Real-time model correction using Kalman filter for Raman-controlled cell culture processes

Raman spectroscopy has found extensive use in monitoring and controlling cell culture *** this context,the prediction accuracy of Raman-based models is of paramount ***,models established with data from manually fed-batch cultures often exhibit poor performance in Raman-controlled ***,there is a need for effective methods to rectify these *** objective of this paper is to investigate the efficacy of Kalman filter(KF)algorithm in correcting Raman-based models during cell ***,partial least squares(PLS)models for different components were constructed using data from manually fed-batch cultures,and the predictive performance of these models was ***,various correction methods including the PLS-KF-KF method proposed in this study were employed to refine the PLS ***,a case study involving the auto-control of glucose concentration demonstrated the application of optimal model correction *** results indicated that the original PLS models exhibited differential performance between manually fed-batch cultures and Raman-controlled *** glucose,the root mean square error of prediction(RMSEP)of manually fed-batch culture and Raman-controlled culture was 0.23 and 0.40 g·L^(-1).With the implementation of model correction methods,there was a significant improvement in model performance within Raman-controlled *** RMSEP for glucose from updating-PLS,KF-PLS,and PLS-KF-KF was 0.38,0.36 and 0.17 g·L^(-1),***,the proposed PLS-KF-KF model correction method was found to be more effective and stable,playing a vital role in the automated nutrient feeding of cell cultures.

Controllable and scalable preparation of electrocatalyst materials holds significant importance for their practical *** sputtering is a highly effective synthesis method,known for its producing uniform films and allowing easy control of component *** this paper,we propose an in-situ synthesis method for layered double hydroxide(LDH)electrocatalysts through sacrificing magnetron sputtered *** resulting FeCo-LDH catalyst demonstrated a low overpotential of only 300 mV at 10 mA·cm^(-2).Furthermore,we conducted spectroscopic analysis to investigate the surface changes of the catalysts during the oxygen evolution reaction(OER)*** findings indicated that the formation of Co oxyhydroxides plays a beneficial role in enhancing the catalytical performance of the FeCo-LDH for OER *** restructuring strategy of converting a magnetron-sputtered sacrificial film into a catalytical LDH introduces a new avenue to the synthesis of transition metal-based electrocatalysts.

Preventive effects of Bifidobacterium lactis Probio-M8 on ovalbumin-induced food allergy in mice

Food allergy is a significant public health concern *** probiotics have been found to enhance food allergy by regulating immune-microbe interactions in animal models and ***,the effects of Bifidobacterium lactis Probio-M8 on food allergy have not been thoroughly *** present study examined the anti-allergic properties of Probio-M8,particularly in relation to immune response and gut microbiota *** demonstrate that oral administration of Probio-M8 effectively mitigated the allergy symptoms triggered by ovalbumin(OVA)by ameliorating the morphological damage in the jejunum,reducing OVA-specific IgE and histamine levels in the serum,and suppressing Th2 cytokines(interleukin(IL)4 and IL-13)while increasing Th1 cytokines(interferon(IFN)γ)and regulatory T(Treg)cytokines(IL-10 and transforming growth factor(TGF)β1)in the culture supernatants of splenic ***,Probio-M8 effectively altered the diversity and composition of gut microbiota,particularly the relative abundances of Akkermansia_muciniphila in OVA-induced *** to the OVA group,the Probio-M8 group showed a decrease in the relative abundance of Akkermansia_*** conclusion,Probio-M8 demonstrates the potential to alleviate food allergy by regulating the Th1/Th2 response and modulating gut microbiota,thereby offering a novel therapeutic strategy for patients with food allergy.

Neurotransmitter-initiated signaling pathway were reported to play an important role in regulating the malignant phenotype of tumor *** cells could exhibit a"neural addiction"property and build up local nerve networks to achieve an enhanced neurotransmitter-initiated signaling through nerve growth factor-mediated *** the dysregulated nervous systems might represent a novel strategy for cancer ***,whether intrahepatic cholangiocarcinoma(ICC)could build its own nerve networks and the role of neurotransmitters in the progression ICC remains largely *** staining and Enzyme-linked immunosorbent assay suggested that IcC cells and the infiltrated nerves could generate a tumor microenvironment rich in acetylcholine that promotes IcC metastasis by inducing epithelial-mesenchymal transition(EMT).Acetylcholine promoted iCC metastasis through interacting with its receptor,alpha 5 nicotine acetylcholine receptor subunits(CHRNA5).Furthermore,acetylcholine/CHRNA5 axis activated GSK3β/β-catenin signaling pathway partially through the influx of Ca^(2+)-mediated activation of Ca/calmodulin-dependent protein kinases(CAMKll).In addition,acetylcholine signaling activation also expanded nerve infiltration through increasing the expression of Brain-Derived Neurotrophic Factor(BDNF),which formed a feedforward acetylcholine-BDNF axis to promote ICC ***93,a small-molecule inhibitor of CAMKIll,significantly inhibited the migration and enhanced the sensitivity to gemcitabine of ICC *** all,Acetylcholine/CHRNA5 axis increased the expression ofβ-catenin to promote the metastasis and resistance to gemcitabine of ICC via CAMKIl/GSK3βsignaling,and the CAMKIl inhibitor KN93 may be an effective therapeutic strategy for combating ICC metastasis.

BACKGROUND As the primary microtubule organizing center in animal cells,centrosome abnormalities are involved in human colon *** To explore the role of centrosome-related genes(CRGs)in colon *** CRGs were collected from public *** clustering analysis was performed to separate the Cancer Genome Atlas *** Cox and least absolute shrinkage selection operator regression analyses were performed to identify candidate prognostic CRGs and construct a centrosome-related signature(CRS)to score colon cancer patients.A nomogram was developed to evaluate the CRS risk in colon cancer *** integrated bioinformatics analysis was conducted to explore the correlation between the CRS and tumor immune microenvironment and response to immunotherapy,chemotherapy,and targeted ***-cell transcriptome analysis was conducted to examine the immune cell landscape of core prognostic *** A total of 726 CRGs were collected from public databases.A CRS was constructed,which consisted of the following four genes:TSC1,AXIN2,COPS7A,and *** cancer patients with a high-risk signature had poor *** with a high-risk signature exhibited decreased levels of plasma cells and activated memory CD4+T *** treatment response,patients with a high-risk signature were resistant to immunotherapy,chemotherapy,and targeted ***7A expression was relatively high in endothelial cells and ***1 expression was high in endothelial cells,fibroblasts,and malignant *** We constructed a centrosome-related prognostic signature that can accurately predict the prognosis of colon cancer patients,contributing to the development of individualized treatment for colon cancer.

Due to the important role ofδphase's quantity and morphology in the mechanical and fatigue properties of Inconel 718 alloy and its components,it is necessary to renew insights into the effect of cold deformation on theδphase precipitation,especially on the morphology ***,the nucleation and growth behavior ofδphase in cold-rolled Inconel 718 alloy during aging were *** results show that the precipitation rate and volume fraction ofδphase increase with increasing the cold rolling reduction from 10%to 50%.The volume fraction ofδphase reaches equilibrium after 5 h,remaining at 5.98%,6.52%,and 6.79%under different rolling reductions(10%,30%,and 50%),*** nucleation ofδphase mainly occurs on different sites(grain boundaries,new twin boundaries and old twin boundaries)under 10%rolling reduction,whileδphase mainly nucleates on the new grain boundaries of static recrystallization due to 50%rolling *** the growth ofδphase undergoes a process of alternate orientation growth from spherical(nucleation)→short rod(longitudinal orientation growth)→short rod(radial orientation growth)→dynamic *** 10%rolling reduction,δphase tends to grow into the matrix,while under 50%rolling reduction,the orientation grows faster and is easily affected by the grain boundary curvature.

Maximum Correntropy Criterion-Based UKF for Loosely Coupling INS and UWB in Indoor Localization

Indoor positioning is a key technology in today’s intelligent environments,and it plays a crucial role in many application *** paper proposed an unscented Kalman filter(UKF)based on the maximum correntropy criterion(MCC)instead of the minimummean square error criterion(MMSE).This innovative approach is applied to the loose coupling of the Inertial Navigation System(INS)and Ultra-Wideband(UWB).By introducing the maximum correntropy criterion,the MCCUKF algorithm dynamically adjusts the covariance matrices of the system noise and the measurement noise,thus enhancing its adaptability to diverse environmental localization *** in the presence of non-Gaussian noise,especially heavy-tailed noise,the MCCUKF exhibits superior accuracy and robustness compared to the traditional *** method initially generates an estimate of the predicted state and covariance matrix through the unscented transform(UT)and then recharacterizes the measurement information using a nonlinear regression method at the cost of ***,the state and covariance matrices of the filter are updated by employing the unscented transformation on the measurement ***,to mitigate the influence of non-line-of-sight(NLOS)errors positioning accuracy,this paper proposes a k-medoid clustering algorithm based on bisection k-means(Bikmeans).This algorithm preprocesses the UWB distance measurements to yield a more precise position *** results demonstrate that MCCUKF is robust to the uncertainty of UWB and realizes stable integration of INS and UWB systems.

Numerical Investigation of the Angle of Attack Effect on Cloud Cavitation Flow around a Clark-Y Hydrofoil

Cavitation is a prevalent phenomenon within the domain of ship and ocean engineering,predominantly occurring in the tail flow fields of high-speed rotating propellers and on the surfaces of high-speed underwater *** re-entrant jet and compression wave resulting from the collapse of cavity vapour are pivotal factors contributing to cavity ***,these phenomena significantly modulate the evolution of cavitation *** this paper,numerical investigations into cloud cavitation over a Clark-Y hydrofoil were conducted,utilizing the Large Eddy Simulation(LES)turbulence model and the Volume of Fluid(VOF)method within the OpenFOAM *** analysis of results obtained at different angles of attack is undertaken.A discernible augmentation in cavity thickness is observed concomitant with the escalation in attack angle,alongside a progressive intensification in pressure at the leading edge of the hydrofoil,contributing to the suction *** results can serve as a fundamental point of reference for gaining a deeper comprehension of cloud cavitation dynamics.
