Construction of fault diagnosis system for control rod drive mechanism based on knowledge graph and Bayesian inference

Knowledge graph technology has distinct advantages in terms of fault *** this study,the control rod drive mechanism(CRDM)of the liquid fuel thorium molten salt reactor(TMSR-LF1)was taken as the research object,and a fault diagnosis system was proposed based on knowledge *** subject–relation–object triples are defined based on CRDM unstructured data,including design specification,operation and maintenance manual,alarm list,and other forms of expert *** this study,we constructed a fault event ontology model to label the entity and relationship involved in the corpus of CRDM fault events.A three-layer robustly optimized bidirectional encoder representation from transformers(RBT3)pre-training approach combined with a text convolutional neural network(TextCNN)was introduced to facilitate the application of the constructed CRDM fault diagnosis graph database for fault *** RBT3-TextCNN model along with the Jieba tool is proposed for extracting entities and recognizing the fault query intent *** on the dataset collected from TMSR-LF1 CRDM fault diagnosis unstructured data demonstrate that this model has the potential to improve the effect of intent recognition and entity ***,a fault alarm monitoring module was developed based on WebSocket protocol to deliver detailed information about the appeared fault to the operator ***,the Bayesian inference method combined with the variable elimination algorithm was proposed to enable the development of a relatively intelligent and reliable fault diagnosis ***,a CRDM fault diagnosis Web interface integrated with graph data visualization was constructed,making the CRDM fault diagnosis process intuitive and effective.

A BIOREDUCIBLE POLYPEPTIDE FOR EFFICIENT GENE TRANSFECTION BOTH IN VITRO AND IN VIVO

A disulfide bond containing polypeptide, PolyK6-R8, was designed and prepared by oxidative polymerization between terminal cysteinyl thiol groups of CHK6HC and CR8C. The molar ratio between CHK6HC and CR8C within obtained PolyK6-Rs was 9：1, and PolyK6-R8 could combine DNA compactly when weight ratio reached 5. Through in vitro investigation, it was found that PolyK6-R8 was an efficient gene vector with low cytotoxicity for delivering DNA in both COS-7 and HeLa cells. Cellular uptake of DNA mediated by PolyK6-R8/DNA complexes was promoted after incubation for 4 h. Moreover, by examining the histological sections of hindlimb ischemia rats through immunohistochemistry, PolyK6- R8/VEGF complexes were proved to be effective in both VEGF protein expression and succeeding vessel formation. The results indicated that polypeptide-based PolyK6-R8 is a promising gene vector for the limb ischemia treatment.

Autologous transplantation of bone marrow mononuclear cells improved heart function after myocardial infarction

AIM: To investigate whether autologous transplantation of adult stem cells could improve post-infarcted heart function. METHODS: Bone marrow mononuclear cells (MNCs) were isolated from adult rabbits’ tibias after coronary ligation. These cells were exposed to 5-azacytidine 10 μmol/L for 24 h on the third day of culture. After being labeled with bromodeoxyuridine (BrdU), the cells were auto-transplanted into bordering zone of the infarcted area at 2 weeks after injury. The animals were killed at 3 days, 2 weeks, 1 month, and 2 months after transplantation, respectively. The left ventricular functions, capillary density, and cardiac nerve density were measured and the differentiation of the engrafted cells was determined by immunostaining. RESULTS: BrdU-labeled MNCs were well aligned with the host cardiomyocytes. Parts of them were incorporated into capillary and arteriolar vessel walls. In addition to inducing angiogenic ligands (basic fibroblast growth factor, vascular endothelial growth factor) and imflammation cytokines (interleukin 1-β) during the early period of MNCs implantation, MNCs induced 2.0-fold increase in capillary density as well. Moreover, GAP43-positive and TH-positive nerve density were markedly higher in the MNCs-treated groups than that in the non-treated hearts. Left ventricular ejection fraction, LV+dp/dtmax, and LV-dp/dtmax were 47 %, 67 %, and 55 % in MNCs-treated heart respectively, which was higher than that of the control heart, whereas left ventricular end-diastolic volume, left ventricular end-diastolic diameter, and left ventricular end-diastolic pressure were 45 %, 22 %, and 50 % respectively in MNCs-treated heart, which was lower than that of the control heart at 2 months after cell transplantation. CONCLUSION: Autologous transplantation of MNCs induced angiogenesis and nerve sprouting and improved left ventricular diastolic function.

Cox7a2 mediates steroidogenesis in TM3 mouse Leydig cells

Aim： To investigate the regulatory function of Cox7a2 on steroidogenesis and the mechanism involved in TM3 mouse Leydig cells. Methods： The cDNA of Cox7a2 was cloned from TM3 mouse Leydig cells. It was subcloned to pDsRed- Express-N 1 and transfected back into TM3 mouse Leydig cells for Cox7a2 overexpression by transient gene transfection. Steroidogenesis affected by overexpressed Cox7a2 was studied by ELISA. To elicit the mechanism of this effect, expression of steroidogenic acute regulatory （STAR） protein and reactive oxygen species （ROS） were examined by Western blot and fluorometer, respectively. Results： The cDNA of Cox7a2 （249 bp） was cloned from Leydig cells and confirmed by DNA sequencing. After constructed pDsRed-Express-N1-Cox7a2 was transfected back into TM3 mouse Leydig cells, Cox7a2 inhibited not only luteinizing hormone （LH）-induced secretion of testosterone but also the expression of StAR protein. At the same time, Cox7a2 increased the activity of ROS in TM3 mouse Leydig cells. Conclusion： Cox7a2 inhibited LH-induced StAR protein expression, and consequent testosterone production, at least in part, by increasing ROS activity in TM3 mouse Leydig cells.

Expression and localization of CKLFSF2 in human spermatogenesis

Aim： To investigate the expression and subcellular localization of chemokine-like factor superfamily 2 （CKLFSF2） in human testis and its potential role in spermatogenesis. Methods： A specific polyclonal antibody against CKLFSF2 was raised. The expression and cellular localization of CKLFSF2 in the seminiferous tubules was checked by immunohistochemistry method. Also, in situ hybridization was applied to localize the mRNA distribution. The EGFP- CKLFSF2 fusion protein was expressed in COS-7 cells to localize its subcellular location in vitro. In addition, the abnormal expression of CKLFSF2 in testes of patients with male infertility was assayed by reverse transcription polymerase chain reaction （RT-PCR） and immunohistochemistry methods. Results： Having a close correlation with spermatogenesis defects, CKLFSF2 was specifically expressed in meiotic and post-meiotic germ cells, which were localized to the endoplasmic reticulum （ER） near the Golgi apparatus. Conclusion： CKLFSF2 could play important roles in the process of meiosis and spermiogenesis, and might be involved in the vesicular transport or membrane apposition events in the endoplasmic reticulum.

Experimental Study on Wire Melting Control Ability of Twin-Body Plasma Arc

The twin-body plasma arc has the decoupling control ability of heat transfer and mass transfer,which is beneficial to shape and property control in wire arc additive *** this paper,with the wire feeding speed as a characteristic quantity,the wire melting control ability of twin-body plasma arc was studied by adjusting the current separation ratio(under the condition of a constant total current),the wire current/main current and the position of the wire in the arc axial *** results showed that under the premise that the total current remains unchanged(100 A),as the current separation ratio increased,the middle and minimum melting amounts increased approximately synchronously under the effect of anode effect power,the first melting mass range remained constant;the maximum melting amount increased twice as fast as the middle melting amount under the effect of the wire feeding speed,and the second melting mass range was *** the wire current increased,the anode effect power and the plasma arc power were both factors causing the increase in the wire melting amount;however,when the main current increased,the plasma arc power was the only factor causing the increase in the wire melting *** average wire melting increment caused by the anode effect power was approximately 2.7 times that caused by the plasma arc *** minimum melting amount was not affected by the wire-torch distance under any current separation ratio *** the current separation ratio increased and reached a threshold,the middle melting amount remained constant with increasing wire-torch *** the current separation ratio continued to increase and reached the next threshold,the maximum melting amount remained constant with the increasing wire-torch *** effect of the wire-torch distance on the wire melting amount reduced with the increase in the current separation *** this study,the decoupling mechanism and ability of this innovative

Origin of natural sulfur-metal chimney in the Tangyin hydrothermal field,Okinawa Trough:constraints from rare earth element and sulfur isotopic compositions

For the first time,we present the rare earth element (REE)and sulfur isotopic composition of hydrothermal precipitates recovered from the Tangyin hydrothermal field (THF),Okinawa Trough at a water depth of 1206 *** natural sulfur samples exhibit the lowest ∑REE concentrations (∑REE= 0.65×10^-6-4.580×10^-6)followed by metal sulfides (∑REE=1.71×10^-6-11.63×10^-6).By contrast,the natural sulfur-sediment samples have maximum ∑REE concentrations (∑REE=1 1.54×10^-6-33.06×10^-6), significantly lower than those of the volcanic and sediment ***,the δEu,δCe,(La/Yb)N, La/Sm,(Gd/Yb)N and normalized patterns of the natural sulfur and metal sulfide show the most similarity to the *** hydrothermal precipitate samples are characterized by enrichments of LREE (LREE/HREE=10.09-24.53)arid slightly negative Eu anomalies or no anomaly (δEu=0.48-0.99),which are different from the hydrothermal fluid from sediment-free mid-oceanic ridges and back-are basins,but identical to the sulfides from the Jade hydrothermal *** lower temperature and more oxidizing conditions produced by the mixing between seawater and hydrothermal fluids further attenuate the leaching ability of hydrothermal fluid,inducing lower REE concentrations for natural sulfur compared with metal sulfide;meanwhile,the negative Eu anomaly is also weakened or almost *** sulfur isotopic compositions of the natural sulfur (δ^34S=3.20‰-5.01‰,mean 4.23 ‰)and metal sulfide samples (δ34S=0.82‰-0.89‰,mean 0.85‰)reveal that the sulfur of the chimney is sourced from magmatic degassing.

Pollution characteristics and quantitative source apportionment of heavy metals within a zinc smelting site by GIS-based PMF and APCS-MLR models

The abandoned smelters present a substantial pollution threat to the nearby soil and groundwater. In this study, 63 surface soil samples were collected from a zinc smelter to quantitatively describe the pollution characteristics, ecological risks, and source apportionment of heavy metal(loid)s (HMs). The results revealed that the average contents of Zn, Cd, Pb, As, and Hg were 0.4, 12.2, 3.3, 5.3, and 12.7 times higher than the risk screening values of the construction sites, respectively. Notably, the smelter was accumulated heavily with Cd and Hg, and the contribution of Cd (0.38) and Hg (0.53) to ecological risk was 91.58%. ZZ3 and ZZ7 were the most polluted workshops, accounting for 25.7% and 35.0% of the pollution load and ecological risk, respectively. The influence of soil parent materials on pollution was minor compared to various workshops within the smelter. Combined with PMF, APCS-MLR and GIS analysis, four sources of HMs were identified: P1(25.5%) and A3(18.4%) were atmospheric deposition from the electric defogging workshop and surface runoff from the smelter; P2(32.7%) and A2(20.9%) were surface runoff of As-Pb foul acid; P3(14.5%) and A4(49.8%) were atmospheric deposition from the leach slag drying workshop; P4(27.3%) and A1(10.8%) were the smelting process of zinc products. This paper described the distribution characteristics and specific sources of HMs in different process workshops, providing a new perspective for the precise remediation of the smelter by determining the priority control factors. Lp-solutions of Multi-dimensional Oblique Reflected BSDEs and Optimal Switching Problem on Finite or Infinite Time Horizon

In this paper, we study mulit-dimensional oblique reflected backward stochastic differential equations（RBSDEs） in a more general framework over finite or infinite time horizon, corresponding to the pricing problem for a type of real option. We prove that the equation can be solved uniquely in Lp（1 < p ≤ 2）-space,when the generators are uniformly continuous but each component taking values independently. Furthermore,if the generator of this equation fulfills the infinite time version of Lipschitzian continuity, we can also conclude that the solution to the oblique RBSDE exists and is unique, despite the fact that the values of some generator components may affect one another.
