Puerariae Radix protects against ulcerative colitis in mice by inhibiting NLRP3 inflammasome activation

Ulcerative colitis(UC)is a common inflammatory disease of the gastrointestinal *** Chinese medicine(TCM)has long been used in Asia as a treatment for UC and Puerariae Radix(PR)is a reliable anti-diarrheal *** aims of this study were to investigate the protective effect of PR using the dextran sulfate sodium salt(DSS)-induced UC model in mice and identify molecular mechanisms of PR *** chemical constituents of PR via ultra-performance liquid chromatography/tandem mass spectrometry and identified potential PR and UC targets using a network pharmacology(NP)approach were obtained to guide mouse experiments.A total of 180 peaks were identified from PR including 48 flavonoids,46 organic acids,14 amino acids,8 phenols,8 carbohydrates,7 alkaloids,6 coumarins and 43 other *** results showed that caspase-1 was the most dysregulated of the core genes associated with UC.A PR dose of 0.136 mg/g administered to DSS treated mice reversed weight loss and decreased colon lengths found in UC *** also alleviated intestinal mucosal shedding,inflammatory cell infiltration and mucin *** treatment suppressed upregulation of NOD-like receptor protein 3(NLRP3),cysteinyl aspartate-specific proteases-1(caspase-1),apoptosis-associated speck-like(ASC)and gasdermin D(GSDMD)at both the protein and m RNA expression *** addition of a small molecule dual-specificity phosphatase inhibitor NSC 95397 inhibited the positive effects of *** results indicated that PR exerts a protective effect on DSS-induced colitis by inhibiting NLRP3 inflammasome activation in mice.

Epilepsy,one of the most common neurological disorders,is characterized by spon-taneous recurrent *** lobe epilepsy(TLE)is one of the most common medically intractable seizure ***2-and NcK-interacting kinase(TNIK)has recently attracted attention as a critical modulation target of many neurological and psychiatric disorders,but its role in epilepsy remains *** this study,we hypothesized the involvement of TNIK in epilepsy and investigated TNIK expression in patients with intractable TLE and in a pilocarpineinduced rat model of epilepsy by western blotting,immunofluorescence,and immunohistochemistry.A pentylenetetrazole(PTZ)-induced epilepsy rat model was used to determine the effect of the TNIK inhibitor NCB-0846 on behavioral manifestations of ***(Co-IP)/mass spectrometry(MS)was used to identify the potential *** Co-IP,we detected and confirmed the main potential TNIK *** fractionation was used to establish the effect of NCB-0846 on the expression of the main interactors in postsynaptic density(PSD)*** found that TNIK was primarily located in neurons and decreased significantly in epilepsy model rats and TLE patients compared with ***-0846 delayed kindling progression and decreased seizure ***-IP/MS identified 63 candidate TNIK interactors in rat hippocampi,notably ***-IP showed that TNIK might correlate with endogenous GRIA1,SyN2,PSD-95,CaMKIV,GABRG1,and *** addition,the significant decrease in GRIA1 in hippocampal total lysate and PSDs after NCB-0846 treatment might help modify the progression of PTZ *** results suggest that TNIK contributes to epileptic pathology and is a potential antiepileptic drug target.

A novel distributed solar refrigeration system based on evacuated U-tube solar collector and elastocaloric refrigerator

Refrigeration challenges in regions with electricity shortages significantly decrease the quality of life for residents. In response to the prevalent refrigeration challenges in power-deficient areas, a novel distributed solar refrigeration system, comprising an evacuated U-tube solar collector and elastocaloric refrigerator, is theoretically introduced. Theoretical formulations for the energy efficiency and cooling power of the solar refrigeration system are presented to facilitate predictive assessments of the performance properties. Under typical conditions, the energy efficiency and cooling power of the solar refrigeration system are,respectively, 4.84% and 200.15 W. Subsequently, an extensive parameter study is conducted to comprehensively uncover key performance influencers and identify avenues for improvement. In addition, local sensitivity analyses identify that the length ratio is the top influential parameter, while the heat transfer fluid flow rate is the least sensitivity. A pragmatic case study,conducted with the weather data of Ningbo City, China, serves to empirically predict the performance of the hybrid system within the constraints of practical circumstances.

Enhancing SAEAs with unevaluated solutions:a case study of relation model for expensive optimization

Surrogate-assisted evolutionary algorithms(SAEAs) hold significant importance in resolving expensive optimization problems. Extensive efforts have been devoted to improving the efficiency of SAEAs through the development of proficient model-assisted solution selection methods, which, however, first requires generating high-quality solutions. The fundamental paradigm of evaluating a limited number of solutions in each generation within SAEAs reduces the variance of adjacent populations, thereby affecting the quality of offspring solutions. This is a frequently encountered issue yet receives little attention. To address the issue, this paper presents a framework using unevaluated solutions to enhance the efficiency of *** surrogate model is employed to identify high-quality solutions for the direct generation of new solutions without evaluation. To ensure dependable selection, we have introduced two tailored relation models to select the optimal solution and the unevaluated population. A comprehensive experimental analysis is performed on two test suites, which showcases the superiority of the relation model over regression and classification models in the solution selection phase. Furthermore, the surrogate-selected unevaluated solutions with high potential have been shown to enhance the efficiency of the algorithm significantly.

Data-driven discovery of partial differential equations(PDEs)has recently made tremendous progress,and many canonical PDEs have been discovered successfully for proof of ***,determining the most proper PDE without prior references remains challenging in terms of practical *** this work,a physics-informed information criterion(PIC)is proposed to measure the parsimony and precision of the discovered PDE *** proposed PIC achieves satisfactory robustness to highly noisy and sparse data on 7 canonical PDEs from different physical scenes,which confirms its ability to handle difficult *** PIC is also employed to discover unrevealed macroscale governing equations from microscopic simulation data in an actual physical *** results show that the discovered macroscale PDE is precise and parsimonious and satisfies underlying symmetries,which facilitates understanding and simulation of the physical *** proposition of the PIC enables practical applications of PDE discovery in discovering unrevealed governing equations in broader physical scenes.

Accurate Distinction of Weak and Strong AC Grids for Emerging Hierarchical-Infeed LCC-UHVDC Systems

Distinction of weak and strong AC grids for emerging hierarchical-infeed LCC-UHVDC systems is important for planning and operation departments. However, accuracy of earlier distinction methods is limited as they were developed by empirical reasoning without rigorous theoretical analysis. Hence in this letter, hierarchical-infeed interactive effective short-circuit ratio (HIESCR) index is first used for strength evaluation of HIDC systems with complex inter-inverter interactions considered. Boundary HIESCR (BHIESCR) is also introduced in the proposed distinction method of weak and strong AC grids. That is, weak (or strong) AC grids are, respectively, identified when HIESCR is less (or greater) than BHIESCR. Second, it is shown BHIESCR remains almost unchanged as 3.0 versus various system parameters and rated operation variables based on rigorous theoretical analysis. This salient feature makes the proposed method more accurate than earlier methods. Finally, the proposed method is validated by simulations based on the PSCAD/EMTDC program.

Highly active copper-intercalated weakly crystallized δ-MnO_(2) for low-temperature oxidation of CO in dry and humid air

Copper intercalated birnessite MnO_(2)(δ-MnO_(2))with weak crystallinity and high specific surface area(421 m^(2)/g)was synthesized by a one-pot redox method and investigated for low-temperature CO *** molar ratio of Cu/Mn was as high as 0.37,which greatly weakened the Mn-O bond and created a lot of low-temperature active oxygen *** situ DRIFTS revealed strong bonding of copper ions with ***-synthesized MnO_(2)-150Cu achieved 100%conversion of 250 ppm CO in normal air(3.1 ppm H_(2)O)even at−10°C under the weight-hourly space velocity(WHSV)of 150 L/(g·h).In addition,it showed high oxygen storage capacity to oxidize CO in inert *** the concurrent moisture in air significantly inhibited CO adsorption and its conversion at ambient temperature,MnO_(2)-150Cu could stably convert CO in 1.3%moisture air at 70°C owing to its great low-temperature activity and reduced competitive adsorption of water with increased *** study discovers the excellent low-temperature activity of weakly crystallized δ-MnO_(2) induced by high content intercalated copper ions.

Performance Improvement of Two-Phase Steam Ejector based on CFD Techniques,Response Surface Methodology and Genetic Algorithm

The steam ejector is a crucial component in the waste heat recovery *** performance determines the amount of recovered heat and system ***,poor ejector performance has always been the main bottleneck for system ***,this study proposes an optimization methodology to improve the steam ejector's performance by utilizing computational fluid dynamics(CFD) techniques,response surface methodology(RSM),and genetic algorithm(GA).Firstly,a homogeneous equilibrium model(HEM) was established to simulate the two-phase flow in the steam ***,the orthogonal test was presented to the screening of the key decision variables that have a significant impact on the entrainment ratio(ER).Next,the RSM was used to fit a response surface regression model(RSRM).Meanwhile,the effect of the interaction of geometric parameters on the performance of the steam ejector was ***,GA was employed to solve the RSRM's global optimal ER *** results show that the RSRM exhibits a good fit for ER(R^(2)=0.997).After RSM and GA optimization,the maximum ejector efficiency is 27.94%,which is 48.38% higher than the initial ejector of 18.83%.Furthermore,the optimized ejector efficiency is increased by 46.4% on average under off-design ***,the results reveal that the combination of CFD,RSM,and GA presents excellent reliability and feasibility in the optimization design of a two-phase steam ejector.

A LiDAR Point Clouds Dataset of Ships in a Maritime Environment

For the first time, this article introduces a LiDAR Point Clouds Dataset of Ships composed of both collected and simulated data to address the scarcity of LiDAR data in maritime applications. The collected data are acquired using specialized maritime LiDAR sensors in both inland waterways and wide-open ocean environments. The simulated data is generated by placing a ship in the LiDAR coordinate system and scanning it with a redeveloped Blensor that emulates the operation of a LiDAR sensor equipped with various laser beams. Furthermore,we also render point clouds for foggy and rainy weather conditions. To describe a realistic shipping environment, a dynamic tail wave is modeled by iterating the wave elevation of each point in a time series. Finally, networks serving small objects are migrated to ship applications by feeding our dataset. The positive effect of simulated data is described in object detection experiments, and the negative impact of tail waves as noise is verified in single-object tracking experiments.
