TCP-1,a novel peptide to diagnose early colon cancer

A nine cyclic peptide(TCP-1)showed excellent specificity for colon ***-1 binds with human tumor tissues at early stages and mice tumor with diameters of 1-4 mm,suggesting that TCP-1 may be used for early diagnosis of colon *** mechanism of the targeted binding of TCP-1 to colon cancer was also studied using immunoprecipitation,LC-MS and *** screening and identifying of the possible binding target proteins of TCP-1,keratin,typeⅡcytoskeletal 5 was speculated to be the specific binding target protein of TCP-1 in human tumor *** studies were conducted to investigate the target-mediated drug disposition of the new tumor-specific peptide by LC-MS/*** tissue distribution study showed that TCP-1 was found only in colon tumors(the target site)in tumor mice did not bind to any other *** TCP-1 to tumor markedly increased its removal rate from blood circulation but mildly extended its staying time in *** tumor mice,a lower AUC of TCP-1(reduced by almost 35%)and 2-fold higher clearance were found compared to that of normal *** proposed metabolic pathway of TCP-1 in the kidney was also determined using LC-MS^(n)-*** high specificity and low toxicity of the peptide may be caused by its extremely tight binding to the *** applications for future clinical use,including MRI and PET/CT were also explored,and this research may promote the development of colon cancer diagnostic technology research and provide new ideas and technical routes for tumor diagnostic technology.

Sedimentary process research is of great significance for understanding the distribution and characteristics of *** the detailed observation and measurement of the Sangyuan outcrop in Luanping Basin,this paper studies the depositional process of the hyperpycnal flow deposits,and divides their depositional process into three phases,namely,acceleration,erosion and *** the acceleration phase,hyperpycnal flow begins to enter the basin nearby,and then speeds up *** developed in the acceleration phase are *** addition,the original deposits become unstable and are taken away by hyperpycnal flows under the eroding *** a result,there are a lot of mixture of red mud pebbles outside the basin and gray mud pebbles within the *** the erosion phase,the reverse deposits are eroded and become thinner or even ***,no reverse grading characteristic is found in the proximal major channel that is closer to the source,but it is still preserved in the middle branch channel that is far from the *** entering the deceleration phase,normally grading deposits appear and cover previous *** final deposits in the basin are *** are reverse,and others are *** are superimposed with each other under the action of hyperpycnal *** analysis of the Sangyuan outcrop demonstrates the sedimentary process and distribution of hyperpycnites,and reasonably explain the sedimentary characteristics of *** is helpful to the prediction of oil and gas exploration targets in gravity flow deposits.

Extracellular membrane proteins are crucial for mediating cell attachment,recognition,and signal transduction in the testicular microenvironment,particularly germline stem *** 18(CDH18),a type Ⅱ classical cadherin,is primarily expressed in the nervous and reproductive ***,we investigated the expression of CDH18in neonatal porcine prospermatogonia(ProSGs)and murine spermatogonial stem cells(SSCs).Disruption of CDH18 expression did not adversely affect cell morphology,proliferation,self-renewal,or differentiation in cultured porcine ProSGs,but enhanced cell adhesion and prolonged cell *** analysis indicated that the down-regulation of CDH18 in ProSGs significantly up-regulated genes and signaling pathways associated with cell *** further elucidate the function of CDH18 in germ cells,Cdh18 knockout mice were generated,which exhibited normal testicular morphology,histology,*** analysis showed increased expression of genes associated with adhesion,consistent with the observations in porcine *** interaction of CDH18withβ-catenin and JAK2 in both porcine ProSGs and murine SSCs suggested an inhibitory effect on the canonical Wnt and JAK-STAT signaling pathways during CDH18 ***,these findings highlight the crucial role of CDH18 in regulating cell adhesion in porcine ProSGs and mouse *** this regulatory mechanism provides significant insights into the testicular niche.

Within the electronic design automation(EDA) domain, artificial intelligence(AI)-driven solutions have emerged as formidable tools, yet they typically augment rather than redefine existing methodologies. These solutions often repurpose deep learning models from other domains, such as vision, text, and graph analytics, applying them to circuit design without tailoring to the unique complexities of electronic circuits. Such an “AI4EDA” approach falls short of achieving a holistic design synthesis and understanding,overlooking the intricate interplay of electrical, logical, and physical facets of circuit data. This study argues for a paradigm shift from AI4EDA towards AI-rooted EDA from the ground up, integrating AI at the core of the design process. Pivotal to this vision is the development of a multimodal circuit representation learning technique, poised to provide a comprehensive understanding by harmonizing and extracting insights from varied data sources, such as functional specifications, register-transfer level(RTL) designs, circuit netlists,and physical layouts. We champion the creation of large circuit models(LCMs) that are inherently multimodal, crafted to decode and express the rich semantics and structures of circuit data, thus fostering more resilient, efficient, and inventive design methodologies. Embracing this AI-rooted philosophy, we foresee a trajectory that transcends the current innovation plateau in EDA, igniting a profound “shift-left” in electronic design methodology. The envisioned advancements herald not just an evolution of existing EDA tools but a revolution, giving rise to novel instruments of design-tools that promise to radically enhance design productivity and inaugurate a new epoch where the optimization of circuit performance, power, and area(PPA) is achieved not incrementally, but through leaps that redefine the benchmarks of electronic systems' capabilities.

This paper takes the total yield of products that need refrigerated transport as the impact factors of transport aggregate of cold chain logistics,such as meat,aquatic products,quick-frozen noodle,fruits,vegetables,dairy,and *** selecting the consumption data of urban residents on transported products via cold chain in Jiangsu Province from 2005 to 2000 as sample,this paper establishes grey prediction model GM(1,1) of cold chain logistics demand and uses DPS7.05 software for test,to predict the cold chain logistics demand of urban residents in Jiangsu Province during the Twelfth Five-Year Plan *** results show that in the period 2010-2015,the cold chain logistics demand of urban residents in Jiangsu Province is 1 151.589 1,1 185.136 6,1 219.661 3,1 255.191 8,1 291.757 3,1 329.388 1 t respectively;in the period 2005-2010,the cold chain logistics demand of urban residents in Jiangsu Province increases at annual growth rate of 3.9%;in the period 2011-2015,the growth rate declines to some extent,increasing slowly at rate of 2.9%.

Heat shock induction of a 65 kDa ATP-binding proteinase in rat C6 glioma cells

The 45, 55, 65 and 100 kDa ATP-binding proteinases (ATP-BPases) of the heat-shocked (44 ℃ for 30 min, recovery for 12h) rat C6 glioma cells were purified by DEAE-ionexchange and ATP-affinity chromatography. Their molecular masses, isoelectric points (pI), pH-optima and other properties were analyzed by native proteinase *** was shown that the 65 kDa ATP-BPase is specifically induced by heat shock and not detectable in control *** N-terminal 1-9 amino acid sequence was determined by Edman degradation, but no homologies to other proteins in the protein data bases were found. 30 and 31 kDa proteinases can be cleaved from the 45, 55 and 65 kDa proteinases to which they are linked. A possible relationship of the heat-induced 65 kDa ATP-BPase with the ATP-dependent proteinases (ATP-DPases) in prokaryotes and eukaryotes is discussed.

表征酶与底物有效吸附是认识纤维素酶促水解反应过程、揭示预处理作用机制的重要手段之一。成功构建了纤维素外切酶(Cel7A)、纤维素内切酶(Cel5A)和葡萄糖苷酶(BglⅠ)荧光探针,优化吸附条件,并考察其对细胞壁的吸附。结果表明,Cel7A、Cel5A、BglⅠ基因N端分别连接至荧光蛋白基因mCherry、eGFP、eBFP C端,依次连入pPICZα载体,成功在毕赤酵母(X33)菌株表达;mCherry-Cel7A、eGFP-Cel5A和eBFP-BglⅠ荧光探针的最佳激发波长分别为573、469 nm和385 nm,最佳发射波长分别为613、511 nm和447 nm;以滤纸为底物,测定最佳吸附浓度分别为0.058、0.19 U/mL和1.93 U/mL,最佳吸附时间分别为7、7 min和6 min,最佳观察时间分别为min、<12 s和<12 s。纤维素酶荧光探针能够结合生物质样品,并反应出不同底物与纤维素酶亲和力的差异。上述工作为生物质抗降解屏障、生物质预处理、纤维素高效酶促水解等研究提供了技术支持。
