applications in aquaculture.

Prediction of miRNA Based on miRNA Biogenesis via One-class SVM

MicroRNAs are a class of small, single-stranded RNAs which are produced by non-protein-coding RNA genes with a length of 21-29 nt. They regulate the expression of protein-encoding genes at the post-transcriptional level and the degradation ofmRNAs by base pairing to mRNAs. Mature miRNAs are processed from 60-90 nt RNA hairpin structures called pre-miRNAs. At present, most of the machine learning computational methods for pre-miRNAs prediction are based on two-class SVM and use structural information of pre-miRNA hairpins. Those methods share a common feature that all of them need a negative dataset in the training dataset and feature selection in both training and testing dataset. In order to avoid selecting false negative examples of miRNA hairpins in the training dataset which may mislead the classifiers, we presented a microRNA prediction algorithm called MirBio based on miRNAs Biogenesis which is trained only on the information of the positive miRNAs class to predict miRNAs. It can predict both pre-miRNAs and miRNAs and get a relatively satisfying result in this study.
