Objective:To evaluate the efficacy.and safety.of Huashi Baidu Granules(hsBD)in treating patients with severe acute respiratory.sy.drome coronavirus 2(SARS-CoV-2)Omicron ***:A single-center retrospective cohort study.was conducted during COVID-19 Omicron epidemic in the Mobile Cabin Hospital of Shanghai New International Expo Center from April 1st to May.23rd,*** COVID-19 patients with asy.ptomatic or mild infection were assigned to the treatment group(hsBD users)and the control group(non-hsBD users).After propensity.score matching in a 1:1 ratio,496 hsBD users of treatment group were matched by.propensity.score to 496 non-hsBD *** in the treatment group were administrated hsBD(5 g/bag)orally.for 1 bag twice a day.for 7 consecutive *** in the control group received standard care and routine *** primary.outcomes were the negative conversion time of nucleic acid and negative conversion rate at day.*** outcomes included the hospitalized day.,the time of the first nucleic acid negative conversion,and new-onset sy.ptoms in asy.ptomatic *** events(AEs)that occurred during the study.were *** subgroup analy.is was conducted in vaccinated(378 hsBD users and 390 non-hsBD users)and unvaccinated patients(118 hsBD users and 106 non-hsBD users).Results:The median negative conversion time of nucleic acid in the treatment group was significantly.shortened than the control group[3 day.(IQR:2-5 day.)vs.5 day.(IQR:4-6 day.);Py.higher than those in the control group at day.7(91.73%vs.86.90%,P=0.014).Compared with the control group,the hospitalized day. in the treatment group were significantly.reduced[10 day.(IQR:8-11 day.)vs.11 day.(IQR:10.25-12 day.);Py.(IQR:2-4 day.)vs.5 day.(IQR:

The conventional perception of astrocy.es as mere supportive cells within the brain has recently.been called into question by.empirical evidence, which has revealed their active involvement in regulating brain function and encoding behaviors associated with ***, astrocy.es in the basolateral amy.dala have been found to play.a role in the modulation of anxiety.like behaviors triggered by.chronic stress. Nevertheless, the precise molecular mechanisms by.which basolateral amy.dala astrocy.es regulate chronic stress–induced anxiety.like behaviors remain to be fully.elucidated. In this study. we found that in a mouse model of anxiety.triggered by.unpredictable chronic mild stress, the expression of excitatory.amino acid transporter 2 was upregulated in the basolateral amy.dala. Interestingly. our findings indicate that the targeted knockdown of excitatory.amino acid transporter 2 specifically.within the basolateral amy.dala astrocy.es was able to rescue the anxiety.like behavior in mice subjected to stress. Furthermore, we found that the overexpression of excitatory.amino acid transporter 2 in the basolateral amy.dala, whether achieved through intracranial administration of excitatory.amino acid transporter 2agonists or through injection of excitatory.amino acid transporter 2-overexpressing viruses with GfaABC1D promoters, evoked anxiety.like behavior in mice. Our single-nucleus RNA sequencing analy.is further confirmed that chronic stress induced an upregulation of excitatory.amino acid transporter 2 specifically.in astrocy.es in the basolateral amy.dala. Urbanization and carbon emissions in China:Analy.is of dy.amic relationships from 1978 to 2020

BACKGROUND Cardioneuroablation(CNA)has shown encouraging results in patients with vasovagal sy.cope(VVS).However,data on different subty.es was *** This observational study.retrospectively.enrolled 141 patients[mean age:40±18 y.ars,51 males(36.2%)]with the diagnosis of *** characteristics among different ty.es of VVS and the outcomes after CNA were *** After a mean follow-up of 4.3±1.5 y.ars,41 patients(29.1%)experienced sy.cope/pre-sy.cope events after ***/pre-sy.cope recurrence significantly.differed in each subty.e(P=0.04).The cardioinhibitory.ty.e of VVS had the lowest recurrence rate after the procedure(n=6,16.7%),followed by.mixed(n=26,30.6%)and vasodepressive(n=9,45.0%).Additionally.a significant difference was observed in the analy.es of the Kaplan-Meier survival curve(P=0.02).Sy.cope/pre-sy.cope burden was significantly.reduced after CNA in the vasodepressive ty.e(Py.es with recurrent sy.cope/pre-sy.cope after CNA have a lower baseline deceleration capacity.DC)level than those without(7.4±1.0 ms vs.9.0±1.6 ms,P=0.01).Patients with DCy.cope/pre-sy.cope recurrence after CNA compared to patients with DC≥8.4 ms,and this association still existed after adjusting for age and sex(HR=8.1,95%CI:2.2-30.1,P=0.02).CONCLUSIONS Different subty.es exhibit different event-free *** vasodepressive ty.e exhibited the lowest event-free rate,but those patients with DC≥8.4 ms might benefit from CNA.

Py.imethamine upregulates BNIP3 to interfere SNARE-mediated autophagosome-ly.osomal fusion in hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is one of the most common tumor ty.es and remains a major clinical challenge. Increasing evidence has revealed that mitophagy.inhibitors can enhance the effect of chemotherapy.on HCC. However, few mitophagy.inhibitors have been approved for clinical use in humans. Py.imethamine (Py.) is used to treat infections caused by.protozoan parasites. Recent studies have reported that Py. may.be beneficial in the treatment of various tumors. However, its mechanism of action is still not clearly.defined. Here, we found that blocking mitophagy.sensitized cells to Py.-induced apoptosis. Mechanistically. Py. potently.induced the accumulation of autophagosomes by.inhibiting autophagosome-ly.osome fusion in human HCC cells. In vitro and in vivo studies revealed that Py. blocked autophagosome-ly.osome fusion by.upregulating BNIP3 to inhibit sy.aptosomal-associated protein 29 (SNAP29)-vesicle-associated membrane protein 8 (VAMP8) interaction. Moreover, Py. acted sy.ergistically.with sorafenib (Sora) to induce apoptosis and inhibit HCC proliferation in vitro and in vivo. Py. enhances the sensitivity.of HCC cells to Sora, a common chemotherapeutic, by.inhibiting mitophagy. Thus, these results provide new insights into the mechanism of action of Py. and imply.that Py. could potentially.be further developed as a novel mitophagy.inhibitor. Notably. Py. and Sora combination therapy.could be a promising treatment for malignant HCC.
