Hybrid liposome-erythrocyte drug delivery system for tumor therapy with enhanced targeting and blood circulation

Liposome,a widely used drug delivery system(DDS),still shows several disadvantages such as dominant clearance by liver and poor target organ *** overcome the drawbacks of liposomes,we developed a novel red blood cell(RBC)-liposome combined DDS to modulate the tumor accumulation and extend the blood circulation life of the existing liposomal ***,RBCs,an ideal natural carrier DDS,were utilized to carry liposomes and avoid them undergo the fast clearance in the *** this study,liposomes could either absorbed onto RBCs’surface or fuse with RBCs’membrane by merely altering the interaction time at 37°C,while the interaction between liposome and RBCs would not affect RBCs’*** the in vivo antitumor therapeutic efficacy study,1,2-dipalmitoyl-sn-glycero-3-phosphocholine(DPPC)liposomes attached onto RBCs’surfaces exhibited lung targeting effect(via RBC-hitchhiking approach)and reduced clearance in the liver,while DPPC liposomes fused with RBCs had prolong blood circulation up to 48 h and no enrichment in any ***,20 mol%of DPPC liposomes were replaced with pH-sensitive phospholipid 1,2-dioleoyl-Sn-glycero-3-phosphoethanolamine(DOPE)as it could respond to the low pH tumor microenvironment and then accumulate in the *** DOPE attached/fusion RBCs showed partial enrichment in lung and about 5-8%tumor accumulation,which were significantly higher than(about 0.7%)the conventional liposomal ***,RBC-liposome composite DDS is able to improve the liposomal tumor accumulation and blood circulation and shows the clinical application promises of using autologous RBCs for antitumor therapy.

Objective:To correlate the utility of the Fundamentals of Laparoscopic Surgery(FLS)manual skills program with the Objective Structured Assessment of Technical Skills(OSATS)global rating scale in evaluating operative ***:The Asian Urological Surgery Training and Educational Group(AUSTEG)Laparoscopic Upper Tract Surgery Course implemented and validated the FLS program for its usage in laparoscopic surgical ***’basic laparoscopic skills were assessed using three different training models(peg transfer,precision cutting,and intra-corporeal suturing).They also performed live porcine laparoscopic surgery at the same *** surgery skills were assessed by blinded faculty using the OSATS rating ***:From March 2016 to March 2019,a total of 81 certified urologists participated in the course,with a median of 5 years of post-residency *** differences in task time did not reach statistical significance,those with more surgical experience were visibly faster at completing the peg transfer and intra-corporeal suturing FLS ***,they took longer to complete the precision cutting task than participants with less *** OSATS scores correlated weakly with all three FLS tasks(peg transfer time:r=0.331,r^(2)=0.110;precision cutting time:r=0.240,r^(2)=0.058;suturing with intracorporeal knot time:r=0.451,r^(2)=0.203).Conclusion:FLS task parameters did not correlate strongly with OSATS globing rating scale *** FLS task models demonstrated strong validity,it is important to assimilate the inconsistencies when benchmarking technical proficiency against real-life operative competence,as evaluated by FLS and OSATS,respectively.

Characterization,expression dynamics,and potential function of OPA1 for regulation of mitochondrial morphology during spermiogenesis in Phascolosoma esculenta

Mitochondria undergo morphological changes during spermatogenesis in some *** mechanism and role of mitochondrial morphology regulation,however,remain somewhat *** this study,we analyzed the molecular characteristics,expression dynamics and subcellular localization of optic atrophy protein 1(OPA1),a mitochondrial fusion and cristae maintenance-related protein,to reveal the possible regulatory mechanisms underlying mitochondrial morphology in Phascolosoma esculenta *** full-length cDNA of the *** opa1 gene(Pe-opa1)is 3743 bp in length and encodes 975 amino *** Pe-OPA1 protein is highly conservative and includes a transmembrane domain,a GTPase domain,two helical bundle domains,and a lipid-interacting *** and protein expression was higher in the coelomic fluid(a site of spermatid development)of male *** and increased first and then decreased from March to ***,their expression during the breeding stage was significantly higher than during the non-breeding stage,suggesting that Pe-OPA1 is involved in *** *** Pe-OPA1 protein was more abundant in components consisting of many spermatids than in components without,indicating that Pe-OPA1 mainly plays a role in the spermatid in coelomic ***,Pe-OPA1 was mainly detected in the spermatid *** experiments showed that the Pe-OPA1 are constitutively expressed and co-localized with mitochondria during spermiogenesis,suggesting its involvement in *** *** results provide evidence for Pe-OPA1's involvement in the regulation of mitochondrial morphology during spermiogenesis.

Anaerobic biodegradation of PAHs in mangrove sediment with amendment of NaHCO_3

Mangrove sediment is unique in chemical and biological properties. Many of them suffer polycyclic aromatic hydrocarbon（PAH） contamination. However, the study on PAH biological remediation for mangrove sediment is deficient. Enriched PAH-degrading microbial consortium and electron acceptor amendment are considered as two effective measures. Compared to other electron acceptors, the study on CO2, which is used by methanogens, is still seldom. This study investigated the effect of Na HCO3 amendment on the anaerobic biodegradation of four mixed PAHs, namely fluorene（Fl）, phenanthrene（Phe）,fluoranthene（Flua） and pyrene（Pyr）, with or without enriched PAH-degrading microbial consortium in mangrove sediment slurry. The trends of various parameters, including PAH concentrations, microbial population size, electron-transport system activities, electron acceptor and anaerobic gas production were monitored. The results revealed that the inoculation of enriched PAH-degrading consortium had a significant effect with half lives shortened by 7–13 days for 3-ring PAHs and 11–24 days for 4-ring PAHs. While Na HCO3 amendment did not have a significant effect on the biodegradation of PAHs and other parameters, except that CO2 gas in the headspace of experimental flasks was *** of the possible reasons is that mangrove sediment contains high concentrations of other electron acceptors which are easier to be utilized by anaerobic bacteria, the other one is that the anaerobes in mangrove sediment can produce enough CO2 gas even without adding Na HCO3.

Treadmill exercise in combination with acousto-optic and olfactory stimulation improves cognitive function in APP/PS1 mice through the brain-derived neurotrophic factor-and Cygb-associated signaling pathways

A reduction in adult neurogenesis is associated with behavioral abnormalities in patients with Alzheimer's ***,enhancing adult neurogenesis represents a promising therapeutic approach for mitigating disease symptoms and ***,nonpharmacological interventions aimed at inducing adult neurogenesis are currently *** individual non-pharmacological interventions,such as aerobic exercise,acousto-optic stimulation,and olfactory stimulation,have shown limited capacity to improve neurogenesis and cognitive function in patients with Alzheimer's disease,the therapeutic effect of a strategy that combines these interventions has not been fully *** this study,we observed an age-dependent decrease in adult neurogenesis and a concurrent increase in amyloid-beta accumulation in the hippocampus of amyloid precursor protein/presenilin 1 mice aged 2-8 *** deposition became evident at 4 months,while neurogenesis declined by 6 months,further deteriorating as the disease ***,following a 4-week multifactor stimulation protocol,which encompassed treadmill running(46 min/d,10 m/min,6 days per week),40 Hz acousto-optic stimulation(1 hour/day,6 days/week),and olfactory stimulation(1 hour/day,6 days/week),we found a significant increase in the number of newborn cells(5'-bromo-2'-deoxyuridine-positive cells),immature neurons(doublecortin-positive cells),newborn immature neurons(5'-bromo-2'-deoxyuridine-positive/doublecortin-positive cells),and newborn astrocytes(5'-bromo-2'-deoxyuridine-positive/glial fibrillary acidic protein-positive cells).Additionally,the amyloid-beta load in the hippocampus *** findings suggest that multifactor stimulation can enhance adult hippocampal neurogenesis and mitigate amyloid-beta neuropathology in amyloid precursor protein/presenilin 1 ***,cognitive abilities were improved,and depressive symptoms were alleviated in amyloid precursor protein/presenilin 1 mice

Hokkien Merchants and the Kian Teik Tong： Economic and Political Influence in Nineteenth- Century Penang and Its Region

This article explores nineteenth-century Penang＇s Hokkkien merchants and their secret society or hui-the Klan Teik Tong diande Tang）--which had a variety of roles and an extensive network. It contextualizes the merchants＇ secret society as a transnational socioeconomic and political organization rather than as an overseas Chinese criminal group in the wider Penang area. By recovering Klan Teik Tong and its network, it can be shown how these merchants secured and mobilized labour, capital, and allies in a way that cut across linguistic, ethnic, class and state boundaries in order to establish control of coolies and the lucrative opium, tin, and rice businesses, in order to exert political influence in the colonial and indigenous milieus of the nineteenth-century Penang region. They established a social contract through their Klan Teik Tong relief activities and initiation rituals, and thus were able to recruit thousands of members who were mainly labourers. With such a substantial social force, the merchants launched organized violence against their rivals to attain dominance in opium revenue farming and tin mining businesses in Penang, Krabi, and Perak. The widespread and strategic location of the Klan Teik Tong in Burma also enabled the same merchants to monopolize the Penang-Burma rice trade. The versatility of the Klan Teik Tong＇s functions allowed them to operate as an alternative political order vis-a-vis the colonial and indigenous powers. This arrangement allowed the Hokkien merchants to gain significant political clout in confronting the Siamese and Dutch authorities.

A Qualitative Examination of Teacher-Student Power-Sharing in Chinese Classrooms:A Study in Hong Kong

This article explores the dynamics of power-sharing between teacher and students in learning and teaching situations,and describes the theoretical bases,implementation,and results of an empirical study in three elementary schools in Hong Kong Special Administrative Region of the People’s Republic of *** from 58 class observations and interviews with 50 students,25 teachers,and three school principals suggest that teachers can empower students by acknowledging their right to and responsibility for learning and by sharing power with ***-sharing classroom practice requires the dual efforts of teachers and students,and can be facilitated by the teachers’interactive teaching mode,students’cooperation and willingness to engage in learning,and school policy and culture,*** the power-sharing classrooms,teachers played the role of facilitators and students played as *** this article,theoretical implications for understanding power-sharing and critical pedagogy are *** empirical evidence from this Hong Kong study contributes to an understanding of teacher-initiated power-sharing in the Chinese context,and the practice of critical pedagogy in classrooms.
