Semaphorin 7a participants in pterygium by regulating vascular endothelial growth factor

AIM: To investigate the relationship between semaphorin 7a expression and cell proliferation and migration in pterygium fibroblasts. METHODS: Twenty-six patients with surgically diagnosed pterygium were enrolled, including 15 cases of primary pterygium and 11 cases of recurrent pterygium. In addition, 12.cases of normal conjunctival tissue were collected. The expression of semaphorin 7a in normal conjunctival tissue, primary pterygium and recurrent pterygium was detected by real-time polymerase chain reaction. Recurrent pterygium fibroblasts were isolated and cultured, and the expression of semaphorin 7a was silenced by small interfering RNA(siRNA) interference technique. Furthermore, the effects of si-semaphorin 7a interference on the mRNA and protein levels of β1-integrin, vascular endothelial growth factor A(VEGFA) and vascular endothelial growth factor receptor(VEGFR), and on fibroblast proliferation were analyzed. Transwell assay was used to detect the effect of semaphorin 7a interference on fibroblast migration. RESULTS: Semaphorin 7a was highly expressed in the primary pterygium and recurrent pterygium samples than that of the normal conjunctival tissue. Compared with the primary pterygium, the expression of semaphoring 7a in the recurrent pterygium samples was significantly increased(P<0.05). The mRNA and protein expression levels of β1-integrin, VEGFA and VEGFR were decreased after si-semaphorin 7a transfection, and as well as the cell proliferation and migration. CONCLUSION: Semaphorin 7a might play important roles in the pathogenesis of pterygium by affecting the expression of β1-integrin, VEGFA and VEGFR.

Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid ***,the therapeutic role of ferroptosis on cancer is gaining increasing *** an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely *** the study,we investigated the effect of baicalin on bladder cancer cells5637 and *** a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer ***,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron *** ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell ***,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced *** of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 *** together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.

目的探讨胆囊癌根治性目的切除术后达到肝脏外科中教科书式结局(TOLS)对远期预后的影响。方法采用回顾性队列研究方法。收集2.14年1月至2.2.年1月中华消化外科菁英荟胆道外科学组全国多中心数据库15家医学中心收治的82.例胆囊癌患者的临床病理资料;男2.5例,女539例;年龄为(62.11)岁。根据TOLS评价标准,将患者分为达到TOLS和未达到TOLS。正态分布的计量资料以x±s表示,组间比较采用独立样本t检验;偏态分布的计量资料以M(Q1,Q3)表示,组间比较采用Mann‑Whitney U检验。计数资料以绝对数表示,组间比较采用χ^(2.检验。等级资料比较采用Mann‑Whitney U检验。采用Kaplan‑Meier法计算生存率和绘制生存曲线,Log‑rank检验进行生存分析。单因素分析和多因素分析采用COX逐步回归模型向后Wald法。结果(1)TOLS情况。82.例行胆囊癌根治性目的切除术患者中,510例达到TOLS,314例未达到TOLS。(2.随访情况。82.例行胆囊癌根治性目的切除术患者,排除出院后90 d内死亡的112.,712.患者纳入生存分析。510例达到TOLS患者的中位随访时间为2.0}(11.4,30.1)个月,中位总生存时间为47.6(30.6,64.6)个月,5年总生存率为47.5%。2.2.未达到TOLS患者的中位随访时间为14.0(6.8,2..5)个月,中位总生存时间为2..3(2..0,2..6)个月,5年总生存率为2..0%。两者总生存情况比较,差异有统计学意义(χ^(2.=58.491,P2.、第八版AJCC T3期和T4期、第八版AJCC N1期、第八版AJCC N2.、辅助治疗是胆囊癌根治性目的切除术后患者总生存时间的独立影响因素(风险比=0.452.1.479,1.373,1.612.1.455,1.481,1.835,1.978,0.538,95%可信区间为0.352.0.581,1.141~1.964,1.052.1.791,1.2.9~2.063,1.102.1.92.,1.02.~2.147,1.380~2.441,1.342.2.915,0.382.0.758,P<0.05)。结论达到TOLS的胆囊癌根治性目的切除术患者能够获得更好的远期预后。

Expression of cyclooxygenase-2.in gastric cancer and its relation to liver metastasis and long-term prognosis

AIM: To investigate the expression of cyclooxygenase-2.COX-2. in gastric cancer and its relation with the liver metastasis and ***: Expression of COX-2.mRNA and protein was examined in gastric cancer and its paired substantial normal tissue by semi-quantitative reverse transcriptionpolymerase chain reaction and *** relation between COX-2.expression and prognosis was investigated in 195 ***: The expression of COX-2.mRNA in gastric cancer tissue was significantly higher than that in normal tissue in 47 cases (w= 792. P＜0.01). The COX-2.mRNA in pT3-4 tissue expressed higher than that in pT1-2.issue (w = 2.4, P＜0.05). The positive expression rate of COX-2.protein was 57.9% (113/195). The COX-2.xpression was significantly related to histological type,lymphnode metastasis, venous invasion and liver metastasis (P＜0.05). No relation was found between COX-2.expression and invasion depth, peritoneal metastasis and International Union against Cancer TNMstage. The multiple regression analysis showed that the COX-2.expression and venous invasion were obviously associated with liver metastasis (P＜0.05). However,there was no significant correlation between COX-2.mmunoreactivity and ***: COX-2.may play an important role in the development of gastric cancer, and the over-expression of COX-2.protein may be a high risk factor for liver metastasis.

目的:观察冠心病稳定型心绞痛牵涉痛分布区域与穴位的关系,并结合动物实验探讨局部敏化出现的规律。方法:临床研究部分,观察1 046例冠心病稳定型心绞痛患者外周压痛敏感部位、局部皮肤形态异常改变,并测量了77位患者疼痛阈值的变化。动物实验部分,以SD大鼠冠状动脉左前降支结扎4h为心肌缺血模型组(n=14),以假手术为对照组(n=4),分别用在自然光下拍摄观察和活体荧光成像的方法,记录大鼠尾静脉注射Evans Blue后体表出现蓝染的部位,并检测痛阈变化。结果:心绞痛患者压痛部位以及皮肤局部组织结构的改变多分布在左侧胸前区、背部、肩部、上肢尺侧部;右侧也有一定数量的分布。压痛点出现在穴位上、穴位附近和穴位以外。心肌缺血的大鼠模型出现类似的变化。结论:心脏发生缺血性病变时,其相应节段(T 1~T 5)神经支配皮节区域发生规律性的牵涉性的"敏化"反应,这些敏感度改变的部位与"穴位"形成有关。

目的:本研究旨在探讨即时和延时的表达性写作对不同情绪价和唤醒度的图片的喜好度的影响。方法:60名被试平均分为4组,分别观看具有积极高唤醒、积极低唤醒、消极高唤醒、消极低唤醒的情绪图片,并评定对图片的喜好度。然后进行即时(实验一)或延时的表达性写作(实验二,写作前被试完成一个30分钟的无关判断任务)。最后对喜好度进行再次评定。结果:对写作文本的分析表明,即时和延时的表达性写作都是有效的;情绪图片的唤醒度对图片的喜好度和文本写作都具有重要的影响;对喜好度的分析表明,在即时性表达中,书写具有显著的影响;但在延时性表达中,没有发现显著的书写效应。结论:即时而非延时的书写表达对改善情绪图片的喜好度具有重要影响。

Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases,including *** ferroptosis in cancer cells may be a potential strategy for cancer ***,ferroptosis-inducing drugs are attracting more attention for cancer ***,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl,exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer ***,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells,which was accompanied by ROS accumulation,lipid peroxidation,and GSH *** ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death,indicating that ferroptosis contributed to erianin-induced cell ***,we demonstrated that Ca^(2.)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing *** together,our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2.)/CaMdependent ferroptosis and inhibiting cell migration,and erianin will hopefully serve as a prospective compound for lung cancer treatment.

Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological *** a broad spectrum of aging biomarkers has been developed,their potential uses and limitations remain poorly *** immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower?This review aims to address this ***,we summarize our current knowledge of biomarkers developed for cellular,organ,and organismal levels of aging,comprising six pillars:physiological characteristics,medical imaging,histological features,cellular alterations,molecular changes,and secretory *** fulfill all these requisites,we propose that aging biomarkers should qualify for being specific,systemic,and clinically relevant.
