Objective:To investigate the anti-tumor effect of γδ T cells on gastric adenocarcinoma SGC-7901 at cellular level in vitro and animal model in vivo,and to explore whether the c-Met antibodies and/or compounds co-administration could enhance the killing effect for tumors and influence the gene and protein expression of c-Met and ***:Firstly,peripheral blood mononuclear cells(PBMCs) were freshly isolated from healthy volunteers,and γδ T cells with high purity were obtained after selectively amplification stimulated with Zol and ***,Real Time Cell Analyser system(RTCA) was applied to measure cytotoxicity of γδ T cells on SGC-7901 cells at different effector-target ratios(E:T).Thirdly,animal model of SGC-7901 in Balb/c nude mice was established to explore the anti-tumor effect of γδ T cells alone or co-administration with c-Met antibodies and/or ***,RT-PCR and Western Blot experiments were carried out to detect the influence for c-Met and Fas ***:The purity of γδ T cells could reach 90.2%for 2-week cultivation in vitro via stimulation with Zol+*** screening results showed that γδ T cells could inhibit SGC-7901 tumor cell growth in a concentration-dependent *** vivo mouse model experiment indicated that y5 T cells could obviously inhibit the tumor growth,and the inhibition ability was enhanced after co-administration with c-Met antibodies and/or ***-PCR and Western Blot experiments showed that c-Met and Fas expressions were effectively inhibited and promoted respectively,and the effects could been enhanced after co-administration with c-Met antibodies and ***:γδ T cells showed the anti-tumor activity both in vitro and in vivo,and the effects could be enhanced after co-administration with c-Met antibodies and *** c-Met and Fas expression may be inhibited or promoted,respectively,during the process of *** study indicated that γδ T cells appear to be a promising tool for n

Predicting submerged vegetation drag with a machine learning-based method

Accurate estimation of the drag forces generated by vegetation stems is crucial for the comprehensive assessment of the impact of aquatic vegetation on hydrodynamic processes in aquatic *** coupling relationship between vegetation layer flow velocity and vegetation drag makes precise prediction of submerged vegetation drag forces particularly *** present study utilized published data on submerged vegetation drag force measurements and employed a genetic programming(GP)algorithm,a machine learning technique,to establish the connection between submerged vegetation drag forces and flow and vegetation *** using the bulk velocity,U,as the reference velocity scale to define the drag coefficient,C_(d),and stem Reynolds number,the GP runs revealed that the drag coefficient of submerged vegetation is related to submergence ratio(H^(*)),aspect ratio(d^(*)),blockage ratio(ψ^(*)),and vegetation density(λ).The relation between vegetation stem drag forces and flow velocity is implicitly embedded in the definition of C_(d).Comparisons with experimental drag force measurements indicate that using the bulk velocity as the reference velocity,as opposed to using the vegetation layer average velocity,U_(v),eliminates the need for complex iterative processes to estimate U_(v)and avoids introducing additional errors associated with U_(v)*** approach significantly enhances the model’s predictive capabilities and results in a simpler and more user-friendly formula expression.

DEM analysis of the influence of stirrer design on die filling with forced powder feeding

Die filling is a critical stage during powder compaction,which can significantly affect the product quality and *** this paper,a forced feeder is introduced attempting to improve the filling performance of a lab-scale die filling *** die filling process is analysed with a graphics processing units(GPU)enhanced discrete element method(DEM).Various stirrer designs are assessed for a wide range of process settings(i.e.,stirrer speed,filling speed)to explore their influence on the die filling performance of free-flowing *** results show that die filing with the novel helical-ribbon(i.e.,type D)stirrer design exhibits the highest filling ratio,implying that it is the most robust stirrer design for the feeder configuration ***,die filling performance with the type D stirrer design is a function of the stirrer speed and the filling speed.A positive variation of filling ratio(ηf>0%)can be ensured over the whole range of filling speed by adjusting the stirrer speed(i.e.,increasing the stirrer speed).The approach used in this study can not only help understand how the stirrer design affects the die filling performance but also guide the optimization of feeder system and process settings.

Prediction of discharge in a tidal river using the LSTM-based sequence-to-sequence models

The complexity of river-tide interaction poses a significant challenge in predicting discharge in tidal *** short-term memory(LSTM)networks excel in processing and predicting crucial events with extended intervals and time delays in time series ***,the sequence-to-sequence(Seq2Seq)model,known for handling temporal relationships,adapting to variable-length sequences,effectively capturing historical information,and accommodating various influencing factors,emerges as a robust and flexible tool in discharge *** this study,we introduce the application of LSTM-based Seq2Seq models for the first time in forecasting the discharge of a tidal reach of the Changjiang River(yangtze River)*** study focuses on discharge forecasting using three key input characteristics:flow velocity,water level,and discharge,which means the structure of multiple input and single output is *** experiment used the discharge data of the whole year of 2020,of which the first 80%is used as the training set,and the last 20%is used as the test *** means that the data covers different tidal cycles,which helps to test the forecasting effect of different models in different tidal cycles and different *** experimental results indicate that the proposed models demonstrate advantages in long-term,mid-term,and short-term discharge *** Seq2Seq models improved by 6%-60%and 5%-20%of the relative standard deviation compared to the harmonic analysis models and improved back propagation neural network models in discharge prediction,*** addition,the relative accuracy of the Seq2Seq model is 1%to 3%higher than that of the LSTM *** assessment of the prediction errors shows that the Seq2Seq models are insensitive to the forecast lead time and they can capture characteristic values such as maximum flood tide flow and maximum ebb tide flow in the tidal cycle *** indicates the significance of the Seq2Seq models.

Heavy-atom engineered hypoxia-responsive probes for precisive photoacoustic imaging and cancer therapy

Photoacoustic agents combining photodynamic therapy(PDT) and photothermal therapy(PTT) functions have emerged as potent theranostic agents for combating cancer. The molecular approaches for enhancing the near-infrared(NIR)-absorption and maximizing non-radiative energy transfer are essential for effective photoacoustic imaging(PAI) and therapy applications. In addition, such molecules with high specificity and affinity to cancer cells are urgently needed, which would further decrease the side effect during treatments. In this study, we applied a heavy-atom engineering strategy and introduced p-aminophenol,-thio, and-seleno moieties into NIR heptamethine cyanine(Cy7) skeleton(Cy7-X-NH_(2), X = O, S, Se) to significantly increase photothermal conversion efficiency for PTT and promote intersystem crossing for ***, we designed a series of nitroreductase(NTR)-activated photoacoustic probes(Cy7-X-NO_(2),X = O, S, Se), and target hypoxic tumors with NTR overexpression. Our prostate cancer targeting probe,Cy7-Se-NO_(2)-KUE, exhibited specific tumor photoacoustic signals and effective tumor killing through outstanding synergistic PTT/PDT in vivo. These findings highlighted a versatile strategy for cancer photoacoustic diagnosis and enhanced phototherapy.

Ultrathin two-dimensional medium-entropy oxide as a highly efficient and stable electrocatalyst for oxygen evolution reaction

Medium-entropy oxides(MEOs)with broad compositional tunability and entropy-driven structural stability,are receiving booming attention as a promising candidate for oxygen evolution reaction(OER)***,ultrathin two-dimensional(2D)nanostructure offers extremely large specific surface area and is therefore considered to be an ideal catalyst ***,it remains a grant challenge to synthesize ultrathin 2D MEOs due to distinct nucleation and growth kinetics of constituent multimetallic elements in 2D anisotropic *** this work,an ultrathin 2D MEO(MnFeCoNi)O was successfully synthesized by a facile and low-temperature ionic layer epitaxy *** from multi-metal synergistic effects within ultrathin 2D nanostructure,this 2D MEO(MnFeCoNi)O revealed excellent OER electrocatalytic performance with a quite low overpotential of 117 mV at 10 mA·cm^(-2) and an impressive stability for 120 h continuous operation with only 6.9%***,the extremely high mass activity(5584.3 A·g^(-1))was three orders of magnitude higher than benchmark RuO_(2)(3.4 A·g^(-1))at the same overpotential of 117 *** work opens up a new avenue for developing highly efficient and stable electrocatalysts by creating 2D nanostructured MEOs.
