Effect of propofol on generation of inflammatory mediator of monocytes

Objective:To evaluate the effect of propofol with different concentrations on the expression of inflammatory mediators of mterleukin and tumor-necrosis factor- α(TNF- α) by stimulating the mouse primary monocytes and human monocytic cell line with lipopolysaccharide(LPS)and also discuss the effeet of propofol on the secretion of inflammatory mediator and its possible molecular ***:The mononuclear cells of mouse spleen were separated and then purified to obtain the primary *** dose-effect relationship of production of pro-inflammatory cytokines by monocytes which were stimulated by LPS,namely the monocytes were stimulated by the dose of 0-500 ng/mL for 24 *** was employed to detect the concentration of IL-6,TL-8 and TNF-α.The effect of propofol on the secretion of above pro-inflammatory cytokines by the monocytes was *** were divided into the control group,the 0.1%DMSO group,the LPS group and the treatment group with LPS + different dose of propofol(propofol 1-100 μg/mL).ELISA was employed to detect the concentration of IL-6,IL-8 and TNF-α.The change in the expression of important signaling molecules in Toll-like receptor and NF-κ B signaling pathway was detected after THP-1 cells were treated with ***:The concentration of TNF- α was(3 863±153)pg/mL after 12 h of stimulation by LPS and then its concentration was decreased *** the concentration of IL-6 and IL-8 was relatively high after 24 h of stimulation by LPS,(5 627±330) pg/mL and(1 626±200) pg/mL,*** propofol could inhibit the expression of IL-6,IL-8 and TNF- α caused by *** the intervention treatment of 50 μg/mL propofol,the concentration of IL-6,IL-8 and TNF-α was significantly decreased(Plusions:The propofol can inhibit the expression of TLR-4 and NF-κ B to inhibit the activation of p38 and the expression of pro-inflammatory cytokines.

PROPULSIVE PERFORMANCE AND VORTEX SHEDDING OF A FOIL IN FLAPPING MOTION

The propulsive performance and vortex shedding of oscillating foil, whichmimics biological locomotion, were numerically investigated. The objectives of this study were todeal with unsteady force, in particular thrust force, exerted on the foil in pitching and plungingmotions, and to explore the relation of the propulsive performance with vortex structures near thefoil and vortex shedding in the near wake. The two-dimensional incompressible Navier-Stokesequations in the vorticity and stream-function formulation were solved by fourth-order essentiallycompact finite difference schemes for the space derivatives and a fourth-order Runge-Kutta schemefor the time advancement. To reveal the mechanism of the propulsive performance, the unsteady forceand the shedding of the trailing- and leading-edge vortices of the foil were analyzed. The effectsof some typical factors, such as the frequency and amplitude of the oscillation, the phasedifference between the pitching and plunging motions, and the thickness ratio of the foil, on thevortex shedding and unsteady force were discussed.

Sensitive determination of 4-O-methylhonokiol in rabbit plasma by high performance liquid chromatography and application to its pharmacokinetic investigation

A novel high performance liquid chromatographic method was developed for the determination of 4-O- methylhonokiol in rabbit plasma and was applied to its pharmacokinetic investigation. Plasma samples were treated by one-fold volume of methanol and acetonitrile to remove the interference proteins. A reverse phase column of SHIM- PACK VP-ODS （150 mm × 4.6 mm, 5.0 μm） was used to separate 4-O-methylhonokiol in the plasma samples. The detection limit of 4-O-methylhonokiol was 0.2 μg/L and the linear range was 0. 012 - 1. 536 mg/L. The good extraction recoveries were obtained for the spiked samples （84.7%, 89.3% and 87.7% for low, middle and high concentrations of added standards, respectively）. The relative standard deviation of intra-day and inter-day precisions was in the range from 0.6% to 13.5%. The pharmacokinetic study of 4-O-methylhonokiol was made and the results from the plasmaconcentration curve of 4-0-methylhonokiol showed a two-apartment open model. This work developed a sensitive, stable and rapid HPLC method for the determination of 4-O-methylhonokiol and the developed method has been successfully applied to a pharmacokinetic study of 4-O-methylhonokiol.

l-Acetoxy allylic phosphonates rearrange regioselectively to γ-acetoxy α,β-unsaturated phosphonates under the catalysis of Pd(0). l-Acetoxy allylic phosphonates act as the precursors of phosphonatedπ-allylic cations in palladium(0) catalyzed reactions[1]. The reactions of l-acetoxy allylic phosphonates under palladium(0) catalysis with nucleophiles have been reported. 1-4 These reactions are highly regio- and stereoselective. We wish to report here the palladium(0) catalyzed reaction of l-acetoxy allylic phosphonates in the absence of a nucleophile.

为实现玉米杂交种的自动化快速分选,提出了应用少量近红外波段光对玉米种子进行成像,获取种子光谱图像并提取纹理特征来鉴定玉米杂交种纯度的方法。采集5个玉米品种的母本和杂交种在4个短波近红外波段的透射光谱图像和4个中波近红外波段的反射光谱图像,采用白板标定校正光谱图像,运用中值滤波、大津法去除噪声,从背景中分割出种子,应用灰度分布统计,灰度共生矩阵提取纹理特征,对同一粒种子拼接其在各波长处的特征数据,应用主成分分析和正交线性判别分析降维并获得子空间的最佳可分性,使用支持向量机建立透射和反射光谱图像纯度鉴定模型。透射和反射模型对5个玉米品种平均正确鉴别率均在85%以上。表明利用少量波段的近红外光谱图像鉴定玉米杂交种纯度是可行的。

Three-dimensional morphological characteristics measurement of ankle joint based on computed tomography image post-processing

As precise positioning of ankle radiography is not possible, quantitative measurement of all syndesmotic parameters on repeated ankle X-ray films may be of little value. The purpose of this study was to provide a set of scientific and objective evaluation criteria for assessing the quality of ankle fracture reduction accurately and reliably by an intelligent combining three-dimensional （3-D） computed tomography （CT） measurement model. Methods From June 2008 to March 2011, all the thin-slice CT images of 100 cases （50 males and 50 females） with normal ankle joint scanned by 16-slice spiral CT were collected. Two-dimensional （2-D） and 3-D images of ankle joints were generated by using multiple planar reconstruction （MPR） and surface shaded display （SSD） respectively. The relevant parameters about bone structures and their relationship were measured and analyzed based on 3-D topological narrow division technique and 3-D measurement techniques combining essential elements of point, line and surface. Results In this study, the mean distance from lateral malleolus tip to talocrural articular surface, the tip of medial malleolus anterior colliculus to talocrural articular surface and lateral malleolus tip to the tip of medial malleolus anterior colliculus were （22.83±1.12） mm, （12.84±1.09） mm, and （61.18±2.03） mm respectively in male group, and （20.16±1.00） mm, （10.30±1.05） mm and （53.00±1.40） mm respectively in female group. The above three parameters were correlated with gender, height and weight （P 〈0.05）. However, the mean perpendicular distance from lateral malleolus tip to the plane through the tip of medial malleolus anterior colliculus, the talocrural angle, later clear space, medial clear space, and the superior clear space were （9.93±0.29） mm, （10.01±0.38）°, （1.94±0.16） mm, （2.78±0.19） mm, and （3.14±0.15） mm respectively in 100 cases, were not significance correlated with gender, height and weight （P 〉0.05）.
