Understanding the micro-coevolution of the human gut microbiome with host genetics is challenging but essential in both evolutionary and medical *** gain insight into the interactions between host genetic variation and the gut microbiome,we analyzed both the human genome and gut microbiome collected from a cohort of 190 students in the same boarding college and representing 3 ethnic groups,Uyghur,Kazakh,and Han *** found that differences in gut microbiome were greater between genetically distinct ethnic groups than those genetically closely related ones in taxonomic composition,functional composition,enterotype stratification,and microbiome genetic *** also observed considerable correlations between host genetic variants and the abundance of a subset of gut microbial ***,interactions between gut microbiome species and host genetic variants might have coordinated effects on specific human *** ovatus,previously reported to modulate intestinal immunity,is significantly correlated with the host genetic variant rs12899811(meta-P=5.55×10^(-5)),which regulates the VPS33B expression in the colon,acting as a tumor suppressor of colorectal *** results advance our understanding of the micro-coevolution of the human gut microbiome and their interactive effects with host genetic variation on phenotypic diversity.

Human genetic adaptation to high altitudes(>2500 m) has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified, largely owing to the lack of deep-genome sequencing data available to previous studies. Here, we build a list of putative adaptive variants, including 63 missense, 7 loss-of-function, 1,298 evolutionarily conserved variants and 509 expression quantitative traits loci. Notably, the top signal of selection is located in TMEM247, a transmembrane protein-coding *** Tibetan version of TMEM247 harbors one high-frequency(76.3%) missense variant, rs116983452(c.248 C > T; ***83 Val), with the T allele derived from archaic ancestry and carried by >94% of Tibetans but absent or in low frequencies(<3%) in non-Tibetan populations. The rs116983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p = 5.78 × 10), red blood cell count(p = 5.72 × 10) and hematocrit(p = 2.57 × 10). In particular, TMEM247-rs116983452 shows greater effect size and better predicts the phenotypic outcome than any EPAS1 variants in association with adaptive traits in Tibetans. Modeling the interaction between TMEM247-rs116983452 and EPAS1 variants indicates weak but statistically significant epistatic *** results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.

基于护士危机意识的预警性管理干预应用于脑卒中偏瘫患者中的效果观察

目的:探讨基于护士危机意识的预警性管理干预在脑卒中偏瘫患者中的应用效果。方法:选取2020年1月-2022年6月在江苏省荣军医院接受治疗的116例脑卒中偏瘫患者为研究对象,将其随机分为两组,各58例。对照组实施常规干预,观察组实施基于护士危机意识的预警性管理干预。比较两组下肢深静脉血栓形成(DVT)与下肢肿胀发生情况,肢体运动功能、负性情绪及生活质量。结果:干预1周后,观察组DVT发生率与下肢肿胀发生率均明显低于对照组,差异有统计学意义(P<0.05)。观察组干预1周后上肢评分、下肢评分及总分均明显高于对照组,差异有统计学意义(P<0.05)。观察组干预1周后焦虑自评量表(SAS)评分、抑郁自评量表(SDS)评分均明显低于对照组,差异有统计学意义(P<0.05)。观察组干预1周后脑卒中专用生活质量量表(SS-QOL)评分明显高于对照组,差异有统计学意义(P<0.05)。结论:基于护士危机意识的预警性管理干预的应用,可降低脑卒中偏瘫患者发生DVT与下肢肿胀等不良事件,提高其肢体运动功能,同时还能够显著缓解患者的负性情绪,促进其生活质量的改善。

目的:探讨脑卒中偏瘫病人日常生活活动能力训练中引入活动分析法的效果。方法:将2021年6月—2022年6月收治的符合纳入标准的脑卒中偏瘫病人80例,按入院先后顺序分为两组,各40例。对照组实施常规日常生活活动能力训练,试验组在此基础上引入活动分析法。两组训练时间为每次30 min,每周1次,持续训练4周。评价两组训练前后运动功能、日常生活活动能力、神经功能及步行能力变化,进行比较分析。结果:试验组训练4周后上肢、下肢功能评分明显提高,且高于对照组(P<0.05);试验组训练4周后日常生活活动能力明显增强,且强于对照组(P<0.05);试验组训练4周后神经功能明显恢复,且优于对照组(P<0.05);试验组训练4周后步行能力明显提高,且优于对照组(P<0.05)。结论:脑卒中偏瘫病人日常生活活动能力训练中引入活动分析法可改善运动功能及日常生活活动能力,促进神经功能恢复,提高步行能力。

Activin A, which was first described in 1986, has been shown to maintain hippocampal neuronal survival. Activin A increases intracellular free Ca2＋ via L-type Ca2＋ channels. Our previous study showed that activin A promotes neurite growth of dorsal root ganglia in embryonic chickens and inhibits nitric oxide secretion. The present study demonstrated for the first time that activin A could maintain cerebral cortex neuronal survival in vitro for a long period, and that activin A was shown to increase voltage-gated Na＋ current （/Na） in Neuro-2a cells, which was recorded by patch clamp technique. The present study revealed a novel mechanism for activin A, as well as the influence of activin A on neurons by regulating expressions of vasoactive intestine peptide and inducible nitric oxide synthase.

Molecular insights on Ca^(2+)/Na+separation via graphene-based nanopores:The role of electrostatic interactions to ionic dehydration

Ca^(2+)/Na+separation is a common problem in industrial applications,biological and medical ***,Ca^(2+)and Na+have similar ionic radii and hydration radii,thus Ca^(2+)/Na+separation is *** by biological channels,group modification is one of the effective methods to improve the separation *** this work,molecular dynamics simulations were performed to investigate the effects of different functional groups(COO,NH3+)on the separation performance of Ca^(2+)and Na+through graphene nanopores under an electric *** pristine graphene nanopore was used for *** showed that three types of nanopores preferred Ca^(2+)to Na+,and Ca^(2+)/Na+selectivity followed the order of ge-COO(4.06)>ge(1.85)>ge-NH3+(1.63).Detailed analysis of ionic hydration microstructure shows that different nanopores result in different hydration factors for the second hydration layer of Ca^(2+)and the first layer of Na+.Such different hydration factors corresponding to the dehydration ability can effectively evaluate the separation *** addition,the breaking of hydrogen bonds between water molecules due to electrostatic effects can directly affect the dehydration ***,the electrostatic effect generated by group modification will affect the ionic hydration microstructure,thus reflecting the differences in dehydration *** in turn affects the permeable and separation performance of *** results of this work provide perceptive guidelines for the application of graphene-based membranes in ion separation.

Direct Effects of Activin A on the Activation of Mouse Macrophage RAW264.7 Cells

Macrophages play critical roles in innate immune and acquired immune v/a secreting pro-inflammatory mediators, phagocytosing microorganisms and presenting antigens. Activin A, a member of transforming growth factor （TGF-β） superfamily, is produced by macrophages and microglia cells. In this study, we reported a direct effect of activin A as a pro-inflammatory factor on mouse macrophage cell line RAW264.7 cells. Our data revealed that activin A could not only increase IL-1β and IL-6 production from RAW264.7 cells, but also promote pinocytic and phagocytic activities of RAW264.7 cells. In addition, activin A obviously up-regulated MHC Ⅱ expression on the surface of RAW264.7 cells, whereas did not influence MHC I expression. Activin A also enhanced CD80 expression, which is a marker of activated macrophages, but did not influence RAW264.7 cell proliferation. These data suggest that activin A may regulate primary macrophage-mediated innate and acquired immune response via promoting the activation of rest macrophages.

A Critical Role of Activin A in Maturation of Mouse Peritoneal Macrophages in vitro and in vivo

Activin A, a multifunctional factor of the transforming growth factor-beta （TGF-β） superfamily, is mainly produced by microglia and macrophages, and its anti-inflammatory and pro-inflammatory activities are both related to macrophage functions. However the direct effect of activin A on the rest macrophages in vivo remains unclear. In the present study, the results showed that activin A not only increased NO and IL-1β release, but also promoted phagocytic abilities of mouse peritoneal macrophages in vitro and in vivo, whereas it did not influence MHC Ⅰ and MHC Ⅱ expression. Moreover, we found that activin A significantly upregulated the expressions of CD14 and CD68, markers of mature macrophages, on the surface of macrophages in vitro and in vivo. These data suggest that activin A can induce primary macrophage maturation in vitro and in vivo, but may not trigger the acquired immune response via regulating expression of MHC molecules involved in presentation of antigen.
