Thiosemicarbazones of 2-amino-5-chlorobenzophenone and 3-aminobenzophenone （L1-La） have been synthesized and their Cu（II） complexes （1-4） were afforded via coordination with cupric chloride. All these compounds were characterized by UV-vis and lR spectroscopy together with CHN elemental analysis. NMR spectroscopy was also applied to characterize the ligands. In vitro cholinesterase inhibitory assays for the complexes （1-4） showed ICso values less than 10 ~molJL, with complex 1 exhibiting the most activity, ICso = 2.15 ~molJL and 2.16 i^molJL for AChE and BuChE, respectively. Molecular modeling simulation revealed the binding interaction template for complex I with the AChE and BuChE receptors. In DPPH assay, the complexes also showed more in vitro antioxidant activities in comt）arison tn their narent li^ands.

AIM:To develop an automated model for subfoveal choroidal thickness(SFCT)detection in optical coherence tomography(OCT)images,addressing manual fovea location and choroidal contour ***:Two procedures were proposed:defining the fovea and segmenting the *** localization from B-scan OCT image sequence with three-dimensional reconstruction(LocBscan-3D)predicted fovea location using central foveal depression features,and fovea localization from two-dimensional en-face OCT(LocEN-2D)used a mask region-based convolutional neural network(Mask R-CNN)model for optic disc detection,and determined the fovea location based on optic disc relative *** segmentation also employed Mask ***:For 53 eyes in 28 healthy subjects,LocBscan-3D’s mean difference between manual and predicted fovea locations was 170.0μm,LocEN-2D yielded 675.9μ***-2D performed better in non-high myopia group(P=0.02).SFCT measurements from Mask R-CNN aligned with manual ***:Our models accurately predict SFCT in OCT ***-3D excels in precise fovea localization even with high ***-2D shows high detection rates but lower accuracy especially in the high myopia *** both models offers a robust SFCT assessment approach,promising efficiency and accuracy for large-scale studies and clinical use.

Constructed wetlands(CWs)are widely applied for decentralized wastewater ***,achieving efficient removal of ammonia(NH+-N)has proven challenging due to insufficient *** this study,natural hematite(Fe,O3)was employed as a CW substrate(H-CWs)for the first time to drive anaerobic ammonia oxidation coupled with iron(I)reduction(Feammox).Compared to gravel constructed wetlands(G-CWs),ammonia removal was enhanced by 38.14%to 54.03%and nitrous oxide(N_(2)0)emissions were reduced by 34.60%in *** synergistic removal of ammonia and nitrate by H-CWs also resulted in the absence of ammoxidation *** and 15N isotope tracer incubations showed that Feammox accounting for approximately 40%of all ammonia removal in the *** enrichment of iron phosphate(Fe_(3)Fe_(4)(PO_(4))_(6))promoted the accumulation of the Feammox intermediate compound *** nanowires were observed on the surface of H-CW substrates as well,suggesting that the observed biological ammoxidation was most likely related to extracellular electron transfer(EET).Microbial and metagenomics analysis revealed that H-CWs elevated the integrity and enhanced the abundance of functional microorganisms and genes associated with nitrogen ***,the efficient ammonia removal in the absence of O2 together with a reduction in N_(2)O emissions as described in this study may provide useful guidance for hematite-mediated anaerobic ammonia removal in CWs.

Arsenic(As) fate in paddy fields has been one of the most significant current issues due to the strong As accumulation potential of rice plants under fooded conditions. However,no attempt was done to explore As methylation and volatilization under non-fooded conditions. Herein, we investigated the effects of water management on As methylation and volatilization in three arsenic-contaminated soils enhanced by biostimulation with strawderived organic matter and bioaugmentation with genetic engineered Pseudomonas putida KT2440(GE P. putida). Under fooded conditions, the application of biochar(BC), rice straw(RS)and their combination(BC+RS) increased total As in porewater. However, these effects were greatly attenuated under non-fooded conditions. Compared with RS amendment alone, the combination of GE P. putida and RS further promoted the As methylation and volatilization,and the promotion percentage under non-fooded conditions were significantly higher than that under fooded conditions. The combined GE P. putida and RS showed the highest efficiency in As methylation(88 μg/L) and volatilization(415.4 μg/(kg·year)) in the non-fooded soil with moderate As contamination. Finally, stepwise multiple linear regression analysis presented that methylated As, DOC and p H in porewater were the most important factors contributing to As volatilization. Overall, our findings suggest that combination of bioaugmentation with GE P. putida and biostimulation with RS/BC+RS is a potential strategy for bioremediation of arsenic-contaminated soils by enhancing As methylation and volatilization under non-fooded conditions.

Objective: To study the in vitro and in vivo antitumor effects of the polysaccharide of Alocasia cucullata(PAC) and the underlying mechanism. Methods: B16F10 and 4T1 cells were cultured with PAC of 40 μg/m L, and PAC was withdrawn after 40 days of administration. The cell viability was detected by cell counting kit-8. The expression of Bcl-2 and Caspase-3 proteins were detected by Western blot and the expressions of ERK1/2 mRNA were detected by quantitative real-time polymerase chain reaction(qRT-PCR). A mouse melanoma model was established to study the effect of PAC during long-time administration. Mice were divided into 3 treatment groups: control group treated with normal saline, positive control group(LNT group) treated with lentinan at 100 mg/(kg·d), and PAC group treated with PAC at 120 mg/(kg·d). The pathological changes of tumor tissues were observed by hematoxylin-eosin staining. The cell apoptosis of tumor tissues was detected by TUNEL staining. Bcl-2 and Caspase-3 protein expressions were detected by immunohistochemistry, and the expressions of ERK1/2, JNK1 and p38 mRNA were detected by qRT-PCR. Results: In vitro, no strong inhibitory effects of PAC were found in various tumor cells after 48 or 72 h of administration. Interestingly however, after 40 days of cultivation under PAC, an inhibitory effect on B16F10 cells was found. Correspondingly, the long-time administration of PAC led to downregulation of Bcl-2 protein(P <0.05), up-regulation of Caspase-3 protein(P<0.05) and ERK1 mRNA(P<0.05) in B16F10 cells. The above results were verified by in vivo experiments. In addition, viability of B16F10 cells under long-time administration culture in vitro decreased after drug withdrawal, and similar results were also observed in 4T1 cells. Conclusions: Long-time administration of PAC can significantly inhibit viability and promote apoptosis of tumor cells, and had obvious antitumor effect in tumor-bearing mice.

Indoleamine 2, 3-dioxygenase (IDO) is a rate-limiting enzyme for the tryptophan catabolism. In human and murine cells, IDO inhibits antigen-specific T cell proliferation in vitro and suppresses T cell responses to fetal alloantigens during murine pregnancy. In mice, IDO expression is an inducible feature of specific subsets of dendritic cells (DCs), and is important for T cell regulatory properties. However, the effect of IDO and tryptophan deprivation on DC func- tions remains unknown. We report here that when tryptophan utilization was prevented by a pharmacological inhibitor of IDO, 1-methyl tryptophan (1MT), DC activation induced by pathogenic stimulus lipopolysaccharide (LPS) or inflam- matory cytokine TNF-α was inhibited both phenotypically and functionally. Such an effect was less remarkable when DC was stimulated by a physiological stimulus, CD40 ligand. Tryptophan deprivation during DC activation also regu- lated the expression of CCR5 and CXCR4, as well as DC responsiveness to chemokines. These results suggest that tryptophan usage in the microenvironment is essential for DC maturation, and may also play a role in the regulation of DC migratory behaviors.

This paper aims to enhance the compression capacity of underwater cylindrical shells by adopting the corrugated sandwich structure of *** cuttlebone suffers uniaxial external compression,while underwater cylindrical shells are in a biaxial compressive stress *** suit the biaxial compressive stress state,a novel bidirectional corrugated sandwich structure is proposed to improve the bearing capacity of cylindrical *** static and buckling analysis for the sandwich shell and the unstiffened cylindrical shell with the same volume-weight ratio are studied by numerical *** is indicated that the proposed sandwich shell can effectively reduce the ratio between circumferential and axial stress from 2 to 1.25 and improve the critical buckling load by about 1.63 *** simulation shows that optimizing and adjusting the structural parameters could significantly improve the advantage of the sandwich ***,the hydrostatic pressure tests for shell models fabricated by 3D printing are carried *** to the experimental results,the overall failure position of the sandwich shell is at the center part of the sandwich *** has been found the average critical load of the proposed sandwich shell models exceeds two times that of the unstiffened shell ***,the proposed bio-inspired bidirectional corrugated sandwich structure can significantly enhance the pressure resistance capability of cylindrical shells.

Toll-like receptors(TLRs),the key sensor molecules in vertebrates,trigger the innate immunity and prime the adaptive immune *** TLR family of rodents,the largest order of mammals,typically contains 13 TLR ***,a clear picture of the evolution of the rodent TLR family has not yet emerged and the TLR evolutionary patterns are unclear in rodent ***,we analyzed the natural variation and the evolutionary processes acting on the TLR family in rodents at both the interspecific and population *** results showed that rodent TLRs were dominated by purifying selection,but a series of positively selected sites(PSSs)primarily located in the ligand-binding domain was also *** numbers of PSSs differed among TLRs,and nonviralsensing TLRs had more PSSs than those in viral-sensing ***-conversion events were found between TLR1 and TLR6 in most rodent *** genetic analyses showed that TLR2,TLR8,and TLR12 were under positive selection in Rattus norvegicus and ***,whereas positive selection also acted on TLR5 and TLR9 in the former species,as well as TLR1 and TLR7 in the latter ***,we found that the proportion of polymorphisms with potentially functional change was much lower in viral-sensing TLRs than in nonviral-sensing TLRs in both of these rat *** findings revealed the first thorough insight into the evolution of the rodent TLR genetic variability and provided important novel insights into the evolutionary history of TLRs over long and short timescales.
