Functional analysis of the novel mitochondrial tRNA^(Trp)and tRNA^(Ser(AGy))variants associated with type 2 diabetes mellitus

BACKGROUND Mutations in mitochondrial tRNA(mt-tRNA)genes that result in mitochondrial dysfunction play important roles in type 2 diabetes mellitus(T2DM).We previously reported a large Chinese pedigree with maternally inherited T2DM that harbors novel mt-tRNA^(Trp)A5514G and tRNA^(Ser(AGy))C12237T variants,however,the effects of these mt-tRNA variants on T2DM progression are largely *** To assess the potential pathogenicity of T2DM-associated m.A5514G and m.C12237T variants at genetic,molecular,and biochemical *** Cytoplasmic hybrid(cybrid)cells carrying both m.A5514G and m.C12237T variants,and healthy control cells without these mitochondrial DNA(mtDNA)variants were generated using trans-mitochondrial *** features,including mt-tRNA steady-state level,levels of adenosine triphosphate(ATP),mitochondrial membrane potential(MMP),reactive oxygen species(ROS),mtDNA copy number,nicotinamide adenine dinucleotide(NAD+)/NADH ratio,enzymatic activities of respiratory chain complexes(RCCs),8-hydroxy-deoxyguanine(8-OhdG),malondialdehyde(MDA),and superoxide dismutase(SOD)were examined in cell lines with and without these mt-tRNA *** Compared with control cells,the m.A5514G variant caused an approximately 35%reduction in the steady-state level of mt-tRNA^(Trp)(Py))steady-state level(P=0.5849).Biochemical analysis revealed that cells with both m.A5514G and m.C12237T variants exhibited more severe mitochondrial dysfunctions and elevated oxidative stress than control cells:ATP,MMP,NAD+/NADH ratio,enzyme activities of RCCs and SOD levels were markedly decreased in mutant cells(Py contrast,the levels of ROS,8-OhdG and MDA were significantly increased(Py number was not affected by m.A5514G and m.C12237T variants(P=0.5942).CONCLUSION The m.A5514G variant impaired mt-tRNA^(Trp)metabolism,which subsequent

Cracking on a nickel-based superalloy fabricated by direct energy deposition

Cracks have consistently been a significant challenge limiting the development of additive manufactured nickel-based *** is essential to investigate the location of cracks and their forming *** study extensively examines the impact of solidification process,microstructural evolution,and stress concentration on crack initiation during direct energy deposition(DED).The results emphasize that the crack formation is significantly related to large-angle grain boundaries,rapid cooling *** caused by large-angle grain boundaries and a fast-cooling rate predominantly appear near the edge of the deposited *** cracks are more likely to form near the top of the deposited sample,due to the presence ofγ/γ'*** secondary dendritic arm and the carbides in the interdendritic regions can obstruct liquid flow during the final stage of solidification,which results in the formation of solidification cracks and *** work paves the way to avoid cracks in nickel-based superalloys fabricated by DED,thereby enhancing the performance of superalloys.

Deciphering TADF Mechanisms in Metal-Free Organic Emitters BrA-HBI:Utilizing Optimally Tuned Range-Separated Functionals for Insight

Metal-free organic emitters,characterized by their thermally activated delayed fluorescence(TADF)properties,offer considerable promise for the creation of highly efficient organic light-emitting diodes(OLEDs).Recently,Shao et *** a novel excited state intramolecular proton transfer(ESIPT)system BrA-HBI,demonstrating an emission quantum yield of up to 50%[***.32,2201256(2022)].However,many open issues cannot be answered solely by experimental means only and require detailed theoretical *** instance,what causes the activation of TADF from the Keto*tautomer and leads to fluorescence quenching in the Enol^(*)form?Herein,we provide a theoretical investigation on the TADF mechanism of the BrA-HBI molecule by optimally tuned range-separated *** findings reveal that ESIPT occurs in the BrA-HBI ***,we have disclosed the reason for the fluorescence quenching of the Enol^(*)form and determined that the T_(2)state plays a dominant role in the TADF *** addition,double hybrid density functionals method was utilized to verify the reliability of optimally tuned range separation functionals on the calculation of the TADF mechanism in *** findings not only provide a theoretical reference for development of highly efficient organic light-emitting diodes,but also demonstrate the effectiveness of the optimally tuned range-separated functionals in predicting the luminescence properties of TADF molecules.

Effect of solidification mode on microstructure evolution and properties of magnesium alloy with long-period stacking ordered phase

How Emotion Nurtures Mentality:The Influencing Mechanism of Social-Emotional Competency on the Mental Health of University Students

Social-Emotional Competency(SEC),regarded as a critical psychological resource for individuals to adapt to social environments,is an effective protective factor for students’mental health,impacting their future success and *** the impact of SEC on university students’mental health can offer valuable insights for nurturing talents with healthy psychological and physical *** on data from two large-scale surveys of Chinese university students,this study designed two comprehensive Multiple Mediation Models involving SEC,stress,coping strategies,and stress reaction to explore the pathway of emotion nurturing *** 1 utilized a parallel mediation model to examine the relationships between SEC,academic stress,interpersonal stress,and stress *** results indicated that SEC negatively predicted academic stress,interpersonal stress,and stress ***,academic and interpersonal stress mediated the relationships between SEC and stress reactions in *** these findings,Study 2 further investigated the role of coping *** constructing a multiple-chain mediation model,it examined the predictive relationships among SEC,academic stress,interpersonal stress,three types of coping strategies,and stress *** findings indicated that SEC negatively predicted stress,problemavoiding strategy,and stress reactions,while positively predicting problem-solving and assistant-seeking ***,the two stress types and three coping strategies significantly mediated the relationship between SEC and stress *** indicated that higher SEC was associated with reduced stress and more adaptive coping strategies and subsequently contributed to more favorable stress *** research explored the impact of university students’SEC on mental health and its relational mechanisms,aiming to provide theoretical reference and practical insights for future efforts in cultivating SEC among uni

The Shanghai Laser Electron Gamma Source(SLEGS, located in BL03SSID) beamline at the Shanghai Synchrotron Radiation Facility(SSRF) is a Laser Compton Scattering(LCS) gamma source used for the investigation of nuclear structure, which is in extensive demand in fields such as nuclear astrophysics, nuclear cluster structure, polarization physics, and nuclear energy. The beamline is based on the inverse Compton scattering of 10640 nm photons on 3.5 GeV electrons and a gamma source with variable energy by changing the scattering angle from 20° to 160°. γ rays of 0.25-21.1 MeV can be extracted by the scheme consisting of the interaction chamber, coarse collimator, fine collimator, and attenuator. The maximum photon flux for 180° is approximately 10~7 photons/s at the target at 21.7 MeV, with a 3-mm-diameter beam. The beamline was equipped with four types of spectrometers for experiments in( γ,γ'),( γ,n),( γ,p), and( γ,α). At present, Nuclear Resonance Fluorescence(NRF) spectrometry, Flat-Efficiency neutron Detector(FED) spectrometry, neutron Time-Of-Flight(TOF) spectrometry, and Light-Charged Particle(LCP) spectrometry methods have been developed.

Thickening progression mechanism of silica fume-oil well cement composite system at high temperatures

This work studied the thickening progression mechanism of the silica fume-oil well cement composite system at high temperatures(110-180.C) in order to provide a theoretical guidance for the rational application of silica fume in the cementing engineering. Results showed that silica fume seldom affected the thickening progression of oil well cement slurry at 110-120.C, but when temperature reached above130.C, it would aggravate the bulging degree of thickening curves and significantly extend the thickening time, meanwhile causing the abnormal “temperature-based thickening time reversal” and“dosage-based thickening time reversal” phenomena in the range of 130-160.C and 170-180.C respectively. At 130-160.C, the thickening time of oil well cement slurry was mainly associated with the generation rate of calcium hydroxide(CH) crystal. The introduced silica fume would be attracted to the cement minerals' surface that were hydrating to produce CH and agglomerate together to form an“adsorptive barrier” to hinder further hydration of the inner cement minerals. This “adsorptive barrier”effect strengthened with the rising temperature which extended the thickening time and caused the occurrence of the “temperature-based thickening time reversal” phenomenon. At 170-180.C, the pozzolanic activity of silica fume significantly enhanced and considerable amount of C-S-H was generated, thus the “temperature-based thickening time reversal” vanished and the “dosage-based thickening time reversal” was presented.

Rational design of superior catalysts from topological semimetals with nontrivial energy window

Materials featuring topological energy bands and nontrivial surface states hold significant promise in unlocking unprecedented opportunities for innovating electrocatalytic ***,it remains a challenge to realize superior topological catalysts which can carry both high catalytic activity and excellent catalytic ***,we propose that a family of Ni-based binary materials hosting fantasying topological conjunct-nodalpoint state and a large nontrivial energy window(NEWD)represents an ideal choice for such superior topological catalysts in hydrogen evolution *** presence of conjunct-nodal-points ensures long Fermi arcs on the surface,thereby enabling an extremely high catalytic *** NEWD plays a crucial role in stabilizing the high catalytic activity against external perturbations,such as strain and electron/hole *** roles for conjunctnodal-points and NEWD are substantiated by the observable weakening of catalytic performance during topological phase transitions,which result in the removal of the conjunct-nodal-points,NEWD and their corresponding long Fermi *** work unveils a hidden mechanism and opens a feasible route for developing superior quantum catalysts from novel topology point of view.

Polymethoxyflavones(PMFs)are a type of uncommon dietary flavonoids,characterized by more than one methoxy group,which exist in limited plant species,like Citrus species and Kaempferia *** addition,different PMFs,such as nobiletin,sinensetin,tangeretin,and casticin,have been isolated from these natural *** have received increasing attention due to their multiple bioactivities,such as antioxidant,anti-inflammatory,anti-cancer,metabolic regulatory,immunoregulatory,neuroprotective,and skin protective *** bioactivities of PMFs should be associated with the regulation of critical molecular targets and the interaction with gut *** order to provide a comprehensive and updated review of PMFs,their natural sources,refined extraction,biosynthesis,metabolism,and bioactivities are summarised and discussed,with the emphasis on the molecular mechanisms of PMFs on regulating different chronic ***,PMFs may be promising flavonoids to the forefront of nutraceuticals for the prevention and/or treatment of certain human chronic diseases.
