Fe_(3)O_(4)@Angelica sinensis polysaccharide nanoparticles as an ultralow-toxicity contrast agent for magnetic resonance imaging

Although iron oxide(Fe_(3)O_(4)) nanoparticles have broad application prospects as magnetic resonance imaging(MRI) contrast agent, their biocompatibility and biotoxicity still need to be improved. In this study, we prepared Fe_(3)O_(4)@Angelica sinensis polysaccharide nanoparticles(Fe_(3)O_(4)@ASP NPs) with a 9 nm Fe_(3)O_(4) core and ASP as the coating material. The Fe_(3)O_(4)@ASP NPs are superparamagnetic, can be taken up by liver and spleen macrophages in the circulatory system in vivo, and are a good-biocompatibility and low-toxicity transverse relaxation time(T_(2)) and T_(2)-star(T_(2)^(*)) magnetic resonance imaging(MRI) contrast agent for the liver. The cytotoxicity assessment using HeLa cells and the pathological tests in mice validate that Fe_(3)O_(4)@ASP NPs have low toxicity and good biocompatibility in vivo, which can be attributed to the ASP as a natural polysaccharide with good biocompatibility and its function of protecting the liver. Fe_(3)O_(4)@ASP NPs are a potential new MRI contrast agent with high signal intensity in vivo.

Sound Velocity in Soap Foams

The velocity of sound in soap foams at high gas volume fractions is experimentally studied by using the time difference *** is found that the sound velocities increase with increasing bubble diameter,and asymptotically approach to the value in air when the diameter is larger than 12.5 *** propose a simple theoretical model for the sound propagation in a disordered *** this model,the attenuation of a sound wave due to the scattering of the bubble wall is equivalently described as the effect of an additional *** simplicity reasonably reproduces the sound velocity in foams and the predicted results are in good agreement with the *** measurements indicate that the increase of frequency markedly slows down the sound velocity,whereas the latter does not display a strong dependence on the solution concentration.

Recombinant adenovirus vector Ad-hIL-10 protects grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats

BACKGROUND: Interleukin 10 (IL-10), a Th2 type cytokine, modulates inflammatory responses by inhibiting the production of proinflammatory cytokines. This study was designed to investigate the protective effects of adenovirus-mediated human IL-10 (Ad-hIL-10) gene transfer on protecting grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats. METHODS: Adenoviruses encoding hIL-10 or beta-galactosidase (Ad-lacZ) were injected via the superior mesenteric vein into prospective donor animals. The donor liver was harvested 48 hours after transduction, and stored for 12 hours at 4 degrees C in lactated Ringer's solution prior to transplantation. The rats were divided into saline, Ad-lacZ, and Ad-hIL-10 groups. Liver function test, histopathological examination, reverse transcriptase-polymerase chain reaction (RT-PCR), and Western blotting were performed at 24 hours after transplantation in the three groups. RESULTS: Liver function (ALT and AST) was significantly improved, and the Suzuki score was significantly decreased in the Ad-hIL-10 group. The levels of hepatic TNF-alpha, MIP-2, ICAM-1 mRNA, and NF-kappa B protein in the Ad-hIL-10 group were significantly decreased. The expression of hIL-10 mRNA was detected by RT-PCR in Ad-hIL-10-treated grafts but not in controls treated with saline or Ad-lacZ. CONCLUSIONS: Donor pretreatment with Ad-hIL-10 down-regulates the expression of proinflammatory cytokines TNF-alpha, MIP-2, and ICAM-1 mRNA. hIL-10 protects against hepatic cold ischemia-reperfusion injury, at least in part, by suppressing NF-kappa B activation and subsequent expression of proinflammatory mediators. Amyloidosis is a multisystem disease that is characterized by deposition of fibrils in extracellular tissue,which mainly involves the kidney,heart,and autonomic nervous *** types of amyloidosis typically infiltrate the heart,including immunoglobulin light-chain(AL)and amyloid transthyretin(ATTR).ATTR is further subdivided into wild-type ATTR and variant ATTR caused by point mutations in the TTR gene.[1]Wild-type ATTR is considered as not uncommon in older patients with heart ***,a comprehensive set of consensus recommendations for the suspicion and diagnosis of ATTR was published,with particular focus on the combined application of noninvasive methods.[2]We present here a case of wild-type TTR cardiac amyloidosis(ATTRwt-CA),which was diagnosed by noninvasive modalities,and provide an overview of the recommended diagnostic approach of ***,to the best of our knowledge,this is the first Chinese case of ATTRwt-CA reported to date.

Cluster synchronization in community network with nonidentical nodes via intermittent pinning control

In this paper, cluster synchronization in community network with nonidentical nodes is investigated. By combining intermittency with a pinning control scheme, some effective controllers are designed. In the control scheme, only one node in each community is controlled and coupling weights of a spanning tree in each community are enhanced. Based on the Lyapunov function method and mathematical analysis technique, two results for achieving cluster synchronization are obtained. Noticeably, by introducing an adaptive strategy, some universal adaptive intermittent pinning controllers are designed for different networks. Finally, two numerical simulations are performed to verify the correctness of the derived results.

Inhibition of Dual Specific Oncolytic Adenovirus on Esophageal Cancer via Activation of Caspases by a Mitochondrial-dependent Pathway

We investigated the anti-tumor effects of dual cancer specific-oncolytic adenovirus Ad-VP on esophageal cancer（EC）. The anti-tumor activity of Ad-VP was compared with that of the control recombinant adenoviruses （Ad-GP, Ad-Apoptin, Ad-EGFP） in human esophageal cancer cell EC-109 and human normal liver cell L02 in vitro. In 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide（MTT） assays, the growth of EC-109 cells was slightly inhibited by Ad-GP, Ad-Apoptin and Ad-EGFE However, Ad-VP induced a significant cytotoxic effect. Infection of EC-109 cells with Ad-VP resulted in a significant induction of apoptosis of them in vitro, detected by 4＇,6-diamidino-2-phenylindole（DAPI） or acridine orange and ethidium bromide staining. The results of Western blot and flow cytometric assay indicate the loss of mitochondrial membrane potential（Aψm）, the release of eytochrome c and the activation of caspase-3, 6 and 7 in Ad-VP infected EC-109 cells. In contrast, all these assays show almost no effects of the recombinant adenoviruses on L02 cells. These results demonstrate that the treatment of tumors with Ad-VP selectively inhibits tumor growth and induces apoptosis of esophageal cancer cells. Ad-VP may provide a novel and powerful strategy for cancer gene therapy.

Porcine acute liver failure model established by two-phase surgery and treated with hollow fiber bioartificial liver support system

AIM： To establish a highly reproducible animal model of acute liver failure （ALF）, for assessing the effect of bioartificial liver support system （BALSS）. METHODS： A two-phase complete liver devascularization procedure was performed in eight Ioco-hybrid pigs. Blood biochemical index and liver biopsy were studied every 2 h after surgery, and survival time was recorded. The BALSS constructed with high volume recirculating technique was a hollow fiber circulating system consisting of a hepatocyte reactor-hollow fiber module inoculated with microcarrieradhering hepatocytes, and a double pump, heparinized, thermostabilized, micro-capsulized activated carbonadsorbing plasmapheresis system. Twelve pigs undergoing two-phase surgery were randomized into： control group （perfused without hepatocytes, n = 6） and treatment group （perfused with hepatocytes, n = 6）. Intergroup liver biochemical indexes, survival time, and liver pathological changes were analyzed at regular intervals. RESULTS： Two-phase surgery was performed in all the experimental pigs, and there was no obvious difference between their biochemical indexes. After 3 h of phase II surgery, ammonia （Amm） increased to （269＋37）μmol/L. After 5 h of the surgery, fibrinogen （Fib） decreased to （1.5±0.2） g/L. After 7 h of the surgery, ALT, AST, Tbil and PT were （7.6±1.8） nka/L, （40±5） nka/L, （55±8）μmol/L and （17.5±1.7） nka/L respectively. After 9 h of surgery, ALB and Cr were （27±4） g/L and （87±9）μmol/L. After 13 h of surgery, BUN was （3.5±0.9） μmol/L. All the above values were different from those determined before surgery. Survival time of pigs averaged 13.5±1.4 h. ALF pigs in the other group were treated with BALSS. The comparison analysis between the treated and control animals showed the changes of Tbil, PT, AIb, BUN, Cr, Fib, and Amm （P〈0.01）, but there was no change of ALT and AST.
