Comparative evaluation of SPE methods for biotoxicity assessment of water and wastewater:Linkage between chemical extracting efficiency and biotoxicity outcome

Biotoxicity assessment results of environmental waters largely depend on the sample extraction protocols that enrich pollutants to meet the effect-trigger thresholds of ***,more chemical mixture does not necessarily translate to higher combined ***,there is a need to establish the link between chemical extracting efficiency and biotoxicity outcome to standardize extraction methods for biotoxicity assessment of environmental *** study compares the performance of five different extraction phases in solid phase extraction(SPE),namely HLB,HLB+Coconut,C18 cartridge,C18 disk and Strata-X,and evaluated their chemical extracting efficiencies and biotoxicity *** quantitatively assessed cytotoxicity,acute toxicity,genotoxicity,estrogenic activity,and neurotoxicity of the extracts using in vitro bioassays and characterized the chemical extracting efficiencies of the SPE methods through chemical recoveries of 23 model compounds with different polarities and total organic *** Pareto ranking,we identified HLB+Coconut as the optimal SPE method,which exhibited the highest level of water sample biotoxicity and recovered the most chemicals in water *** found that the biotoxicity outcomes of the extracted water samples significantly and positively correlated with the chemical extracting efficiencies of the SPE ***,we observed synchronous changing patterns in biotoxicity outcome and chemical extracting efficiencies in response to increasing sample volumes per cartridge(SVPC)during *** findings underscore that higher chemical extracting efficiency of SPE corresponds to higher biotoxicity outcome of environmental water samples,providing a scientific basis for standardization of SPE methods for adequate assessment of biotoxicities of environmental waters.

A new scheme of climate-vegetation regionalization in the Hengduan Mountains Region

The Hengduan Mountains Region(HMR) is essential for the future ecological protection, clean energy production,Sichuan-Xizang and Yunnan-Xizang railways, and other major infrastructure projects in China. The distributions of climate and vegetation exhibit significant regional differentiation and vertical zonality due to the rugged longitudinal ranges and gorges and the complex disaster-prone environments in HMR. Therefore, it is urgent to develop the climate-vegetation regionalization in HMR to effectively satisfy the national requirements such as agricultural production and ecological protection, mountain disaster risk prevention, and major project construction. We here develop a new scheme of climate-vegetation regionalization with the latest demarcation outcome of HMR, the ground observation from 122 meteorological stations in HMR and its surrounding areas during 1990–2019, and the high-precision remote sensing data of land cover types. The new scheme first constructs the regionalization index system, fully considering the extraordinarily complicated geomorphic pattern of mountains and valleys, the scarcity of meteorological observations, and the remarkable differentiation of climate and vegetation in HMR. The system consists of three primary regionalization indices(i.e., days with daily average temperature steady above 10°C, aridity index, and main vegetation types, dividing the temperature zones, moisture regions, and vegetation subregions, respectively) and three auxiliary indices of the accumulated temperature above 10°C, and the temperatures in January and July. Then, the HMR is divided into five temperature zones, 20 moisture regions, and 55 vegetation subregions. Compared with previous regionalization schemes, the new scheme optimizes the climate spatial interpolation model of thin plate smoothing spline suitable for the unique terrain in HMR. Developing a new type of photocatalyst(PC) and catalytic mechanism for near-infrared(NIR) photocontrolled reversibledeactivation radical polymerization(RDRP) system is charming but ***,a novel PC of the persistent radical anion(PRA)(possessing the properties of both radical and anion) was developed for NIR photocontrolled reversible additionfragmentation chain transfer(RAFT) polymerization,enabling successful polymerization while gaining a deep insight into the mechanism of photo-induced electron transfer RAFT(PET-RAFT) *** from the conventional and wellaccepted reductive quenching(RQ) pathway,in which the radical anion intermediates of PCs(PCs^(·-)) must be generated in an excited state(ES),here,the PRA(3,4,9,10-perylenetetracarboxylic dianhydride radical anion(PTCDA^(·-))) could generate conveniently in situ in the ground state(GS) and subsequently serve as highly efficient PC in the NIR region(740–850 nm).The successful implementation of this strategy elucidates the peculiar role played by light and the real way of electron transfer *** fact,the transfer of a single electron from PRA to chain transfer agent(CTA) and cleavage of the C–S bonds is a process from ES to GS,rather than always from GS(PCs^(·-)) to GS(CTA) in the RQ pathway as is well known to *** addition,the excellent spatial-temporal control and powerful penetration ability of the NIR light were also confirmed by this PRAcatalyzed polymerization system.

Resolving the geometric structure of trastuzumab by mobility capillary electrophoresis and native mass spectrometry

Available online Immunoglobulins G(IgGs)are Y-shaped globular proteins,however,their high flexibility and heterogeneity pose great challenges to their structure and conformation *** structure of IgG closely correlates to its biofunctions,such as the antibody escape of human immunodeficiency virus(HIV)could attribute to the distance mismatch between the ends of two Fab arms(antigen-binding sites)and envelope glycoprotein spikes on virion ***,we report the first use of mobility capillary electrophoresis(MCE)and native mass spectrometry(nMS)to resolve the internal geometric structure and conformation of an IgG(trastuzumab)in solution *** proteolysis,the ellipsoid dimensions of IgG and its subunits were measured by MCE-nMS *** was then reconstructed,in which the sizes and relative positions of these three subunits in three-dimensional space were *** was found that the two Fab arms have an angle of~102.1°and a distance of~11.0 nm between the two antigen-binding sites under native condition,and the Fc arm was tilted~16.0°towards one of the Fab *** was not on the plane of Fab-Fab,but has an angle of no larger than 103.1°.Under acidic environment(pH 3.0),each subunit of the IgG would unfold into larger dimensions,and the angles between these subunits also *** great potential for tumor imaging and therapy,the structure of F(ab')_(2)fragments was also measured and validated by molecular dynamic *** was found that the electrostatic force among these three subunits and steric hindrance stemming from Fc help maintaining the angle between two Fab arms.

Development of a novel staging classification for Siewert Ⅱ adenocarcinoma of the esophagogastric junction after neoadjuvant chemotherapy

BACKGROUND Stage classification for Siewert Ⅱ adenocarcinoma of the esophagogastric junction(AEG)treated with neoadjuvant chemotherapy(NAC)has not been *** To investigate the optimal stage classification for Siewert Ⅱ AEG with *** A nomogram was established based on Cox regression model that analyzed variables associated with overall survival(OS)and disease-specific survival(DSS).The nomogram performance in terms of discrimination and calibration ability was evaluated using the likelihood-ratio test,Akaike information criterion,Harrell concordance index,time-receiver operating characteristic curve,and decision curve *** Data from 725 patients with Siewert type Ⅱ AEG who underwent neoadjuvant therapy and gastrectomy were obtained from the Surveillance,Epidemiology,and End Results *** and multivariate analyses revealed that sex,marital status,race,ypT stage,and ypN stage were independent prognostic factors of OS,whereas sex,race,ypT stage,and ypN stage were independent prognostic factors for *** factors were incorporated into the OS and DSS *** novel nomogram model performed better in terms of OS and DSS prediction compared to the 8th American Joint Committee of Cancer pathological staging system for esophageal and gastric ***,a user-friendly web application was developed for clinical *** The nomogram established specifically for patients with Siewert type Ⅱ AEG receiving NAC demonstrated good prognostic *** using external data is warranted before its widespread clinical application.

Dual-ion carrier storage through Mg^(2+) addition for high-energy and long-life zinc-ion hybrid capacitor

Cation additives can efficiently enhance the total electrochemical capabilities of zinc-ion hybrid capacitors (ZHCs).However their energy storage mechanisms in zinc-based systems are still under ***,we modulate the electrolyte and achieve dual-ion storage by adding magnesium *** we assemble several Zn//activated carbon devices with different electrolyte concentrations and investigate their electrochemical reaction dynamic *** zinc-ion capacitor with Mg^(2+)mixed solution delivers 82 mAh·g^(-1)capacity at 1 A·g^(-1) and maintains 91%of the original capacitance after 10000 *** is superior to the other assembled zinc-ion devices in single-component *** finding demonstrates that the double-ion storage mechanism enables the superior rate performance and long cycle lifetime of ZHCs.

Iron–doped bismuth oxybromides as visible-light-responsive Fenton catalysts for the degradation of atrazine in aqueous phases
