Hepatitis C virus(HCV)infection represents an important public health problem *** of HCV morbidity and mortality is a current challenge owned to several viral and host *** molecular evolution plays an important role in HCV transmission,disease progression and therapy *** high degree of genetic heterogeneity characteristic of HCV is a key element for the rapid adaptation of the intrahost viral population to different selection pressures(e.g.,host immune responses and antiviral therapy).HCV molecular evolution is shaped by different mechanisms including a high mutation rate,genetic bottlenecks,genetic drift,recombination,temporal variations and *** evolutionary processes constantly rearrange the composition of the HCV intrahost population in a staging *** advances in the understanding of the molecular mechanism controlling HCV replication have facilitated the development of a plethora of direct-acting antiviral agents against *** a result,superior sustained viral responses have been *** rapidly evolving field of anti-HCV therapy is expected to broad its landscape even further with newer,more potent antivirals,bringing us one step closer to the interferon-free era.

G protein-coupled receptor kinase 2 （GRK2） is an important serine/threonine-kinase regulating different membrane receptors and intraceUular proteins. Attenuation of Drosophila Gprk2 in embryos or adult flies induced a defective differentiation of somatic muscles, loss of fibers, and a flightless phenotype. In vertebrates, GRK2 hemizygous mice contained less but more hypertrophied skeletal muscle fibers than wild-type littermates. In C2C12 myoblasts, overexpression of a GRK2 kinase-deficient mutant （K220R） caused precocious differentiation of ceUs into immature myotubes, which were wider in size and contained more fused nuclei, while GRK2 overexpression blunted differentiation. Moreover, p38MAPK and Akt pathways were activated at an earlier stage and to a greater extent in K220R-expressing cells or upon kinase downregulation, while the activation of both kinases was impaired in GRK2-overexpressing cells. The impaired differentiation and fewer fusion events promoted by enhanced GRK2 levels were recapitulated by a p38MAPK mutant, which was able to mimic the inhibitory phosphorylation of p38MAPK by GRK2, whereas the blunted differentiation observed in GRK2-expressing clones was rescued in the presence of a constitutively active upstream stimulator of the p38MAPK pathway. These results suggest that balanced GRK2 function is necessary for a timely and complete myogenic process.

Association between Cancer and Environmental Exposure to Glyphosate

Background: Argentina, Brazil, Paraguay and Uruguay farm transgenic seeds glyphosate resistant. Argentina annually utilizes 240,000 tonnes of glyphosate in agriculture. A change in the profile of morbidity and mortality is perceived in agricultural areas;cancer seems to prevail. Monte Maíz is a typical argentine agricultural town with 8000 inhabitants;the Mayor and residents of Monte Maiz requested an environmental health study due to perceived increase in cancer frequencies. Methods: An exploratory ecological study was developed to assess the urban environmental contamination and the frequencies and distribution of cancer through an environmental analysis of pollution sources including measurements of pesticides in water, soil and grain dust, and a cross-sectional study of cancer patients that explore associations with different variables. Results: Glyphosate was detected in soil and grain dust and was found to be at an even higher concentration in the village soil than in the rural area. 650 tonnes are used annually in the region and manipulated inner town. We do not find other relevant sources of pollution. Cancer incidence, prevalence, and mortality are between two and three times higher than the reference values (Globocan 2012, WHO) for the entire nation (706/100,000 persons vs. 217/100,000;2123/100,000 persons vs. 883.82/100,000 and 383/100,000 persons vs. 115.13/100,000, respectively). Conclusion: This study detects high glyphosate pollution in association with increased frequencies of cancer in a typical argentine agricultural village, and by design, cannot make claims of causality. Other study designs are required, but if we corroborate the concrescence of high exposure to glyphosate and cancer.

Deposition of the H2A.Z histone variant by the SWR1 complex （SWRI-C） in regulatory regions of specific loci modulates transcription. Characterization of mutations in Arabidopsis thaliana homologs of yeast SWRI-C has revealed a role for H2A.Z exchange in a variety of developmental processes. Nevertheless, the exact composition of plant SWRI-C and how it is recruited to target genes remains to be established. Here we show that SWC4, the Arabidopsis homolog of yeast SANT domain protein Swc4/Eaf2, is a DNA-binding protein that interacts with SWR1-C subunits. We demonstrate that the swc4-1 knockout mutant is embryo- lethal, while SWC4 RNAi knockdown lines display pleiotropic phenotypic alterations in vegetative and repro- ductive traits, including acceleration of flowering time, indicating that SWC4 controls post-embryonic processes. Transcriptomic analyses and genome-wide profiling of H2A.Z indicate that SWC4 represses tran- scription of a number of genes, including the floral integrator FT and key transcription factors, mainly by modulating H2A.Z deposition. Interestingly, SWC4 silencing does not affect H2A.Z deposition at the FLC locus nor expression of this gene, a master regulator of flowering previously shown to be controlled by SWR1-C. Importantly, we find that SWC4 recognizes specific AT-rich DNA elements in the chromatin regions of target genes and that SWC4 silencing impairs SWRI-C binding at FT. Collectively, our data suggest that SWC4 regulates plant growth and development by aiding SWR1-C recruitment and modulating H2A.Z deposition.

The lack of proper maintenance in buildings is responsible for various anomalies, which can cause property and personal damage. The objective of this study is to analyze the maintenance management of the administration building at the Technological Park UFRJ/Brazil, trying to emphasize the relation between maintenance and good performance of the building and to demonstrate the need to develop a consistent maintenance system to achieve an efficient maintenance management. The study allowed to present strategies and possibilities of interventions, in addition to proposing the essential aspects for the development of a structured maintenance system. After reviewing the issue of building maintenance under the various perspectives presented, the importance of planning a preventive building maintenance, according to standards, to achieve the level of performance expected for the building, besides being essential the search for tools and methods that can increase the efficiency of management can be concluded. It is also pertinent to conclude that prevention is the most effective solution against the depreciation factors that affect a building, being the most economical viable, and ensuring other benefits for the building and its users, such as security, property valuation and attendance to the warranty period.

Sick Neighborhood Syndrome: Population with Multiple Chemical Sensitivity Adjacent to Bioethanol Distillery

The population of a neighborhood in the city of Córdoba presents general irritant symptoms since a bioethanol distillery began to operate there, from which formaldehyde, toluene, xylene mainly emanate. At the request of the community, collective environmental health was evaluated with a cross-sectional study of prevalence of these conditions. 53% of the residents reported respiratory disorders, conjunctivitis 31%, headaches 27%, dermatitis 23% and dyspepsia 22%. 57% of children aged 6 - 7 use bronchodilators (an indicator of asthma). 66% of the population has one of these conditions, 46% has two and 26% has three;children are the most affected. The results coincide with the Consensus on Multiple Chemical Sensitivity Syndrome in that it is reproducible by repeated chemical exposure, is chronic and does not improve without eliminating exposure, affecting multiple organs and systems. 14.8% of newborns feature malformations, while in the rest of the city, only 1.7% did. Similar to the Sick Building Syndrome, the collective health picture found suggests a “Sick Neighborhood Syndrome”.

Recovery of Gold and Silver and Removal of Copper, Zinc and Lead Ions in Pregnant and Barren Cyanide Solutions

Over the past decade the concern about toxic metals in freshwater has increased. Environmental laws such as the Clean Water Act have forced industries that produce metal containing wastewater to treat their wastewater prior to discharge. The purpose of this study was to investigate the use of a novel method for the minimization of heavy metals in the wastewater from the mining industry. A very promising electrochemical treatment technique that does not require chemical additions is electrocoagulation (EC) and sulphide precipitation. The present study has been done for the recovery of gold and silver contained in pregnant solution from the cyanidation process using the electrocoagulation technology with iron electrodes;that is a developed alternative technology for the Merril-Crowe process. The average gold and silver content in pregnant solution was 4.27 and 283 ppm respectively and the recoveries were 92% for gold and 95% for silver, with optimum operating parameters of pH 10, residence time of 20 minutes and addition of sodium chloride of 4 gr/L. The results of precipitation process show that the elimination of lead, zinc, cooper and iron ions from the barren solution was successful, with optimum operating parameters of pH 3 and residence time of 15 minutes, and the recoveries were 99% of these ions. Finally the characterization of the solid products of gold and silver formed during the EC process with Scanning Electronic Microscope was performed. Results suggest that magnetite particles and amorphous iron oxyhydroxides (lepidocrocite) were present.

Argentina annually utilizes 240,000 tones of glyphosate in industrial agriculture and a change in the profile of morbidity is perceived for physicians of agricultural areas;now reproductive disorders seem to prevail. The objective of this study is to determine concurrence of glyphosate exposure and reproductive disorders in a typical argentine agricultural town (Monte Maíz). An ecological study was developed with an environmental analysis of pollution sources including measurements of glyphosate and other pesticides and a cross-sectional study of spontaneous abortions and congenital abnormalities prevalence. Glyphosate was detected in soil and grain dust and was found to be at an even higher concentration in the village soil than in the rural area;650 tonnes of glyphosate are used annually in the region and manipulated inner town contaminating the soil and dust in suspension of the town creating an burden of environmental exposure to glyphosate of 79 kg per person per year. We do not find other relevant sources of pollution. The spontaneous abortion and congenital abnormalities rates are three and two times higher than the national average reported by the national health (10% vs. 3% and 3% - 4.3% vs 1.4% respectively). Our study verified high environmental exposure to glyphosate in association with increased frequencies of reproductive disorders (spontaneous abortion and congenital abnormalities) in argentine agricultural village, but is unable to make assertions cause-effect. Further studies are required with designs for such purposes.

The Mexican oregano ＂Lippia berlandieri Shauer＂ is a versatile plant, for multiple purposes and high potential adaptative capabilities. To evaluate the possibility to use oregano as a cover surface on tailings deposits and thereby mitigate the emission of particulate material with metals content, an experiment was carried out comparing the plant growing rate over a period of six-month in four substrates： tailings, vermicompost, ＂soil＂ of the region and a mixture of ＂tailings plus vermicompost＂. The statistical analysis shows a significant difference between treatments, resulting in the ＂soil＂ substrate with the best performance, followed by the mixture of＂tailings plus vermicompost＂ treatment. The low permeability oftailings strongly affects the adaptability and growth of the oregano plant; however, the adaptation possibilities increase in the mixture oftailings with vermicompost or ＂soil＂ of the region.
