Rapid alkalinization factors(RALFs)in plants have been reported to dampen pathogenassociated molecular pattern(PAMP)-triggered immunity via suppressing PAMP-induced complex formation between the pattern recognition receptor(PRR)and its co-receptor ***,the direct and positive role of RALFs in plant immunity remains largely ***,we report the direct and positive roles of a typical RALF,RALF22,in plant ***22alone directly elicited a variety of typical immune responses and triggered resistance against the devastating necrotrophic fungal pathogen Sclerotinia sclerotiorum in a FERONIA(FER)-dependent ***(LRE)-like glycosylphosphatidylinositol(GPI)-anchored protein 1(LLG1)and NADPH oxidase RBOHD were required for RALF22-elicited reactive oxygen species(ROS)*** mutation of cysteines conserved in the C terminus of RALFs abolished,while the constitutive formation of two disulfide bridges between these cysteines promoted the RALF22-elicited ROS production and resistance against ***,demonstrating the requirement of these cysteines in the functions of RALF22 in plant ***,RALF22 amplified the Pep3-induced immune signal by dramatically increasing the abundance of PROPEP3 transcript and *** with RALF22 induced resistance against *** in Brassica crop ***,our results reveal that RALF22 triggers immune responses and augments the Pep3-induced immune signal in a FER-dependent manner,and exhibits the potential to be exploited as an immune elicitor in crop protection.

Extrachromosomal DNA(ecDNA) is a form of circular DNA mostly found in tumor cells. Unlike the typical chromosomal DNA, ecDNA is circular, self-replicating, and carries complete or partial gene fragments. Although ecDNA occurrence remains a rare event in cancer, recent studies have shown that oncogene amplification on ecDNA is widespread throughout many types of cancer, implying that ecDNA plays a central role in accelerating tumor evolution. ecDNA has also been associated with increased tumor mutation burden, chromosomal instability, and even tumor microenvironment remodeling. ecDNA may be crucial in influencing tumor heterogeneity, drug sensitivity,oncogenic senescence, and tumor immunogenicity, leading to a worsening prognosis for tumor patients. In this way, several clinical trials have been conducted to investigate the importance of ecDNA in clinical treatment. In this review, we summarize the biogenesis, characteristics, and current research methods of ecDNA, discuss the vital role of ecDNA-caused tumor heterogeneity in cancers, and highlight the potential role of ecDNA in cancer therapy.

舰载光电跟踪设备在跟踪百公里以上的目标时,由于受到障碍物干扰,目标有时可能从视场中丢失,需采用记忆跟踪算法对目标的未来时刻位置进行预测,重新找回目标。常规的CA、CV模型预测目标时忽略了残差,记忆跟踪时间短,从而造成预测目标不够精确。针对以上问题,提出了Kalman目标预测模型,延长记忆跟踪时间。首先,由船地坐标转换公式推导了甲板坐标系下船摇速度,前馈到伺服控制系统速度回路中,保证视轴自稳定,同时提高跟踪精度;其次,概述了CA、CV、Kalman目标预测模型;最后,重点论述了3种目标预测模型记忆跟踪和实时雷达引导二维位置信息之间的关系。试验结果表明,本文由于引入了Kalman目标预测模型,使得记忆跟踪时间比传统的CA、CV模型的预测目标时间提高了一个数量级。解决了工程中舰载光电跟踪设备受船摇影响时跟踪精度低和记忆跟踪时间短的问题。

Background Circulating tumor DNA(ctDNA)is a promising biomarker for predicting relapse in multiple solid ***,the predictive value of ctDNA for disease recurrence remains indefinite in locoregional gastric cancer(GC).Here,we aimed to evaluate the predictive value of ctDNA in this *** From 2016 to 2019,100 patients with stage II/III resectable GC were recruited in this prospective cohort study(NCT02887612).Primary tumors were collected during surgical resection,and plasma samples were collected perioperatively and within 3 months after adjuvant chemotherapy(ACT).Somatic variants were captured via a targeted sequencing panel of 425 cancer-related *** plasma was defined as ctDNA-positive only if one or more variants detected in the plasma were presented in at least 2%of the primary *** Compared with ctDNA-negative patients,patients with positive postoperative ctDNA had moderately higher risk of recurrence[hazard ratio(HR)=2.74,95%confidence interval(CI)=1.37–5.48;P=0.003],while patients with positive post-ACT ctDNA showed remarkably higher risk(HR=14.99,95%CI=3.08-72.96;P<0.001).Multivariate analyses indicated that both postoperative and post-ACT ctDNA positivity were independent predictors of recurrence-free survival(RFS).Moreover,post-ACT ctDNA achieved better predictive performance(sensitivity,77.8%;specificity,90.6%)than both postoperative ctDNA and serial cancer antigen.A comprehensive model incorporating ctDNA for recurrence risk prediction showed a higher C-index(0.78;95%CI=0.71–0.84)than the model without ctDNA(0.71;95%CI=0.64–0.79;P=0.009).Conclusions Residual ctDNA after ACT effectively predicts high recurrence risk in stage II/III GC,and the combination of tissue-based and circulating tumor features could achieve better risk prediction.

Molecular Characteristics of S1 Gene of Infectious Bronchitis Virus Isolated from Chicken Proventriculus

Infectious bronchitis virus was isolated from swollen proventriculi of clinically ill chicken. The suspected virus samples (2/97, 3/97, 1/98) were adapted in SPF chicken embryos for virus isolation and identification. All the virus isolates were able to agglutinate chicken erythrocytes after treatment with trypsin, and interfer with the reproduction of Newcastle disease virus in chicken embryos, and have low antigenic relat-edness values with reference positive IBV. The isolates 2/97, 3/97, 1/98 RNAs extracted from the allantoic fluid of inoculated embryonated eggs were converted to cDNA by reverse transcription with 3'-primer of S1 gene of (IBV). Polymerase chain reaction (PCR) was performed with two primers which span the S1 gene. Amplified product of 1. 93 kb was subjected to EcoR I and BamH I digestion and the fragments obtained were the same as expected size. The PCR product was ligated to pBlueScript-SK ( + ) vector, and its nucleotide sequence was determined by the dideoxy-mediated chain termination method. Nucleotide sequence analysis showed 73. 6 - 99. 7% homology between the isolated IBV and the IBV strains in GenBank. The homology of amino acid was 71. 4 - 99.4%.
