产前日粮碳水化合物来源和莫能菌素对经产奶牛围产期代谢和泌乳的影响

选用85头经产荷斯坦奶牛随机设计,评估产前碳水化合物来源及莫能菌素对围产期奶牛干物质采食量、血液参数和泌乳性能的影响。日粮处理按照2×2阶乘排列设计,包括传统的干奶料(CONV)和不含有草料纤维来源的干奶料(NFFS),补充或不补充330 mg/d.头莫能菌素。CONV日粮包括70%的草料,NFFS日粮28%的草料用棉籽壳和大豆壳替代。在整个干奶期(产前60 d)饲喂试验日粮(CONV和NFFS)。在预期产犊日期之前的28 d开始每天追加一次莫能菌素直到分娩。分娩后,所有奶牛饲喂相同的泌乳期奶牛料。结果发现,在妊娠的最后28 d,与饲喂CONV日粮相比,饲喂NFFS产前日粮干物质采食量(15.8 vs 11.9 kg/d)、干物质采食量体重百分比(2.1%vs1.6%体重)、血糖(67.4 vs 64.6 mg/dl)和血清胰岛素浓度(0.59 vs 0.45 ng/ml)更高,非酯化脂肪酸浓度(185 vs 245μEq/L)较低。在泌乳的前56 d,产前Ch.、莫能菌素或两者混合效应对平均产奶量或乳成分没有明显的影响。但随着时间的延长,产前碳水化合物来源有影响产奶量的趋势。产前28 d补给莫能菌素对围产期产奶量没有影响。产前日粮对产后干物质采食量、血糖、非酯化脂肪酸、胰岛素浓度或肝脏甘油三酯没有影响。研究结果表明,在干奶期日粮中用NFFS部分替换传统日粮碳水化合物提高或维持了产前干物质采食量,因此增强了分娩前代谢水平。产前更高的干物质采食量可能潜在增加泌乳早期的产奶量。

Early.appearance of th. serotonergic sy.tem in th. fetal brain and th. various effects of serotonin (5-h.) on brain morph.genesis, h.ve given support to a neurotroph.c role of serotonin. Th.s function of serotonin is accomplish.d th.ough.a sy.tem of serotonin nerve terminals in th. target regions th.t involves various 5-h. receptors. In visual, auditory.and somatosensory.cortex an early.and intense serotonergic innervation is particularly.important. Th. neuronal somata of th.se terminals are normally.located in th. mesenceph.lon and th.y.h.ve not been observed in th. maturing cerebral cortex, neith.r in th. adult brain. By.using immunolabeling tech.iques, fluorescence and confocal microscopy. we observe th. presence of both. 5-h. terminals and 5-h. cells in mesenceph.lon (Me, E17) and in th. neopallium (Np, E13-E16) cocultures. Cells immunopositive to 5-h. and to try.toph.n-5-h.0}roxilase are also observed in th. Np on day.12 of culture. Th.se results concerning th. unexpected presence of serotonergic cells in th. fetal cerebral cortex are interesting and may.be of importance in corticogenesis. As it h.ppens with.oth.r elements of th. serotonergic sy.tem, th. presence of th.se ph.noty.ically.serotonergic cells in th. early.cerebral cortex may.be transitory.and probably.supporting cortex maturation processes. Th. molecular signaling path.of th. 5-h.1A receptor h.s also been identified.

To increase protein stability.and test protein function, th.ee double-cy.teine mutations were individually.introduced by.protein engineering into th. cy.teine-free Cry.Aa δ-endotoxin from Bacillus th.ringiensis. Th.se mutations were designed to create disulfide bonds between α-h.lices 2 and 5 (positions 110 - 193), and α-h.lices 5 and 7 (positions 195 - 276 and 198 - 276). Comparison of th. CD spectra of th. wild-ty.e and th. double-cy.teine mutant proteins indicates a tigh.er h.lical packing consistent with.formation of at least two of th. disulfide bonds between th. central and th. outer h.lices. Th.rmal stability.analy.is indi-cates th.t potential covalent linkages between th. central α-h.lix 5 and th. oth.r h.lices increase resistance to th.rmal denaturation by.10?C to 14?C com-pared to th. th.rmal stability.of th. wild-ty.e protein. Spectroscopic analy.is of th. disulfide-specific absorbance band indicates th.t th. double mutant proteins are more stable to temperature and denaturant (guanidine h.0}roch.oride) th.n th. wild-ty.e protein, as a result of th. formation of two of th. disulfide bridges. Th.se results indicate th.t th. double muta-tions M110C/F193C and A198C/V276C successfully.establish.d disulfide bonds, resulting in a more stable structure of th. entire toxin. Despite th. increase in stability.and structural ch.nges introduced by.th. disulfide bonds, no effect on toxicity.was observed. A possible mech.nism involving th. insertion of all of domain I of Cry.Aa toxin into th. target membrane accounts for th.se observations.

我们对同一Ｂｉ２２１２晶须样品加不同方向的磁场，比较了Ｈ∥ｂ和Ｈ∥ｃ两种情况下的ＩＶ曲线．我们发现，当Ｈ∥ｃ时，驱动磁通线运动的临界电流Ｉｃ与温度的关系遵循一般的单调地随温度上升而下降的关系．但是，当Ｈ∥ｂ时，Ｉｃ在Ｔ＊＝２５．５Ｋ附近出现了一个峰值：当Ｔ＞Ｔ＊时，Ｉｃ随温度的下降而升高；Ｔ＜Ｔ＊时，Ｉｃ随温度的下降而下降．我们在文章中对此现象进行了讨论．
