Objective： To investigate the effects ofursolic acid on the proliferation and apoptosis of human HT-29 colon cancer cells. Methods： 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） and flow cytometry assays were performed to evaluate the effects of ursolic acid on the growth and apoptosis of HT-29 cells. Western blot analysis was applied to investigate the inhibitory efibcts of ursolic acid on the phosphorylation of the epidermal growth factor receptor （EGFR）, extracellular signal-regulated kinase （ERK）, c-Jun N-terminal kinase （JNK）, and p38 mitogen-activated protein kinase （p38 MAPK）, and the activity of B cell leukemia-2 （Bcl-2）, B cell leukemia-xL （Bcl-xL）, caspase-3, and caspase-9. Results： Ursolic acid inhibited the growth of HT-29 cells in dose- and time-dependent manners. The median inhibition concentration （IC50） values for 24, 48, and 72 h treatment were 26, 20, and 18 pmol/L, respectively. The apoptotic rates of 10, 20, and 40 μmol/L ursolic acid treatments for 24 h were 5.74%, 14.49%, and 33.05%, and for 48 h were 9%, 21.39%, and 40.49%, respectively. Ursolic acid suppressed the phosphorylation of EGFR, ERK1/2, p38 MAPK, and JNK, which is well correlated with its growth inhibitory effect. 10, 20, and 40 μmol/L ursolic acid significantly inhibited the proliferation of EGF-stimulated HT-29 cells （P〈0.05）. Cell proliferation was most significantly inhibited when treated with 10 and 20 μmol/L ursolic acid combined with 200 nmol/L AG 1478 or 10 μmol/L U0126 （P〈0.01）. Besides, it also down-regulated the expression of Bcl-2 and Bcl-xL and activated caspase-3 and caspase-9. Conclusion： Ursolic acid induces apoptosis in HT-29 cells by suppressing the EGFR/MAPK pathway, suggesting that it may be a potent agent for the treatment of colorectal cancer.

Enhanced ozonation degradation of atrazine in the presence of nano-ZnO： Performance, kinetics and effects

Enhanced ozonation degradation of atrazine（ATZ） with nano-ZnO（nZnO） as catalyst and the influences of the operational parameters have been investigated through semi-continuous experiments in this study. The results demonstrated that the combination of ozone（O3） and nZnO showed an obvious synergetic effect and the ATZ degradation conformed to pseudo-first-order kinetics. An improvement of ATZ degradation efficiency by 41.8% and pseudo-first-order rate constant by more than a factor of four was obtained in the O3/nZnO process after 5 min of reaction compared to O3 alone. Meanwhile, the degradation efficiency of ATZ was gradually enhanced with increasing nZnO dosage and initial pH in the range from 3.0 to 8.0, and a higher amount of ATZ was degraded when the initial concentration of ATZ rose from 0.5 to 5 mg/L. Additionally, sulfate ion, chloride ion, nitrate ion and low concentrations of humic acid substances led to enhancement of the ATZ degradation. The notable decrease of ATZ removal efficiency observed in the presence of radical scavengers and the results of free radical tests indicated thatUOH is the dominant active radical species. The mechanism investigation demonstrated that the enhancement effect could be attributed to the introduction of nZnO,which could promote the utilization of O3, enhance the formation of superoxide radical, and further accelerate the production of hydrogen peroxide and the generation of OH/O2-.

Chlorinated organic pollutants(COPs)have caused serious contaminants in soil and groundwater,hence developing methods to remove these pollutants is necessary and *** a simple hydrothermal method,we synthesized the bimetallic iron-nickel sulfide(FeNiS)particles which exhibited excellent catalytic property of COPs *** was chosen as the peroxydisulfate(PDS)activator to removal COPs including 4-chlorophenol(4-CP),1,4-dichlorophenol(1,4-DCP)and 2,4,6-trichlorophenol(2,4,6-TCP).The results show that FeNiS can efficiently activate PDS to produce sulfate radical(SO4·-)which plays major role in the oxidative dechlorination and degradation due to its strong oxidizing property and the ability of producing hydroxyl radicals(·OH)in the alkaline ***,the Cl-abscised from COPs during the dechlorination can turn into the chlorine radicals and enhance the degradation and cause further mineralization of intermediate *** bimetallic FeNiS catalyst is a promising PDS activator for removal of chlorinated organics.

Objective: To explore the role of the abnormal expression of miRNAs in the development process of non-small cell lung cancer and the feasibility of ultrasound microbubble-mediated gene therapy after transfecting antisense miRNA-224 and miRNA-122 a plasmids into nonsmall cell lung cancer A549 cells. Methods: Antisense miRNA-224 and miRNA-122 a plasmids were transfected into non-small cell lung cancer A549 cells on the optimal ultrasound microbubble mediated condition. We set up a control group. The cell proliferation activity, apoptosis, invasion ability were detected by MTT assay, Annexin V-PE, Transwell invasion experiment and colony formation assay, respectively. Results: The expression of mi RNA-224 decreased and the expression of miRNA-122 a rose after the plasmids of target genes were transfected into non-small cell lung cancer A549 cells, and there were significant differences when compared with those of the control group(P<0.05). After the plasmids of target genes were transfected into A549 cells, the growth of antisense miRNA-224 and miRNA-122 a were inhibited, and the differences were significant as compared with the control group(P < 0.05). Besides, the inhibition of miRNA-122 a group was the most significant and there was statistically significant difference as compared with miRNA-224 group(t =-4.694, P = 0.009). After the plasmids of target genes were transfected into A549 cells, the proportion of apoptotic cells increased, the invasive cells were decreased and the clone ability reduced, and also there was a significant difference as compared with those of the control group(P < 0.05). What's more, the apoptotic peak appeared in miRNA-122 a group. Its invasion ability decreased most obviously(40.25 ± 3.97/visual field), the number of clone ability was 104.93 ± 4.87 and the inhibitory effect was the most obviously. Grapevine(Vitis vinifera L.)is an economically important fruit crop in the world,and China ranks first in the production of grapes with approximately 15%of the world’s total ***,diseases that cause the death of grapevine shoots pose a severe threat to the production of *** this study,the fungus Neopestalotiopsis eucalypti was identified as a causal pathogen of grapevine shoot rot based on the morphology of conidia and a phylogenetic *** phylogenetic analysis was performed with three isolates based on the combined sequence of internal transcribed spacer(ITS)region of ribosomal DNA,part of the translation elongation factor 1-alpha(Tef)and theβ-tubulin(Tub2)*** three isolates were all identified as *** tests of the three fungal isolates were conducted on grapevines shoots in vitro and in *** results showed that all three fungal isolates caused severe rot lesions on the inoculated grapevine shoots,and *** was re-isolated from the inoculated grapevine ***,*** was confirmed as a causal agent of the grapevine shoot *** is the first report of *** causing grapevine shoot disease in China.

人工免疫算法已被证明是用于入侵检测的一种有效技术手段.通过引入疫苗接种算子和小生境技术,设计了一种基于免疫进化算法的动态入侵检测模型,提出了一个在免疫入侵检测中实现疫苗提取和接种的方案.在疫苗接种算子的作用下,算法可以在下一代个体中保留父代的优良基因;小生境技术的应用,保证了疫苗提取和接种操作的可行性,并使得算法能更好地保持抗体的多样性,同时具有较高的收敛速度.最后通过模拟实验,验证了模型的可行性和有效性.

[目的]通过分析南亚热带林地利用方式变化对土壤团聚体组成及化学计量特征的影响,为人工林土壤有机碳(SOC)固持及其养分循环提供科学依据。[方法]以云南省永德县大雪山季风常绿阔叶林、茶园、澳洲坚果林和核桃林为研究对象,测定土壤表层(0~20 cm)团聚体不同粒径的质量百分含量、SOC、全氮(TN)和全磷(TP)含量以及土壤碳氮比(C∶N)、碳磷比(C∶P)和氮磷比(N∶P)等化学计量特征在不同林地利用方式的变化趋势。[结果](1)土壤团聚体的质量百分含量随粒径增大而显著增加。天然林转换为核桃林和澳洲坚果林,粒径为>2 mm和1~2 mm的土壤团聚体质量百分含量显著增加,而粒径为0.5~1 mm、0.25~0.5mm和<0.25 mm的土壤团聚体则下降。(2)林地利用方式下土壤团聚体SOC、TN和TP含量随粒径减小呈增大趋势。天然林转换为核桃林和澳洲坚果林,显著降低了不同粒径土壤团聚体SOC和TN含量,而对茶园无显著影响;核桃林土壤团聚体不同粒径的TP含量要显著高于其他林地类型。(3)天然林转为核桃林和澳洲坚果林后土壤团聚体的C∶N、C∶P和N∶P显著降低,茶园无显著变化。(4)土壤速效氮和含水量促进土壤团聚体SOC、TN、TP的积累,提高化学计量特征,而SD的作用则相反。[结论]天然林转换为人工林后,澳洲坚果和核桃林SOC、TN、TP及其化学计量特征受影响最大,而茶园则无显著影响,小粒径的土壤团聚体能固定更多的SOC、TN和TP。
