The leopard coral grouper(Plectropomus leopardus)is a species of significant economic *** artificial cultivation of *** has thrived in recent decades,the advancement of selective breeding has been hindered by the lack of comprehensive population genomic *** this study,we identified over 8.73 million single nucleotide polymorphisms(SNPs)through whole-genome resequencing of 326 individuals spanning six distinct ***,we categorized 226 individuals with high-coverage sequencing depth(≥14×)into eight clusters based on their genetic profiles and phylogenetic ***,four of these clusters exhibited pronounced genetic differentiation compared with the other *** identify potentially advantageous loci for ***,we examined genomic regions exhibiting selective sweeps by analyzing the nucleotide diversity(θπ)and fixation index(FST)in these four *** these high-coverage resequencing data,we successfully constructed the first haplotype reference panel specific to *** achievement holds promise for enabling high-quality,***,we combined low-coverage sequencing data with imputation techniques for a genome-wide association study,aiming to identify candidate SNP loci and genes associated with growth traits.A significant concentration of these genes was observed on chromosome 17,which is primarily involved in skeletal muscle and embryonic development and cell ***,our detailed investigation of growth-related SNPs across the eight clusters revealed that cluster 5 harbored the most promising candidate SNPs,showing potential for genetic selective breeding *** findings provide a robust toolkit and valuable insights into the management of germplasm resources and genome-driven breeding initiatives targeting ***.

Demethylase-assisted site-specific detection of N^(1)-methyladenosine in RNA

The dynamic RNA modifications have been viewed as new posttranscriptional regulator in modulating gene expression as well as in a broad range of physiological processes.N^(1)-methyladenosine(m^(1)A)is one of the most prevalent modifications existing in multiple types of ***-depth investigation of the functions of m^(1)A requires the site-specific assessment of m^(1)A stoichiometry in ***,we established a demethylase-assisted method(DA-m^(1)A)for the site-specific detection and quantification of m^(1)A in RNA.N^(1)-methyl group in m^(1)A could result in the stalling of reverse transcription at m^(1)A site,thus producing the truncated *** AlkB is a demethylase that can demethylate m^(1)A to produce adenine in RNA,thus generating full-length cDNA from AlkB-treated *** of the produced amounts of full-length cDNA by quantitative real-time PCR can achieve the site-specific detection and quantification of m^(1)A in *** the DA-m^(1)A method,we examined and successfully confirmed the previously well-characterized m^(1)A sites in various types of RNAs with low false positive *** addition,we found that the level of m^(1)A was significantly decreased at the bromodomain containing 2(BRD2)mRNA position 1674 and CST telomere replication complex component 1(CTC1)mRNA position 5643 in human hepatocellular carcinoma *** results suggest that these two m^(1)A sites in mRNA may be involved in liver *** together,the DA-m^(1)A method is simple and enables the rapid,cost-effective,and site-specific detection and quantification of m^(1)A in RNA,which provides a valuable tool to decipher the functions of m^(1)A in human diseases.

表情变换时序特征下的驾驶人情绪识别研究

针对现有驾驶人情绪识别方法存在的识别实时性不足、识别精度较低等问题,提出一种表情识别及其时序情绪表达的驾驶人情绪识别方法。首先,建立VGG Lite驾驶人表情识别模型,在传统VGG Net模型结构上,通过改变卷积层堆叠结构以大幅减少模型的参数量,修改激活函数以增强模型对人脸表情中细节特征的表达能力,并在模型中增加性能优化层来提升模型的收敛性和泛化性。其次,分析表情时序变化与情绪状态之间的关系,研究时间序列演变的情绪表达方式,设计了包含表情时序转化、表情-情绪量化映射和时序情绪表达的驾驶人时序情绪识别方法。然后,采用Fer2013数据集,将所提出的VGG Lite驾驶人表情识别模型与其他模型进行比较验证,证明了该模型不仅可以保持高识别准确率,还有效降低了模型参数量,从而提高了识别速度,此外,采用自制数据集识别表情获得了98.8%的高准确率,证明了该模型能有效识别不同驾驶情境中的驾驶人表情。最后,以公交车驾驶人情绪识别为例对提出的时序情绪识别方法进行试验验证,结果表明,该方法能够准确识别驾驶人各种表情转换下的复杂情绪状态,平均识别率高于95%,比单帧情绪识别方法提升5%以上,每帧图像的情绪识别耗时平均低于0.03 s,每秒平均识别超过10帧,满足交通驾驶情绪识别的实时性要求。所提方法能够及时、准确地评估驾驶人的情绪状态,为提高交通系统整体安全性和效率提供了更有效的手段。

随着2000国家大地坐标系的全面启用,原有地方坐标系已无法满足现代城市建设和社会发展的需要。论文阐述了地方坐标系建立原则和方法,结合投影变形理论对相对独立平面坐标系建立影响因素中中央子午线和投影面的选取进行了详细分析,并以淮安区域为例,结合淮安DEM,详细分析了淮安2000坐标系在淮安市各区县的投影变形情况,同时用格网占比法分析了其与原有坐标系的面积变化。结果表明,淮安2000坐标系在淮安区域内小于2.5 cm/km面积占淮安陆域面积的99.8%,最大投影变形为3.303 cm/km,位于丘陵区域;与原有坐标系相比,在面积变化上优于万分之一,具有良好的适用性。

Research on an innovative structure of an open-ribbed steel–ultra-high performance concrete composite bridg

To completely solve the problem of fatigue cracking issue of orthotropic steel bridge decks(OSDs),the authors proposed a steel–ultra-high performance concrete(UHPC)lightweight composite deck(LWCD)with closed ribs in *** on the successful application of that LWCD,an adaptation incorporating an innovative composite deck structure,i.e.,the hot-rolled section steel–UHPC composite deck with open ribs(SSD)is proposed in this paper,aiming to simplify the fabrication process as well as to reduce the cost of *** on a long-span cable-stayed bridge,a design scheme is proposed and is compared with the conventional OSD ***,a finite element(FE)calculation is conducted to reflect both the global and local behavior of the SSD scheme,and it is found that the peaked stresses in the SSD components are less than the corresponding allowable values.A static test is performed for an SSD strip specimen to understand the anti-cracking behavior of the UHPC layer under negative bending *** static test results indicate that the UHPC layer exhibited a satisfactory tensile toughness,the UHPC tensile strength obtained from the test is 1.8 times the calculated stress by the FE model of the real *** addition,the fatigue stresses of typical fatigue-prone details in the SSD are calculated and evaluated,and the influences of key design parameters on the fatigue performance of the SSD are *** to the fatigue results,the peaked stress ranges for all of the 10 fatigue-prone details are within the corresponding constant amplitude fatigue *** a fatigue test is carried out for another SSD strip specimen to explore the fatigue behavior of the fillet weld between the longitudinal and transverse *** specimen failed at the fillet weld after equivalent 47.5 million cycles of loading under the design fatigue stress range,indicating that the fatigue performance of the SSD could meet the fatigue design *** calculations and experim

Predictive value of serum alpha-fetoprotein for tumor regression after preoperative chemotherapy for rectal cancer

BACKGROUND Preoperative therapy is widely used in locally advanced rectal *** can improve local control of rectal ***,there are few indicators that can predict the effect of preoperative chemotherapy *** To investigate whether the increase in serumα-fetoprotein(AFP)can predict better efficacy of preoperative *** This was a retrospective *** analyzed 125 patients admitted between 2017 and 2019 with locally advanced rectal *** patients received six cycles of preoperative chemotherapy(mFOLFOX6 every 2 wk).Serum AFP of 26 patients rose slightly after three or four cycles of chemotherapy,and fell to normal again within 2 *** other 99 patients had a normal level of serum AFP during *** were divided into two groups(AFP risen and AFP normal).According to postoperative pathology,we compared tumor regression and complete response rate between the two *** primary outcome measure was the tumor regression grade(TRG)after *** difference in pathological complete response between the two groups was also *** There were no tumor progression and distant metastasis in both groups during preoperative *** in the AFP risen group achieved better TRG 0/1 than those in the AFP normal group(61.5%vs 39.4%).The increase in AFP was a significant predictor for better tumor regression[χ2=4.144,odds ratio(OR)=2.666,P=0.04].In the AFP risen group,the complete response rate was 30.8%,which was higher than in the AFP normal group(30.8%vs 12.1%,χ2=4.542,OR=3.251,P=0.03).CONCLUSION Patients with a slight increase in serum AFP can achieve better tumor regression during preoperative chemotherapy,and are more likely to achieve pathological complete response.

Osteosarcoma is the most common primary malignancy of bones and primarily occurs in adolescents and young ***,a second smaller peak of osteosarcoma incidence was reported in the elderly aged more than *** patients with osteosarcoma exhibit different characteristics compared to young patients,which usually results in a poor *** mechanism underlying osteosarcoma development in elderly patients is intriguing and of significant value in clinical *** cells can accelerate tumor progression by metabolic *** research has shown that methylmalonic acid(MMA)was significantly up-regulated in the serum of older individuals and played a central role in the development of aggressive *** found that the significant accumulation of MMA in elderly patients imparted proliferative potential to osteosarcoma *** expression of MAFB was excessively up-regulated in osteosarcoma specimens and was further enhanced in response to MMA accumulation as the patient ***,we first confirmed a novel molecular mechanism between cellular senescence and cancer,in which the MMA-driven transcriptional reprogramming of the MAFB-NOTCH3 axis accelerated osteosarcoma progression via the activation of PI3K-AKT ***,the down-regulation of the MAFB-NOTCH3 axis increased the sensitivity and effect of AKT inhibitors in osteosarcoma through significant inhibition of AKT *** conclusion,we confirmed that MAFB is a novel age-dependent biomarker for osteosarcoma,and targeting the MAFB-NOTCH3 axis in combination with AKT inhibition can serve as a novel therapeutic strategy for elderly patients with osteosarcoma in experimental and clinical trials.
