BACKGROUND Gastric cancer is a leading cause of cancer-related mortality *** somatic mutations have been identified based on next-generation sequencing;they likely play a vital role in cancer treatment ***,nextgeneration sequencing has not been widely used to diagnose and treat gastric cancer in the *** To test the mutant gene frequency as a guide for molecular diagnosis and personalized therapy in gastric cancer by use of next-generation *** We constructed a panel of 24 mutant genes to detect somatic nucleotide variations and copy number variations based on a next-generation sequencing *** custom panel included high-mutation frequency cancer driver and tumour suppressor *** genes were also analyzed using the cBioPortal *** clinical annotation of important variant mutation sites was evaluated in the ClinVar *** searched for candidate drugs for targeted therapy and immunotherapy from the OncoKB *** In our study,the top 16 frequently mutated genes were TP53(58%),ERBB2(28%),BRCA2(23%),NF1(19%),PIK3CA(14%),ATR(14%),MSH2(12%),FBXW7(12%),BMPR1A(12%),ERBB3(11%),ATM(9%),FGFR2(8%),MET(8%),PTEN(6%),CHD4(6%),and KRAS(5%).TP53 is a commonly mutated gene in gastric cancer and has a similar frequency to that in the cBioPortal database.33 gastric cancer patients(51.6%)with microsatellite stability and eight patients(12.5%)with microsatellite instability-high were *** analyses demonstrated that high-frequency mutated genes had transmembrane receptor protein kinase *** discovered that BRCA2,PIK3CA,and FGFR2 gene mutations represent promising biomarkers in gastric *** We developed a powerful panel of 24 genes with high frequencies of mutation that could detect common somatic *** observed mutations provide potential targets for the clinical treatment of gastric cancer.

In recent years,significant increases in waste processing and material engineering have been seen by using advanced oxidation *** treatment results and energy yields of these processes are largely determined by the generation and retention of reactive oxygen species(ROS).However,increasing the amount of ROS remains a key challenge because of the unavailability of performance-and energy-efficient *** this study,plasma electrolysis,ultrasound,and plasma electrolysis combined with ultrasound were used to treat dimethyl sulfoxide(DMSO)solutions,and the results showed that the two methods can synergistically convert filament discharge into spark discharge,and the conversion of the discharge mode can significantly increase the concentration of OH radicals and effectively improve the efficiency of DMSO *** verified the rationality of the results by analyzing the mass transfer path of ROS based on the reaction coefficients and found that the OH radicals in aqueous solution were mainly derived from the decomposition of hydrogen *** findings indicated that the synergistic action of plasma electrolysis and ultrasound can enhance the production of chemically reactive species,and provide new insights and guiding principles for the future translation of this combined strategy into real-life *** results demonstrated that the synergistic strategy of ultrasound and plasma electrolysis is feasible in the switching mode and increasing the ROS,and may open new routes for materials engineering and pollutant degradation.

Sericin is a kind of water-soluble protein expressed specifically in the middle silk gland ofBombyx mori. When the sericin-1 gene promoter was cloned and a transgenic vector was constructed to express a foreign protein, a specific Helitron, Bmhel-8, was identified in the sericin-1 gene promoter sequence in some genotypes of Bombyx mori and Bombyx mandarina. Given that the Bmhel-8 Helitron transposon was present only in some genotypes, it could be the source of allelic variation in the sericin-I promoter. The length of the sericin-1 promoter sequence is approximately 1063 or 643 bp. The larger size of the sequence or allele is ascribed to the presence of Bmhel-8. Silkworm genotypes can be homozygous for either the shorter or larger promoter sequence or heterozygous, containing both alleles. Bmhel-8 in the sericin-1 promoter exhibits enhancer activity, as demonstrated by a dual-luciferase reporter system in BraE cell lines. Furthermore, Bmhel-8 displays enhancer activity in a sericin-1 promoter-driven gene expression system but does not regulate the tissue-specific expression of sericin-1.

In this study,we investigated the effects of high ambient temperature on lipid metabolism in finishing *** pigs（（79.6±1.2） kg） were randomly assigned to two groups：（1） ambient temperature of 30℃ with ad libitum access to feed（HT;n=8） and（2） ambient temperature of 22℃ and fed the average amount consumed by eight pigs in HT group on the previous day（PF;n=8）.After 21 days of constant exposure to different environmental conditions,all the pigs were euthanized,and blood and tissue samples were *** ambient temperature increased the proportion of backfat（P=0.04,＋21.6%）and flare fat（P〈0.01,＋43.3%）.Compared with pair-fed pigs,the activities of fatty acid synthase（FAS） and malic enzyme in backfat and flare fat were lower（P〈0.05） in heat-stressed pigs,as were the amounts of acetyl-CoA-carboxylase and FAS in the longissimus muscle（LM）,the amount of FAS in backfat（P〈0.01）,and FAS activity in the liver（P〈0.01）.Ambient temperature did not affect the amount of hormone-sensitive lipase in different *** amount of lipoprotein lipase in flare fat tended to be higher（P=0.09,＋28.3%）,and the activities of β-hydroxyacyl coenzyme A dehydrogenase in front and back of LM were lower（P〈0.01,-48.3 and-49.8%,respectively） at 30℃ than at 22℃.The plasma concentration of high-density lipoprotein tended to be lower（P=0.08）,but the plasma concentrations of very low-density lipoprotein（VLDL）（P=0.09,＋50.0%） and nonestesterified fatty acid（NEFA）（P=0.04,＋44.2%） were higher in heat-stressed *** concluded that high ambient temperature depressed de novo fatty acid synthesis in both adipose tissues and the ***,β-oxidation of fatty acid in skeletal muscles was also inhibited in the high-temperature *** a result,more plasma NEFAs were used to synthesize VLDLs in the liver and were absorbed by adipose *** may be one reason that high ambient temperature enhances the accumulation of backfat and flare fat in finishing pigs.
