Endomembrane-biased dimerization of ABCG16 and ABCG25 transporters determines their substrate selectivity in ABA-regulated plant growth and stress responses

ATP-binding cassette(ABC)transporters are integral membrane proteins that have evolved diverse func-tions fulfilled via the transport of various *** Arabidopsis,the G subfamily of ABC proteins is particularly abundant and participates in multiple signaling pathways during plant development and stress *** this study,we revealed that two Arabidopsis ABCG transporters,ABCG16 and ABCG25,engage in ABA-mediated stress responses and early plant growth through endomembrane-specific dimerization-coupled transport of ABA and ABA-glucosyl ester(ABA-GE),*** first revealed that ABCG16 contributes to osmotic stress tolerance via ABA *** specifically,ABCG16 induces cellular ABA efflux in both yeast and plant *** FRET analysis,we showed that ABCG16 forms oblig-atory homodimers for ABA export activity and that the plasma membrane-resident ABCG16 homodimers specifically respond to ABA,undergoing notable conformational ***,we demonstrated that ABCG16 heterodimerizes with ABCG25 at the endoplasmic reticulum(ER)membrane and facilitates the ER entry of ABA-GE in both Arabidopsis and tobacco *** specific responsiveness of the ABCG16-ABCG25 heterodimer to ABA-GE and the superior growth of their double mutant support an inhib-itory role of these twoABCGs in early seedling establishment via regulation of ABA-GE translocation across the ER *** endomembrane-specific analysis of the FRET signals derived from the homo-or heterodimerized ABcG complexes allowed us to link endomembrane-biased dimerization to the transloca-tion of distinct substrates by ABcG transporters,providing a prototypic framework for understanding the omnipotence of ABcG transporters in plant development and stress responses.

Cellular hormone homeostasis is essential for precise spatial and temporal signaling responses and *** acid(ABA)plays pivotal roles in orchestrating various developmental and stress re-sponses and confersfitness benefits over ecological and evolutionary timescales in terrestrial *** ABA level is regulated by complex processes,including biosynthesis,catabolism,and ***25 is thefirst ABA exporter identified through genetic screening and affects diverse ABA *** the structural basis of ABA export by ABCG25 is critical for further manipulations of ABA homeostasis and *** used cryo-electron microscopy to elucidate the structural dynamics of AtABCG25 and successfully characterized different states,including apo AtABCG25,ABA-bound AtABCG25,and ATP-bound AtABCG25(E232Q).Notably,AtABCG25 forms a homodimer that fea-tures a deep,slit-like cavity in the transmembrane domain,and we precisely characterized the critical residues in the cavity where ABA *** binding triggers closure of the nucleotide-binding domains and conformational transitions in the transmembrane *** show that AtABCG25 belongs to a conserved ABCG subfamily that originated during the evolution of *** subfamily neofunctionalized to regulate seed germination via the endosperm,in concert with the evolution of this angio-sperm-specific,embryo-nourishing ***,thesefindings provide valuable insights into the intricate substrate recognition and transport mechanisms of the ABA exporter AtABCG25,paving the way for genetic manipulation of ABA homeostasis and plantfitness.

RACK1A promotes hypocotyl elongation by scaffolding light signaling components in Arabidopsis

Scaffold protein RACK1A positively regulates leaf senescence by coordinating the EIN3-miR164-ORE1 transcriptional cascade in Arabidopsis

Plants have adopted versatile scaffold proteins to facilitate the crosstalk between multiple signaling pathways. Leaf senescence is a well-programmed developmental stage that is coordinated by various external and internal signals. However,the functions of plant scaffold proteins in response to senescence signals are not well understood. Here, we report that the scaffold protein RACK1A(RECEPTOR FOR ACTIVATED C KINASE1A) participates in leaf senescence mediated by ethylene signaling via the coordination of the EIN3-miR164-ORE1 transcriptional regulatory cascade. RACK1A is a novel positive regulator of ethylene-mediated leaf senescence. The rack1a mutant exhibits delayed leaf senescence, while transgenic lines overexpressing RACK1A display early leaf senescence. Moreover, RACK1A promotes EIN3(ETHYLENE INSENSITIVE 3) protein accumulation, and directly interacts with EIN3 to enhance its DNA-binding activity. Together, they then associate with the miR164 promoter to inhibit its transcription, leading to the release of the inhibition on downstream ORE1(ORESARA 1) transcription and the promotion of leaf *** study reveals a mechanistic framework by which RACK1A promotes leaf senescence via the EIN3-miR164-ORE1 transcriptional cascade, and provides a paradigm for how scaffold proteins finely tune phytohormone signaling to control plant development.

实行分级诊疗是对医疗资源配置与利用的重构,有利于促进基本医疗服务均等化,也是实现健康中国战略目标的必然要求。自2009年我国正式提出建立分级诊疗制度以来,医疗资源配置不充分、不平衡的问题仍然比较突出。通过对医院和基层医疗卫生机构在人力资源、物力资源配置及资源利用效率方面的对比分析,发现医疗资源配置与分级诊疗的目标要求仍存在一定差距,应推动优质医疗资源下沉基层,加快制度创新,拓宽资源配置链条,推进资源整合共享,以实现分级诊疗的政策目标。

目的:研究参连汤在大白兔心肌缺血时防治T波电交替(T wave alternans,TWA)的离子基础。方法:建立大白兔左冠状动脉前降支结扎造成急性心肌梗死模型,测定假手术组、模型组、参连汤大剂量组、参连汤中剂量组以及参连汤小剂量组TWA、恶性室性心律失常和心性猝死的发生率,测定Ca2+浓度以及ATP酶活性。结果:参连汤各组动物的各种ATP酶活性升高,TWA的发生率减少,恶性室性心律失常减少,实验动物死亡率明显降低。同时我们还发现使用中大剂量参连汤疗效显著,明显优于低剂量组,尽管低剂量组也有效。结论:参连汤能有效控制TWA的发生发展,显著减少了恶性室性心律失常,疗效与剂量有关。

以耐盐指数、相对生长率和相对叶绿素指数为指标,在组培条件下对5种野生百合21份百合材料进行耐盐性评价,利用各指标的隶属函数值对所有材料进行聚类分析。结果表明,不论从耐盐指数、相对生长率,还是从相对叶绿素指数来看,供试百合材料间的耐盐性差异都达极显著水平。从整体上看,宜昌百合、卷丹与野百合耐盐性较强,山丹次之,宝兴最弱。聚类分析结果将供试材料分为3大类群,耐盐类、中等耐盐类和盐敏感类。其中耐盐材料主要包括宜昌百合、野百合和卷丹,盐敏感材料主要是山丹和宝兴百合。

Carbon dots(CDs)have gradually become a widely favored type of fluorescent nanomaterials with unlimited and promising *** work reports a means of breaking the shackle of method to develop new red and white CDs by introducing free *** resulting white emissive CDs in this strategy are employed to demonstrate electroluminescent white-light-emitting diodes(WLEDs)and achieve a record-high external quantum efficiency(0.95%)of one-step-produced white CDs(WCD)-LEDs,which dramatically simplifies the whole fabrication processes of *** additional passivation treatment,the red CDs(RCD2)with excellent properties such as N and S co-doping,bright(quantum yield=49%)and stable photoluminescence(PL),large positive zeta potential(+20.5 mV),and two-photon fluorescence are *** RCD2 are used for rapid staining(5 min)of yeast *** two CDs synthesized via this method have outstanding performance in different aspects,which provides new promise of CDs for further functionalization and applications.
