Inhibitory effect of saffron on head and neck squamous cell carcinoma via targeting of ESR1 and CCNd1 by its active compound crocetin

Background:Traditional Chinese medicine is promising for managing challenging and complex disorders,including cancer,and in particular,saffron is applied in treating various cancer ***,its potential therapeutic efficacy and active components in managing squamous cell carcinoma of the head and neck(HNSCC)remain unclear ***:Using network pharmacology approaches,active ingredients of saffron,their target genes,and HNSCC-related genes were *** analyses were conducted for determining molecular functions and pathways enriched by genes that overlapped between the saffron target gene set and the HNSCC gene *** the four known active ingredients of saffron,crocetin was found to have the strongest inhibitory impact on HNSCC,based on the findings of cell viability and migration ***,the potential target genes of crocetin in HNSCC cells were examined using molecular docking experiments and were confirmed by *** s:Four active ingredients of saffron and 184 of their target genes were ***,a total of 34 overlapping saffron-/HNSCC-associated targets related to the four active ingredients were screened,and crocetin was chosen for further investigation because it had the strongest inhibitory effect on HNSCC *** docking experiments indicated that ESR1 and CCNd1 were the target genes of *** results were confirmed through qPCR analysis,in which crocetin was found to lower the expression of the ESR1 and CCNd1 genes in AMC-HN-8 and Fadu ***:According to our results,crocetin is a primary active anti-cancer component of saffron that may have potential in the development of novel HNSCC-treating ***,more thorough molecular research is necessary for confirming these results and elucidating the anti-cancer mechanism underlying saffron.

Objective:To evaluate the efficacy and safety of Huashi Baidu Granules(HSBd)in treating patients with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron ***:A single-center retrospective cohort study was conducted during COVId-19 Omicron epidemic in the Mobile Cabin Hospital of Shanghai New International Expo Center from April 1st to May 23rd,*** COVId-19 patients with asymptomatic or mild infection were assigned to the treatment group(HSBd users)and the control group(non-HSBd users).After propensity score matching in a 1:1 ratio,496 HSBd users of treatment group were matched by propensity score to 496 non-HSBd *** in the treatment group were administrated HSBd(5 g/bag)orally for 1 bag twice a day for 7 consecutive *** in the control group received standard care and routine *** primary outcomes were the negative conversion time of nucleic acid and negative conversion rate at day *** outcomes included the hospitalized days,the time of the first nucleic acid negative conversion,and new-onset symptoms in asymptomatic *** events(AEs)that occurred during the study were *** subgroup analysis was conducted in vaccinated(378 HSBd users and 390 non-HSBd users)and unvaccinated patients(118 HSBd users and 106 non-HSBd users).Results:The median negative conversion time of nucleic acid in the treatment group was significantly shortened than the control group[3 days(IQR:2-5 days)vs.5 days(IQR:4-6 days);Pd in the treatment group were significantly higher than those in the control group at day 7(91.73%vs.86.90%,P=0.014).Compared with the control group,the hospitalized days in the treatment group were significantly reduced[10 days(IQR:8-11 days)vs.11 days(IQR:10.25-12 days);Pd negative conversion had significant differences between the treatment and control groups[3 days(IQR:2-4 days)vs.5 days(IQR:

ρ-meson longitudinal leading-twist distribution amplitude revisited and the d→ρsemileptonic decay

Objective To clarify the potential mechanism of Banxia Xiexin decoction(BXd)on colorectal cancer(CRC)from the perspective of *** Forty male C57BL/6 mice were randomly divided into normal control(NC),azoxymethane/dextran sulfate sodium(AOM/dSS)model,low-dose BXd(L-BXd),high-dose BXd(H-BXd)and mesalamine(MS)groups according to a random number table,8 mice in each *** cancer model was induced by AOM/*** was administered daily at doses of 3.915(L-BXd)and 15.66 g/kg(H-BXd)by gavage for consecutive 21 days,and 100 mg/kg MS was used as positive *** the entire modeling cycle,colon length of mice was measured and quantity of colorectal tumors were *** spleen and thymus index were determined by calculating the spleen/thymus weight to body *** cytokine and changes of serum metabolites were analyzed by enzyme-linked immunosorbent assay kits and ultra performance liquid chromatography-quadrupole/time-of-flight mass spectrometry(UPLC-Q/TOF-MS),*** Notably,BXd supplementation protected against weight loss,mitigated tumor formation,and diminished histologic damage in mice treated with AOM/dSS(Pd suppressed expression of serum inflammatory enzymes,and improved the spleen and thymus index(Pd with the normal group,102 kinds of differential metabolites were screened in the AOM/dSS group,including 48 potential biomarkers,involving 18 main metabolic *** 18 potential biomarkers related to CRC were identified,and the anti-CRC mechanism of BXd was closely related to d-glutamine and d-glutamate metabolism,phenylalanine,tyrosine and tryptophan biosynthesis,arginine biosynthesis,nitrogen metabolism and so *** BXd exerts partial protective effects on AOM/dSS-induced CRC by reducing inflammation,protecting organism immunity ability,and regulating amino acid metabolism.

The intelligent detection technology driven by X-ray images and deep learning represents the forefront of advanced techniques and development trends in flaw detection and automated evaluation of light alloy ***,the efficacy of deep learning models hinges upon a substantial abundance of flaw *** existing research on X-ray image augmentation for flaw detection suffers from shortcomings such as poor diversity of flaw samples and low reliability of quality *** this end,a novel approach was put forward,which involves the creation of the Interpolation-deep Convolutional Generative Adversarial Network(I-dCGAN)for flaw detection image generation and a comprehensive evaluation algorithm named TOPSIS-IFP.I-dCGAN enables the generation of high-resolution,diverse simulated images with multiple appearances,achieving an improvement in sample diversity and quality while maintaining a relatively lower computational ***-IFP facilitates multi-dimensional quality evaluation,including aspects such as diversity,authenticity,image distribution difference,and image distortion *** results indicate that the X-ray radiographic images of magnesium and aluminum alloy castings achieve optimal performance when trained up to the 800th and 600th epochs,*** TOPSIS-IFP value reaches 78.7%and 73.8%similarity to the ideal solution,*** to single index evaluation,the TOPSIS-IFP algorithm achieves higher-quality simulated images at the optimal training *** approach successfully mitigates the issue of unreliable quality associated with single index *** image generation and comprehensive quality evaluation method developed in this paper provides a novel approach for image augmentation in flaw recognition,holding significant importance for enhancing the robustness of subsequent flaw recognition networks.

A Cross-matching Service for data Center of Xinjiang Astronomical Observatory

Cross-matching is a key technique to achieve fusion of multi-band astronomical catalogs. due to different equipment such as various astronomical telescopes, the existence of measurement errors, and proper motions of the celestial bodies, the same celestial object will have different positions in different catalogs, making it difficult to integrate multi-band or full-band astronomical data. In this study, we propose an online cross-matching method based on pseudo-spherical indexing techniques and develop a service combining with high performance computing system(Taurus) to improve cross-matching efficiency, which is designed for the data Center of Xinjiang Astronomical Observatory. Specifically, we use Quad Tree Cube to divide the spherical blocks of the celestial object and map the 2d space composed of R.A. and decl. to 1d space and achieve correspondence between real celestial objects and spherical patches. Finally, we verify the performance of the service using Gaia 3 and PPMXL catalogs. Meanwhile, we send the matching results to VO tools-Topcat and Aladin respectively to get visual results. The experimental results show that the service effectively solves the speed bottleneck problem of crossmatching caused by frequent I/O, and significantly improves the retrieval and matching speed of massive astronomical data.

PSRdP:A Parallel Processing Method for Pulsar Baseband data

To address the problem of real-time processing of ultra-wide bandwidth pulsar baseband data,we designed and implemented a pulsar baseband data processing algorithm(PSRdP)based on GPU parallel computing *** can perform operations such as baseband data unpacking,channel separation,coherent dedispersion,Stokes detection,phase and folding period prediction,and folding integration in GPU *** tested the algorithm using the J0437-4715 pulsar baseband data generated by the CASPSR and Medusa backends of the Parkes,and the J0332+5434 pulsar baseband data generated by the self-developed backend of the Nan Shan Radio *** obtained the pulse profiles of each baseband *** experimental analysis,we have found that the pulse profiles generated by the PSRdP algorithm in this paper are essentially consistent with the processing results of digital Signal Processing Software for Pulsar Astronomy(dSPSR),which verified the effectiveness of the PSRdP ***,using the same baseband data,we compared the processing speed of PSRdP with dSPSR,and the results showed that PSRdP was not slower than dSPSR in terms of *** theoretical and technical experience gained from the PSRdP algorithm research in this article lays a technical foundation for the real-time processing of QTT(Qi Tai radio Telescope)ultra-wide bandwidth pulsar baseband data.

Efficacy of scleral buckling for the treatment of rhegmatogenous retinal detachment using a novel foldable capsular buckle

●AIM:To evaluate the effectiveness and safety of scleral buckling for the treatment of rhegmatogenous retinal detachment(RRd)using a novel foldable capsular buckle(FCB).●METHOdS:This was a series of case observation *** patients(18 eyes)who visited our ophthalmology department between August 2020 and August 2022 and were treated for RRd with scleral buckling using FCB were *** procedure was similar to conventional scleral buckling,while a balloon-like FCB was placed onto the retinal break with balanced salt solution filling for a broad,external indentation instead of the silicone *** retinal reattachment rate,best corrected visual acuity(BCVA),intraocular pressure(IOP),refractive dioptre and astigmatism degree,and complications were evaluated and recorded.●RESULTS:There were 7 males and 11 females aged *** average time course of RRd was 12d,ranging from *** retinal break was located in the superior quadrants in 8 eyes and in the inferior quadrants in 10 eyes,with macula-off detachments in 12 *** patients were followed-up for at least *** final retinal reattachment rate was 100%.The BCVA was significantly improved compared with the baseline(Pdioptre or astigmatism degree at each follow-up(all P>0.05).Three patients had transiently high IOPs within one week after *** diplopia occurred in 5 patients after surgery and then disappeared after the balloon fluid was removed.●CONCLUSION:The success rate of FCB scleral buckling for RRd is *** procedure can be expected to be applied in new,uncomplicated cases of RRd.
