Molecular cloning of growth hormone receptor (GHR) from common carp(Cyprinus carpio L.)and identification of its two forms of mRNA transcripts

The cDNA of growth hormone receptor (GHR) was cloned from the liver of 2.year common carp (Cyprinus carpio L.) by reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA end (RACE). Its open reading frame (ORF) of 1806 nucleotides is translated into a putative peptide of 602.amino acids, including an extracellular ligand-binding domain of 2.4 amino acids (aa), a single transmembrane domain of 2. aa and an intracellular signal-transduction domain of 334 aa. Sequence analysis indicated that common carp GHR is highly homologous to goldfish (Carassius auratus) GHR at both gene and protein levels. Using a pair of gene-specific primers, a GHR fragment was amplified from the cDNA of 2.year common carp, a 2.4 bp product was identified in liver and a 32. bp product in other tissues. The sequencing of the products and the partial genomic DNA indicated that the difference in product size was the result of a 97 bp intron that alternatively spliced. In addition, the 32. bp fragment could be amplified from all the tissues of 4-month common carp including liver, demonstrating the occurrence of the alternative splicing of this intron during the development of common carp. Moreover, a semi-quantitative RT-PCR was performed to analyze the expression level of GHR in tissues of 2.year common carp and 4-month common carp. The result revealed that in the tissues of gill, thymus and brain, the expression level of GHR in 2.year common carp was significantly lower than that of 4-month common carp.

The deep thermal characteristic of continental margin of the northern South china Sea

Heat flow plays an important role in the study ot thermal structure and thermal evolutionof continental margin of the northern South china Sea. The analysis of heat flow value shows that margin heat flow in the northern South china Sea is relatively high setting, but the percentage of crustal heat flow is lower than 35% in terrestrial heat flow. The terrestrial heat flow exhibited a current of rise from the Northern Continental Margin to the Southern Central Basin. However, the proportion of crustal heat flow in terrestrial heat flow slowly dropped down in the same direction. It is suggested that the main factor causing high heat flow setting is the moving up of hot material from asthenosphere.

Impacts of the Wenchuan Earthquake on the Giant Panda Nature Reserves in china

The Wenchuan Earthquake that occurred in May of 2.08 caused damages to large areas of Sichuan,Gansu,and Shaanxi provinces in *** from local governments and related management agencies show that the giant panda nature reserves in the earthquake-hit areas were heavily *** estimates in this paper of the impacts of the earthquake on the giant panda in the earthquake-hit areas were made based on the interpretation of remote sensing images and information collected by field survey.A rapid assessment method was designed to estimate the damages of the earthquake on giant panda *** using visual interpretation methods,we decoded the remote sensing images of the disaster area in the 49 giant panda nature *** results showed that the Wenchuan Earthquake and the succeeding secondary geological disasters caused great damages to the giant panda nature reserves and disturbed the normal life of the giant pandas there (e.g.,landscape fragmentation increased significantly).Undoubtedly,the life of the giant pandas there was ***,although the earthquake caused certain impacts on the giant pandas,it did not really threat their *** so,we still strongly advocate for protection of the giant pandas,and have prioritized a couple of measures to be taken to restore the giant panda nature reserves in the earthquake-hit areas.

Interannual variability of Antarctic Oscillation and its influence on East Asian climate during boreal winter and spring

The interannual variability of Antarctic Oscillation (AAO) and its influence on East Asian climate during both boreal winter and spring are addressed. The results show that the positive AAO anomaly decreases the cold activity over East Asia during both boreal winter and spring. AAO-related barotropic meridional teleconnection from Antarctic to Arctic is found through analysis of mean me- ridional circulations. This meridional teleconnection is remarkable over Eurasia during boreal winter and over the Pacific Ocean during boreal spring. The results also show that zonal mean zonal wind at high latitudes in Southern Hemisphere has well positive correlation with that of Eurasia during boreal winter and has negative correlation with Pacific North American teleconnection (PNA) during boreal spring, which again display the meridional teleconnection. Thus, local meridional teleconnection is a possible linkage for interaction of circulations at mid-high latitudes between both hemispheres.

Cloning and characterization of interferon stimulated genes Viperin and ISG15,and their promoters from snakehead Channa argus

By suppression subtractive hybridization, rapid amplification of cDNA ends and gene walking methods, interferon stimulated genes (ISGs), Viperin and ISG15, and their promoters have been cloned and characterized from snakehead Channa argus. The Viperin cDNA was found to be 1474 nt and contain an open reading frame (ORF) of 1059 nt that translates into a putative peptide of 352.amino acid (aa). The putative peptide of Viperin shows high identity to that in teleosts and mammals except for the N-terminal 70 aa. The ISG15 cDNA was found to be 758 nt and contain an ORF of 468 nt that translates into a putative peptide of 155 aa. The putative peptide of ISG15 is composed of two tandem repeats of ubiquitin-like (UBL) domains, and a canonical conjugation motif (LRGG) at C-terminal. Viperin and ISG15 promoter regions were characterized by the presence of interferon stimulating response elements (ISRE) and γ-IFN activation sites (GAS). ISRE is a feature of IFN-induced gene promoter and partially overlaps interferon regulatory factor (IRF) 1 and IRF2.recognition sites. GAS is responsible for the γ-IFN mediated transcription. One conserved site for NF-κB was found in the promoter region of Viperin. This is the first report of conservative binding motif for NF-κB in accordance with the consensus sequence (GGGRNNYYCC) among teleost ISG promoters. Moreover, there were also TATA, CAAT and Sp1 transcription factor sites in Viperin and ISG15 promoters. In 5′ untranslated region (UTR), snakehead ISG15 gene contains a single intron, which differs from Viperin gene. The transcripts of Vipeirn and ISG15 mRNA were mainly expressed in head kidney, posterior kidney, spleen and gill. The expression levels in liver were found to increase obviously in response to induction by IFN-inducer poly I∶C.

Environmental Implication of Intensive Farming Practices on North china Plain

The current pressure on production resources of North china Plain, such as land and water to feed the growing population, necessitates the assessment of the sustainability of farming practices. This study focuses on the sustainability of farming practices related to groundwater and soil fertility management. The assessment is based on selected site-specific key indicators and their established threshold limits. The current farming practices in the study area are clearly unsustainable. Only about 6% of the surveyed farm households practice sustainable farming. The study stresses that farming practice, which is economically sustainable, should not be promoted at the cost of environment. Holistic strategies need to be developed and implemented that aim at balanced use of inputs, which satisfy both productivity and environmental concerns.

The study on Sanmenxia annual flow forecasting in the Yellow River with mix regression model

This paper established mix regression model for simulating annual flow, in which annual runoff is auto-regression factor, precipitation, air temperature and water consumption are regression factors; we adopted 9 hypothesis climate change schemes to forecast the change of annual flow of Sanmenxia Station. The results show : (1) When temperature is steady, the average annual runoff will increase by 8.3% if precipitation increases by 10%; when precipitation decreases by 10%, the average annual runoff will decrease by 8.2.; when precipitation is steady, the average annual runoff will decrease by 2.4% if temperature increases 1℃; if temperature decreases 1℃, runoff will increase by 1.2.. The mix regression model can well simulate annual runoff. (2. As to 9 different temperature and precipitation scenarios, scenario 9 is the most adverse to the runoff of Sanmenxia Station of Yellow River; i.e. temperature increases 1℃ and precipitation decreases by 10%. Under this condition, the simulated average annual runoff decreases by 10.8%. On the contrary, scenario 1 is the best to the enhancement of runoff; i.e. when temperature decreases 1 ℃ precipitation will increase by 10%, which will make the annual runoff of Sanmenxia increase by 10.6%.

Variations of δ^(18)O in precipitation from the Muztagata Glacier, East Pamirs

Expeditions to Muztagata (in the eastern Pamirs) during the summer seasons of2.02.and 2.03 collected precipitation samples and measured their oxygen isotopes. The δ^(18)O inprecipitation displays a wide range, varying from -17.40per thousand to +1.33per thousand inJune—September 2.02.and from -2..31per thousand to +4.59per thousand in May—August 2.03. Theδ^(18)O in precipitation correlates with the initial temperature of precipitation during theobserving periods. The positive correlation between δ^(18)O and temperature suggests that δ^(18)Ocan be used as an indicator of temperature in this region. The δ^(18)O values in fresh-snow samplescollected from two snow events at different elevations on the Muztagata Glacier show a strong''altitude effect'', with a ratio of nearly -0.40per thousand per 100 m from 5500 m to 7450 m.

Climatic and environmental changes over the last millennium recorded in the Malan ice core from the northern Tibetan Plateau

In this paper, climatic and environmental changes were reconstructed since 112.A.D. based on the Malan ice core from Hol Xil, the northern Tibetan Plateau. The record of δ 18O in the Malan ice core indicated that the warm-season air temperature variations displayed a general increase trend, the 2.th-century warming was within the range of natural climate variability, and the warmest century was the 17th century while the warmest decade was the 1610s, over the entire study period. The “Medieval Warm Epoch” and “Little Ice Age” were also reflected by the ice core record. The dust ratio in the Malan ice core is a good proxy for dust event frequency. The 870-year record of the dust ratio showed that dust events occurred much frequently in the 19th century. Comparing the variations of δ 18O and the dust ratio, it is found that there was a strong negative correlation between them on the time scales of 101―102.years. By analyses of all the climatic records of ice cores and tree rings from the northern Tibetan Plateau, it was revealed that dust events were more frequent in the cold and dry periods than in the warm and wet periods.

Indoor imitation experimental study on driving factors of rainfall-runoff process

The driving actions of rainfall-runoff process can be attributed to two aspects. The first is the influence of precipitation process, and the second is that of the ground pad. The re- search results of 179 indoor experiments conducted to imitate rainfall-runoff process indicate that both precipitation duration and intensity play important roles in affecting confluence lag time, which is obviously inconsistent with the traditional hypotheses. The nonlinear relationship is of great significance to the confluence curve especially when the precipitation duration is less than the total confluence time or the precipitation intensity is small. Therefore it can be concluded that the unit hydrograph (UH) can be applied to rainfall-runoff process imitation in the humid areas in the south china region. However, the UH application should be strictly modified in accordance with precipitation conditions in the arid and semiarid region of north china where the precipitation duration is short and the intensity is unstable. It will be hard to get ideal imitation results if the UH is applied blindly without considering specific conditions in the north china region. This also explains the unsatisfactory imitation results caused by using various hydrological models in the north china region. When the precipitation duration is short, and the watershed has not reached total watershed concentration, the characteristics of confluence change greatly, which reflects the actual situation in the north china region. Therefore necessary nonlinear corrections should be made when UH is applied. If the duration is longer than the total confluence time and the balance between pondage and discharge is stricken, the imitation research results will be applicable to both rainfall-runoff relation with longer duration in the south china region and the basic theoreti- cal research on runoff generation and concentration.
