The role of exosomes in adult neurogenesis:implications for neurodegenerative diseases

Exosomes are cup-shaped extracellular vesicles with a lipid bilayer that is approximately 30 to 200 nm in thickness.Exosomes are widely distributed in a range of body fluids,including urine,blood,milk,and saliva.Exosomes exert biological function by transporting factors between different cells and by regulating biological pathways in recipient cells.As an important form of intercellular communication,exosomes are increasingly being investigated due to their ability to transfer bioactive molecules such as lipids,proteins,mRNAs,and microRNAs between cells,and because they can regulate physiological and pathological processes in the central nervous system.Adult neurogenesis is a multistage process by which new neurons are generated and migrate to be integrated into existing neuronal circuits.In the adult brain,neurogenesis is mainly localized in two specialized niches:the subventricular zone adjacent to the lateral ventricles and the subgranular zone of the dentate gyrus.An increasing body of evidence indicates that adult neurogenesis is tightly controlled by environmental conditions with the niches.In recent studies,exosomes released from different sources of cells were shown to play an active role in regulating neurogenesis both in vitro and in vivo,thereby participating in the progression of neurodegenerative disorders in patients and in various disease models.Here,we provide a state-of-the-art synopsis of existing research that aimed to identify the diverse components of exosome cargoes and elucidate the therapeutic potential of exosomal contents in the regulation of neurogenesis in several neurodegenerative diseases.We emphasize that exosomal cargoes could serve as a potential biomarker to monitor functional neurogenesis in adults.In addition,exosomes can also be considered as a novel therapeutic approach to treat various neurodegenerative disorders by improving endogenous neurogenesis to mitigate neuronal loss in the central nervous system.

An observed nocturnal ozone transport event in the Sichuan Basin,Southwestern China

The ozone(O_(3))pollution in China drew lots of attention in recent years,and the Sichuan Basin(SCB)was one of the regions confronting worsening O_(3)pollution problem.Many previous studies have shown that regional transport is an important contributor to O_(3)pollution.However,very few features of the O_(3)profile during transport have been reported,especially in the border regions between different administrative divisions.In this study,we conducted tethered balloon soundings in SCB during the summer of 2020 and captured a nocturnal O_(3)transport event during the campaign.Vertically,the O_(3)transport occurred in the bottom of the residual layer,between 200 and 500 m above ground level.Horizontally,the transport pathway was directed from southeast to northwest based on the analysis of the wind field and air mass trajectories.The effect of transport in the residual layer on the surface O_(3)concentration was related to the spatial distribution of O_(3).For cities with high O_(3)concentrations in the upwind region,the transport process would bring clean air masses and abate pollution.For downwind lightly polluted cities,the transport process would slow down the decreasing or even increase the surface O_(3)concentration during the night.We provided observational facts on the profile features of a transboundary O_(3)transport event between two provincial administrative divisions,which implicated the importance of joint prevention and control measures.However,the sounding parameters were limited and the quantitative analysis was preliminary,more integrated,and thorough studies of this topic were called for in the future.

Step Sintering of Microwave Heating and MicrowavePlasma Heating for Alumina Ceramics

The invention of microwave induced plasma during microwave heating makes it possible to confine plasma inside a crucible, in the pressure range of 1.33 Pa to 0.5 MPa, and above 900 degrees C. In this study, the step sintering of microwave heating and microwave plasma heating (SSMP) was proposed. In this process, microwave heating and plasma heating was ingeniously combined in one chamber, and vacuum was unnecessary for microwave induced plasma. In the first step, the microwave energy directly heated alumina ceramics to a given temperature, above which microwave will mainly ionize the gas and sustain a plasma. Then the plasma further heated alumina ceramics to the final temperature to finish sintering. Maintaining the advantages of microwave heating and plasma heating, this step sintering approach will overcome the limitation of both processes for processing ceramics, and it will be suitable for sintering all kinds of ceramics in principle.

Biomimetic“Gemini nanoimmunoregulators”orchestrated for boosted photoimmunotherapy by spatiotemporally modulating PD-L1 and tumor-associated macrophages

A novel strategy of not only stimulating the immune cycle but also modulating the immunosuppressive tumor microenvironment is of vital importance to efficient cancer immunotherapy.Here,a new type of spatiotemporal biomimetic“Gemini nanoimmunoregulators”was engineered to activate robust systemic photoimmunotherapy by integrating the triple-punch of amplified immunogenic cell death(ICD),tumor-associated macrophages(TAMs)phenotype reprogramming and programmed cell death ligand 1(PD-L1)degradation.The“Gemini nanoimmunoregulators”PM@RM-T7 and PR@RM-M2 were constructed by taking the biocompatible mesoporous polydopamine(mPDA)as nanovectors to deliver metformin(Met)and toll-like receptor 7/8 agonist resiquimod(R848)to cancer cells and TAMs by specific biorecognition via wrapping of red blood cell membrane(RM)inlaid with T7or M2 peptides.mPDA/Met@RM-T7(abbreviated as PM@RM-T7)was constructed to elicit an amplified in situ ICD effect through the targeted PTT and effectively stimulated the anticancer immunity.Meanwhile,PD-L1 on the remaining cancer cells was degraded by the burst metformin to prevent immune evasion.Subsequently,mPDA/R848@RM-M2(abbreviated as PR@RM-M2)specifically recognized TAMs and reset the phenotype from M2 to M1 state,thus disrupting the immunosuppressive microenvironment and further boosting the function of cytotoxic T lymphocytes.This pair of sister nanoimmunoregulators cooperatively orchestrated the comprehensive anticancer activity,which remarkably inhibited the growth of primary and distant 4T1 tumors and prevented malignant metastasis.This study highlights the spatiotemporal cooperative modalities using multiple nanomedicines and provides a new paradigm for efficient cancer immunotherapy against metastatic-prone tumors.

The material point method(MPM)has been gaining increasing popularity as an appropriate approach to the solution of coupled hydro-mechanical problems involving large deformation.In this paper,we survey the current state-of-the-art in the MPM simulation of hydro-mechanical behaviour in two-phase porous geomaterials.The review covers the recent advances and developments in the MPM and their extensions to capture the coupled hydro-mechanical problems involving large deformations.The focus of this review is aiming at providing a clear picture of what has or has not been developed or implemented for simulating two-phase coupled large deformation problems,which will provide some direct reference for both practitioners and researchers.

Molecular mechanism of flowering time regulation in Brassica rapa:similarities and differences with Arabidopsis

Properly regulated flowering time is pivotal for successful plant reproduction.The floral transition from vegetative growth to reproductive growth is regulated by a complex gene regulatory network that integrates environmental signals and internal conditions to ensure that flowering takes place under favorable conditions.Brassica rapa is a diploid Cruciferae species that includes several varieties that are cultivated as vegetable or oil crops.Flowering time is one of the most important agricultural traits of B.rapa crops because of its influence on yield and quality.The transition to flowering in B.rapa is regulated by several environmental and developmental cues,which are perceived by several signaling pathways,including the vernalization pathway,the autonomous pathway,the circadian clock,the thermosensory pathway,and gibberellin(GA)signaling.These signals are integrated to control the expression of floral integrators BrFTs and BrSOC1s to regulate flowering.In this review,we summarized current research advances on the molecular mechanisms that govern flowering time regulation in B.rapa and compare this to what is known in Arabidopsis.

Connections Between Induction Generator Effect and Open-loop Modal Proximity to Cause Sub-synchronous Oscillations in Series-compensated Power System

The induction generator effect(IGE)and the openloop modal proximity(OLMP)are two different reasons why subsynchronous oscillations(SSOs)in a series-compensated power system(SCPS)may occur.The IGE attributes the growing SSOs to negative resistance,while the OLMP explains the SSO mechanism from the standpoint of modal conditions.In this paper,we investigate the connections between the IGE and the OLMP through equivalent RLC circuit and open-loop modal analysis.Our investigation is conducted for two types of seriescompensated power systems where either a synchronous generator or a DFIG is connected at the sending end.The investigation reveals the conditions,in which the IGE and the OLMP may jointly cause the growing SSOs,i.e.,both the IGE and the OLMP can explain why the growing SSOs occur.Furthermore,the investigation indicates that the IGE and the OLMP may be totally irrelevant and lead to growing SSOs separately.This implies that it is possible that in a SCPS,the growing SSOs are only due to the IGE,and the OLMP is non-existent,and vice versa.Hence,when the growing SSOs occurs in a SCPS,examination based on both the IGE and the OLMP should be carefully conducted in order to find if the oscillatory instability is due to the IGE,or the OLMP,or both of them.

Low-frequency oscillation of train-network system considering traction power supply mode

The low-frequency oscillation(LFO)has occurred in the train-network system due to the introduction of the power electronics of the trains.The modeling and analyzing method in current researches based on electrified railway unilateral power supply system are not suitable for the LFO analysis in a bilateral power supply system,where the trains are supplied by two traction substations.In this work,based on the single-input and single-output impedance model of China CRH5 trains,the node admittance matrices of the train-network system both in unilateral and bilateral power supply modes are established,including three-phase power grid,traction transformers and traction network.Then the modal analysis is used to study the oscillation modes and propagation characteristics of the unilateral and bilateral power supply systems.Moreover,the influence of the equivalent inductance of the power grid,the length of the transmission line,and the length of the traction network are analyzed on the critical oscillation mode of the bilateral power supply system.Finally,the theoretical analysis results are verified by the time-domain simulation model in MATLAB/Simulink.

Enhanced red luminescence of Ca_(3)Si_(2−x)M_(x)O_(7):Eu^(3+)(M=Al,P)phosphors via partial substitution of Si^(4+) for applications in white light-emitting diodes

A red-emitting phosphor Ca_(2.91)Si_(2)O_(7):0.09Eu^(3+) with partial Al^(3+)/P^(5+) substitution on Si^(4+) was synthesized via a simple solid-state method,and the effects of the introduction of the M^(3+/5+)(M=Al,P)ions on the crystal structure and photoluminescence performance of Ca_(2.91)Si_(2−x)M_(x)O_(7):0.09Eu^(3+) phosphors were investigated.The X-ray diffraction(XRD),energy-dispersive X-ray spectroscopy(EDS),and X-ray photoelectron spectroscopy(XPS)results revealed that the structure of Ca_(3)Si_(2)O_(7) remained the same after the introduction of Al^(3+) and P^(5+) ions.The characteristic emission of Eu^(3+)-doped Ca_(3)Si_(2−x)M_(x)O_(7) phosphors exhibited two main peaks at 617 nm(red)and 593 nm(orange)under excitation at 394 nm,which originated from the^(5)D_(0)→^(7)F_(2)and^(5)D_(0)→^(7)F_(1) electron transitions of Eu^(3+) ions.After the partial substitution of Al^(3+) and P^(5+),the red emission intensities of the Ca_(2.91)Si_(2)O_(7):0.09Eu^(3+) phosphors were significantly enhanced by 1.88-and 1.42-fold,respectively,which is attributed to the crystal-field effect around Eu^(3+).Meanwhile,the luminescence intensities of the Ca_(2.91)Si_(1.96)Al_(0.04)O_(7):0.09Eu^(3+) and Ca_(2.91)Si_(1.94)P_(0.06)O_(7):0.09Eu^(3+) phosphors at 210℃ were 79.36%and 77.53%of those at 30°C,respectively,indicating their excellent thermal stability.Moreover,the as-prepared Ca_(2.91)Si_(1.96)Al_(0.04)O_(7):0.09Eu^(3+)and Ca_(2.91)Si_(1.94)P_(0.06)O_(7):0.09Eu^(3+) red-emitting phosphors were combined with a near-ultraviolet chip of 395 nm to fabricate red-light-emitting diode(LED)and white(w)-LED devices with excellent chromaticity features.In summary,Al^(3+)/P^(5+)-substituted Ca_(2.91)Si_(2)O_(7):0.09Eu^(3+) can serve as red-emitting phosphors for applications in w-LEDs.

Objective Human adenovirus(HAdV)infection is common and can develop to serious conditions with high mortality,yet the mechanism of HAdV infection remains unclear.In the present study,the serum metabolite profiles of HAdV-7-infected patients with pneumonia or upper respiratory tract infection(URTI)were explored.Methods In total,35 patients were enrolled in the study following an outbreak of HAdV-7 in the army,of whom 14 had pneumonia and 21 had URTI.Blood samples were collected at the acute stage and at the recovery stage and were analyzed by untargeted metabolomics.Results Over 90% of the differential metabolites identified between the pneumonia patients and URTI patients were lipids and lipid-like molecules,including glycerophospholipids,fatty acyls,and sphingolipids.The metabolic pathways that were significantly enriched were primarily the lipid metabolism pathways,including sphingolipid metabolism,glycerophospholipid metabolism,and linoleic acid metabolism.The sphingolipid metabolism was identified as a significantly differential pathway between the pneumonia patients and URTI patients and between the acute and recovery stages for the pneumonia patients,but not between the acute and recovery stages for the URTI patients.Ceramide and lactosylceramide,involved in sphingolipid metabolism,were significantly higher in the pneumonia patients than in the URTI patients with good discrimination abilities[area under curve(AUC)0.742 and 0.716,respectively;combination AUC 0.801].Conclusion Our results suggested that HAdV modulated lipid metabolism for both the patients with URTI and pneumonia,especially the sphingolipid metabolism involving ceramide and lactosylceramide,which might thus be a potential intervention target in the treatment of HAdV infection.
