Surgical experience in splitting donor liver into left lateral and right extended lobes

AIM: To outline the surgical experience with donor liver splitting in split liver transplantation.METHODS: From March 1 to September 1 in 2004, 10donor livers were split ex situ into a left lateral lobe (segments Ⅱ and Ⅲ) and a right extended lobe (segments Ⅰ, Ⅳ-Ⅷ) in Medical School of Hannover, and thereafter split liver transplantation was performed successfully in 19 cases. The average age, weight and ICU staying period of the donors were 32.7 years (15-51 years), 64.5 kg (45-75 kg) and 2.4 d (1-8 d) respectively.RESULTS: The average weight of the whole graft and the left lateral lobe was 1 322.6 g (956-1 665 g) and 281.8 g (198-373 g) respectively, and the average ratio of left lateral lobe to the whole graft was 0.215 (0.178-0.274).The average graft to recipient weight ratio (GRWR) of the left lateral lobe and the right extended lobe reached 2.44% (1.22-5.41%) and 1.73% (1.31-2.30%) respectively.On average it took approximately 105 min (85-135 min)to split the donor liver. Five donor organs showed anatomic variation including the left hepatic vein variation in two cases, the left hepatic artery variation in two cases and the bile duct variation in one case.CONCLUSION: Split liver transplantation has become a mature surgical technique to expand the donor pool with promising results. In the process of graft splitting, close attention needs to be paid to potential anatomic variations,especially to variations of the left hepatic vein, the left hepatic artery, and the bile duct.

Structural biology revolution led by technical breakthroughs in cryo-electron microscopy

Recent technical breakthroughs in cryo-electron microscopy（cryo-EM） revolutionized structural biology, which led to the 2017 Nobel Prize in chemistry being awarded to three scientists, Jacques Dubochet, Joachim frank, and Richard Henderson, who made groundbreaking contributions to the development of cryo-EM. In this review, I will give a comprehensive review of the developmental history of cryo-EM, the technical aspects of the breakthrough in cryo-EM leading to the structural biology revolution, including electron microscopy, image recording devices and image processing algorithms,and the major scientific achievements by Chinese researchers employing cryo-EM, covering protein complexes involved in or related to gene expression and regulation, protein synthesis and degradation, membrane proteins, immunity, and viruses.Finally, I will give a perspective outlook on the development of cryo-EM in the future.

为适应日益增长的对生物细胞形态精密测定的技术需求,应用VirtualLab虚拟仿真实验系统,根据红细胞(RBC)的形体特征和光学特性,设计了双曲面旋转对称体的红细胞光散射模型。最佳组合选用了仿真系统中的运算功能模块,仿真了球形模型和双曲面旋转对称模型下,红细胞在几种典型光源入射情况下的光散射,并获得了这几种典型情况下的前向光散射强度图样、幅值分布曲线和相位分布图。其中生物细胞光散射相位分布局域图表明了相位分布与细胞的形态与结构以及其在光路中的位置密切相关。生物细胞前向光散射相位分布图谱可望成为一种新的测定细胞类别的判读依据。

针对中俄东线天然气管道的实际服役状况,分析了土壤运动(冻胀与解冻沉降)对管道结构完整性的影响以及基于管体结构-土壤弹簧模型在确定管-土交互作用方面的局限性,即非线性、大应变与多轴加载评估的保守性、土壤本构模拟与真实状况的偏离,建议发展新型多模块耦合集成技术确定土壤运动产生的机械效应。明确了X80高强管线钢在服役条件下发生应变时效及其导致管线钢(尤其是焊缝区)材料韧性和止裂能力的降低,建议使用时效活化能与等效时效时间模拟、评估管线钢在漫长服役过程中发生应变时效的敏感性,并建立相应的理论基础。此外,详细分析3种常见的管道缺陷(机械损伤、腐蚀缺陷、裂纹)对管道完整性影响的评估技术现状。针对高压、大口径、高强钢天然气管道(特别是焊接金属与热影响区)在地质不稳定地区的材料韧性、裂纹扩展以及止裂能力开展实验与评价技术,建立精确的多物理场协同作用下的管道缺陷评估模型,是当前的国际性技术难题,这些问题的解决将有力保障中俄东线天然气管道以及相关油气管道的长期安全运行。

Fungi are an understudied resource possessing huge potential for developing products that can greatly improve human well-being.In the current paper,we highlight some important discoveries and developments in applied mycology and interdisciplinary Life Science research.These examples concern recently introduced drugs for the treatment of infections and neurological diseases;application of–OMICS techniques and genetic tools in medical mycology and the regulation of mycotoxin production;as well as some highlights of mushroom cultivaton in Asia.Examples for new diagnostic tools in medical mycology and the exploitation of new candidates for therapeutic drugs,are also given.In addition,two entries illustrating the latest developments in the use of fungi for biodegradation and fungal biomaterial production are provided.Some other areas where there have been and/or will be significant developments are also included.It is our hope that this paper will help realise the importance of fungi as a potential industrial resource and see the next two decades bring forward many new fungal and fungus-derived products.

Repeated Physical Training and Environmental Enrichment Induce Neurogenesis and Synaptogenesis Following Neuronal Injury in an Inducible Mouse Model

Neuronal loss as a consequence of brain injury, stroke and neurodegenerative disorders causes functional impairments ranging from cognitive impairments to physical disabilities. Extensive rehabilitation and trainning may lead to neuroprotection and promote functional recovery, although little is known about the molecular and cellular mechanisms driving this event. To investigate the underlying mechanisms and levels of functional recovery elicited by repeated physical training or environmental enrichment, we generated an inducible mouse model of selective CA1 hippocampal neuronal loss. Following the CA1 neuronal injury, mice underwent one of the above mentioned conditions for 3 months. Exposure to either of these stimuli promoted functional cognitive recovery, which was associated with increased neurogenesis in the subgranular zone of dentate gyrus and enhanced synaptogenesis in the CA1 subfield. Notably, a significant correlation was found between the functional recovery and increased synaptogenesis among survived CA1 neurons. Collectively, these results support the utilization of cognitive and physical stimulation as approaches to promote recovery after neuronal loss and demonstrate the potential of this novel mouse model for the development of therapeutic strategies for various neurological disorders associated with focal neuronal loss.

Background.Increasing evidence from human studies has revealed the adverse impact of ambient fine particles(PM_(2.5))on health outcomes related to metabolic disorders and distant organs.Whether exposure to ambient PM_(2.5)leads to kidney impairment remains unclear.The rapid air quality improvement driven by the clean air actions in China since 2013 provides an opportunity for a quasiexperiment to investigate the beneficial effect of PM_(2.5)reduction on kidney function.Methods.Based on two repeated nationwide surveys of the same population of 5115 adults in 2011 and 2015,we conducted a difference-indifference study.Variations in long-term exposure to ambient PM_(2.5)were associated with changes in kidney function biomarkers,including estimated glomerular filtration rate by serum creatinine(GFRscr)or cystatin C(GFRcys),blood urea nitrogen(BUN),and uric acid(UA).Results.For a 10μg/m^(3)reduction in PM_(2.5),a significant improvement was observed for multiple kidney functional biomarkers,including GFRscr,BUN and UA,with a change of 0.42(95%confidence interval[CI]:0.06,0.78)mL/min/1.73m^(2),-0.38(-0.64,-0.12)mg/dL,and-0.06(-0.12,-0.00)mg/dL,respectively.A lower socioeconomic status,indicated by rural residence or low educational level,enhanced the adverse effect of PM_(2.5)on kidney function.Conclusions.These results support a significant nephrotoxicity of PM_(2.5)based on multiple serum biomarkers and indicate a beneficial effect of improved air quality on kidney function.

Effects of virtual environment and self-representations on perception and physical performance in redirected jumping

Background Redirected jumping(RDJ)allows users to explore virtual environments(VEs)naturally by scaling a small real-world jump to a larger virtual jump with virtual camera motion manipulation,thereby addressing the problem of limited physical space in VR applications.Previous RDJ studies have mainly focused on detection threshold estimation.However,the effect VE or self representation(SR)has on the perception or performance of RDJs remains unclear.Methods In this paper,we report experiments to measure the perception(detection thresholds for gains,presence,embodiment,intrinsic motivation,and cybersickness)and physical performance(heart rate intensity,preparation time,and actual jumping distance)of redirected forward jumping under six different combinations of VE(low and high visual richness)and SRs(invisible,shoes,and human-like).Results Our results indicated that the detection threshold ranges for horizontal translation gains were significantly smaller in the VE with high rather than low visual richness.When different SRs were applied,our results did not suggest significant differences in detection thresholds,but it did report longer actual jumping distances in the invisible body case compared with the other two SRs.In the high visual richness VE,the preparation time for jumping with a human-like avatar was significantly longer than that with other SRs.Finally,some correlations were found between perception and physical performance measures.Conclusions All these findings suggest that both VE and SRs influence users'perception and performance in RDJ and must be considered when designing locomotion techniques.
