Rational surface charge engineering of haloalkane dehalogenase for boosting the enzymatic performance in organic solvent solutions

Biocatalysis in organic solvents(OSs)has numerous important applications,but native enzymes in OSs often exhibit limited catalytic ***,we proposed a computation-aided surface charge engineering strategy to improve the catalytic performance of haloalkane dehalogenase DhaA in OSs based on the energetic analysis of substrate binding to the DhaA *** variants with enhanced OS resistance were obtained by replacing negative charged residues on the surface with positive charged residue(Arg).Particularly,a four-substitution variant E16R/E93R/E121R/E257R exhibited the best catalytic performance(five-fold improvement in OS resistance and seven-fold half-life increase in 40%(vol)dimethylsulfoxide).As a result,the overall catalytic performance of the variant could be at least 26 times higher than the wild-type *** spectroscopy and molecular dynamics simulation studies revealed that the residue substitution mainly enhanced OS resistance from four aspects:(a)improved the overall structural stability,(b)increased the hydrophobicity of the local microenvironment around the catalytic triad,(c)enriched the hydrophobic substrate around the enzyme molecule,and(d)lowered the contact frequency between OS molecules and the catalytic *** findings validate that computationaided surface charge engineering is an effective and ingenious rational strategy for tailoring enzyme performance in OSs.

Noise-induced hearing loss(NIHL) is a highly prevalent form of sensorineural hearing damage that has significant negative effects on individuals of all ages and there are no effective drugs approved by the US Food and Drug Administration. In this study, we unveil the potential of superparamagnetic iron oxide nanoparticle assembly(SPIOCA) to reshape the dysbiosis of gut microbiota for treating NIHL. This modulation inhibits intestinal inflammation and oxidative stress responses, protecting the integrity of the intestinal barrier. Consequently, it reduces the transportation of pathogens and inflammatory factors from the bloodstream to the cochlea. Additionally, gut microbiota-modulated SPIOCA-induced metabolic reprogramming in the gut–inner ear axis mainly depends on the regulation of the sphingolipid metabolic pathway, which further contributes to the restoration of hearing function. Our study confirms the role of the microbiota–gut–inner ear axis in NIHL and provides a novel alternative for the treatment of NIHL and other microbiota dysbiosis-related diseases.

Using data from the regional broadband dense temporary array deployed by the ChinArray project,we applied the three-dimensional(3D)Kirchhoff migration method of the teleseismic P-wave receiver function to investigate discontinuity structures of the mantle transition zone(MTZ)in the central and western parts of the North China Craton(NCC)using a highresolution 3D velocity model of the East Asian *** results show that the 410-km discontinuity beneath the Datong Volcano is depressed by~10 km,indicating the presence of a high-temperature anomaly near the depth of 410 km,which is likely related to small-scale mantle upwelling caused by the dehydration of the stagnant Pacific Plate in the *** upwelling of hot material provides a heat source for surface magmatic *** the Bohai Bay Basin,significant subsidence of the 660-km discontinuity is observed,and the transition zone here is extensively ***’s suggested that the anomalies in this region are related to the stagnation of the Pacific slab in the *** the thickness of the transition zone west of the North-South Gravity Lineament appears normal,we propose that the subducting front of the Pacific slab did not cross the gravity lineament in the *** comparison,the small-scale subsidence of the 660-km discontinuity and the thickening of the MTZ observed north of the Hannuoba Volcano likely indicate that the slab crossed the gravity lineament at its turning point and remained in the ***,a local thickening of the MTZ is observed in the Dabie orogenic belt of the Qinling *** is believed to be a combined effect of lithospheric delamination into the transition zone in the lower yangtze region and the stagnation of the Pacific Plate.

We present a numerically stable one-point quadrature rule for the stiffness matrix and mass matrix of the three-dimensional numerical manifold method(3D NMM).The rule simplifies the integration over irregularly shaped manifold elements and overcomes locking issues,and it does not cause spurious modes in modal *** essential idea is to transfer the integral over a manifold element to a few moments to the element center,thereby deriving a one-point integration rule by the moments and making modifications to avoid locking *** the stiffness matrix,after the virtual work is decomposed into moments,higher-order moments are modified to overcome locking issues in nearly incompressible and bending-dominated *** the mass matrix,the consistent and lumped types are derived by *** particular,the lumped type has the clear advantage of *** proposed method is naturally suitable for 3D NMM meshes automatically generated from a regular *** tests justify the accuracy improvements and the stability of the proposed procedure.

The design of heat exchangers in the advanced supercritical power conversion system cannot be separated from the study of heat transfer ***-side heating mode is often encountered for solar receiver and supercritical ***,the characteristics of supercritical CO_(2)(sCO_(2))convection heat transfer in vertical tubes with circumferentially half-side heating was numerically investigated through the SST k-ωturbulent model which matches well with the experimental ***,heat transfer between sCO_(2)upflow and downflow was *** to film boiling heat transfer at subcritical pressure,numerical results were processed according to the supercritical pseudo-phase transition hypothesis,with liquid-like phase in the tube core region and vapor-like film in the region near the heated tube *** structure of two layers was demarcated by pseudo-critical temperature T_(pc).Therefore,sCO_(2)heat transfer was assessed according to double thermal resistances caused by vapor-like film near the wall and core liquid-like *** findings suggest that wall temperature for upflow is higher than that for downflow,which is attributed to larger thermal resistance in the fluid domain for upflow than that for *** difference guarantees the excellent heat transfer performance for downflow than *** is also further concluded that the formation of vapor-like film near the wall due to pseudo-phase transition plays a key role in dominating wall temperature and inducing heat transfer deterioration in half-side heating *** present contribution is significant to the design of supercritical heat exchanger under half-side heating mode.

Improvement of the piezoelectricity of PVDF-HFP by CoFe_(2)O_(4)nanoparticles

High piezoelectric composite films composed of poly(vinylidene fluoride-co-hexafluoropropylene)(PVDF-HFP)and ferromagnetic cobalt ferrite(CoFe_(2)O_(4))(0.00 wt%to 0.2 wt%)are prepared by a solution casting method accompanied by uniaxial stretching and high electric field *** decisive effect of the poling electric field on the power generating capability was confirmed by the *** pure PVDF-HFP films,when the maximum electric field Emax is 120 MV/m,the calibrated open circuit voltage reaches 2.93 V,which is much higher than those poled at lower electric fields(70 MV/m:1.41 V;90 MV/m:2.11 V).Furthermore,the addition of CoFe_(2)O_(4)also influences the piezoelectricity *** the samples containing 0.15 wt%CoFe_(2)O_(4),the calibrated open circuit voltage increases to the maximum value of 3.57 ***,the relative fraction of theβ-phase and the crystallinity degree are 99%and 48%,*** effects of CoFe_(2)O_(4)nanoparticles on initial crystallization,uniaxial stretching and high electric field poling are investigated by XRD,FTIR and DSC.
