Proteomic analysis of trans-hemispheric motor cortex reorganization following contralateral C7 nerve transfer

Nerve transfer is the most common treatment for total brachial plexus avulsion injury. After nerve transfer, the movement of the injured limb may be activated by certain movements of the healthy limb at the early stage of recovery, i.e., trans-hemispheric reorganization. Pre- vious studies have focused on functional magnetic resonance imaging and changes in brain-derived neurotrophic factor and growth asso- ciated protein 43, but there have been no proteomics studies. In this study, we designed a rat model of total brachial plexus avulsion injury involving contralateral C7 nerve transfer. Isobaric tags for relative and absolute quantitation and western blot assay were then used to screen differentially expressed proteins in bilateral motor cortices. We found that most differentially expressed proteins in both cortices of upper limb were associated with nervous system development and function （including neuron differentiation and development, axonogenesis, and guidance）, microtubule and cytoskeleton organization, synapse plasticity, and transmission of nerve impulses. Two key differentially expressed proteins, neurofilament light （NFL） and Thy-1, were identified. In contralateral cortex, the NFL level was upregulated 2 weeks after transfer and downregulated at 1 and 5 months. The Thy-1 level was upregulated from 1 to 5 months. In the affected cortex, the NFL level increased gradually from 1 to 5 months. Western blot results of key differentially expressed proteins were consistent with the proteom- ic findings. These results indicate that NFL and Thy-1 play an important role in trans-hemispheric organization following total brachial plexus root avulsion and contralateral C7 nerve transfer.

Ziyin Huatan Recipe, a Chinese herbal compound, inhibits migration and invasion of gastric cancer by upregulating RUNX3 expression

Objectives: Ziyin Huatan Recipe(ZYHT), a traditional Chinese medicine comprised of Lilii Bulbus, Pinelliae Rhizoma, and Hedyotis Diffusa, has shown promise in treating gastric cancer(GC). However, its potential mechanism has not yet been clearly addressed. This study aimed to predict targets and molecular mechanisms of ZYHT in treating GC by network pharmacology analysis and to explore the role of ZYHT in GC both in vitro and in vivo.Methods: Targets and molecular mechanisms of ZYHT were predicted via network pharmacology analysis. The effects of ZYHT on the expression of metastasis-associated targets were further validated by Western blot and quantitative real-time polymerase chain reaction. To explore the specific molecular mechanisms of the effects of ZYHT on migration and invasion, the runt-related transcription factor 3(RUNX3) gene was knocked out by clustered regularly interspaced short palindromic repeats/Cas9, and lentiviral vectors were transfected into SGC-7901 cells. Then lung metastasis model of GC in nude mice was established to explore the anti-metastasis effect of ZYHT. Western blot and immunohistochemistry were used to explore the impact of ZYHT on the expression of metastasis-related proteins with or without RUNX3 gene.Results: The network pharmacology analysis showed that ZYHT might inhibit focal adhesion, migration,invasion and metastasis of GC. ZYHT inhibited the proliferation, migration and invasion of GC cells in vitro via regulating the expression of metastasis-associated targets. Knocking out RUNX3 almost completely reversed the cell phenotypes(migration and invasion) and protein expression levels elicited by ZYHT.In vivo studies showed that ZYHT inhibited the metastasis of GC cells to the lung and prolonged the survival time of the nude mice. Macrophages(MΦ) differe ntiate from blood monocytes and participate in innate and adaptive immunity.Because of their abilities to recognize pathogens and activate bactericidal activities,MΦ are always discovered at the site of immune defense.MΦ in the intestine are unique,such that in the healthy intestine,they possess complex mechanisms to protect the gut from inflammation.In these complex mechanisms,they produce anti-inflammatory cytokines,such as interleukin-10 and transforming growth factor-β,and inhibit the inflammatory pathways mediated by Toll-like receptors.It has been demonstrated that resident MΦ play a crucial role in maintaining intestinal homeostasis,and they can be recognized by their unique markers.Nonetheless,in the inflamed intestine,the function of MΦ will change because of environmental variation,which may be one of the mechanisms of inflammatory bowel disease(IBD).We provide further explanation about these mechanisms in our review.In addition,we review recent discoveries that MΦ may be involved in the development of gastrointestinal tumors.We will highlight the possible therapeutic targets for the management of IBD and gastrointestinal tumors,and we also discuss why more details are needed to fully understand all other effects of intestinal MΦ.

Effects of Continuous Positive Airway Pressure on Cognitive Deficits in Middle-aged Patients with Obstructive Sleep Apnea Syndrome： A Meta-analysis of Randomized Controlled Trials

Background： Current views on continuous positive airway pressure （CPAP） treatment to improve the cognitive deficits of patients with obstructive sleep apnea syndrome （OSAS） are controversial, so we performed a meta-analysis. Methods： A comprehensive literature search was undertaken in PubMed, CINAHL, Medline, Psyclnfo, EMBASE, Cochrane Library, CNKI, WanFang, VIP, and CBMdisc for studies published from June 1971 to July 2014. The outcome measures included neuropsychological tests of the 7 cognitive domains detailed below. Results： After screening the titles and abstracts and thoroughly reading the fuU text, we obtained 13 studies with little risk of bias that incorporated 1744 middle-aged obese participants with mild to severe OSAS. The studies were published from 1994 to 2012. Treatment durations varied from 1 to 24 weeks. The effect sizes of attention, vigilance, processing speed, working memory, memory, verbal fluency, and visuoconstructive skills domains were -0.10 （P = 0.24）, -0.12 （P = 0.04）, -0.08 （P = 0.16）, 0.00 （P = 0.95）, -0.04 （P = 0.30）, -0.06 （P = 0.34）, and -0.01 （P = 0.92）, respectively. Conclusions： Cognition partially improved in patients with OSAS after CPAP treatment. The only domain with significant improvement was vigilance. Rigorous randomized controlled trials need to be performed to obtain clear results.

Microscale endometrial sampling biopsy in detecting endometrial cancer and atypical hyperplasia in a population of 1551 women:a comparative study with hysteroscopic endometrial biopsy

Background:Endometrial cancer is one of the most common malignancies of the reproductive system.Effective and cost-effective screening method for populations at high risk is not available.This study aimed to investigate specimen adequacy and the influencing factors in microscale endometrial sampling biopsy and to evaluate the diagnostic accuracy and medical cost of biopsy in endometrial cancer and atypical hyperplasia screenings in comparison with hysteroscopic endometrial biopsy.Methods:A total of 1551 patients at high risk for endometrial lesions who required hysteroscopic endometrial biopsy from November 2017 to August 2018 were included.Microscale endometrial sampling biopsy was performed,followed by hysteroscopic endometrial biopsy.We evaluated the specimen adequacy and influencing factors of microscale endometrial sampling.Diagnostic consistency between microscale endometrial sampling biopsy and hysteroscopic endometrial biopsy was evaluated.The sensitivity,specificity,positive predictive value,and negative predictive value of microscale endometrial sampling biopsy in screening for endometrial cancer and atypical hyperplasia were analyzed,and the medical costs of the two procedures were compared.Results:The specimen adequacy was 81.2%.Patient age,menopausal status,endometrial thickness,and endometrial lesion type were correlated with specimen adequacy.There was good consistency in distinguishing benign and malignant endometrial diseases between microscale endometrial sampling biopsy and hysteroscopic biopsy(kappa 0.950,95%CI 0.925-0.975).The sensitivity,specificity,positive predictive value,and negative predictive value of microscale endometrial sampling biopsy were 91.7%,100.0%,100.0%,and 99.3%for endometrial cancer screening,respectively,and 82.0%,100.0%,100.0%,and 99.4%for atypical hyperplasia screening.The medical cost of endometrial sampling biopsy was only 22.1%of the cost of hysteroscopic biopsy.Conclusions:Microscale endometrial sampling biopsy is a minimally invasive alternative technique for obtai

The process of plane strain compression of 06Cr19Ni9NbN steel was carried out in the temperature range of 1000-1200℃ and the reduction ratio range of 10-50%.Combining the finite element numerical simulation,a new constitutive model of thermal deformation was established, which provides the theoretical basis to optimize the plarie strain compression process of the steel.The temperature and grain size at different regions were achieved by experiment and simulation,respectively.According to the results,the mathematical models of stress and temperature during the plane strain compression were established by mathematical analysis method.The new temperature models were established in three regions,respectively,and the stress models took account of the variation of temperature and'st^rain rate.Finally,by comparing the results of calculation,numerical simulation and experiment,the accuracy and validity of these mathematical models were verified.

Effect of spatial spectrum overlap on Fourier ptychographic microscopy

Fourier ptychographic microscopy(FPM)is a newly developed imaging technique which stands out by virtue of its high-resolution and wide FOV.It improves a microscope's imaging perfor-mance beyond the diffraction limit of the employed optical components by illuminating the sample with oblique waves of different incident angles,similar to the concept of synthetic aperture.We propose to use an objective lens with high-NA to generate oblique illuminating waves in FPM.We demonstrate utilizing an objective lens with higher NA to iluminate the sample leads to better resolution by simulations,in which a resolution of 0.28 pum is achieved by using a high-NA illuminating objective lens(NA=1.49)and a low-NA collecting objective lens(NA=0.2)in coherent imaging(λ=488 nm).We then deeply study FPM's exact relevance of convergence speed to spatial spectrum overlap in frequency domain.The simulation results show that an overlap of about 60%is the optimal choice to acquire a high-quality recovery(520*520 pixels)with about 2 min's computing time.In addition,we testify the robustness of the algorithm of FPM to additive noises and its suitability for phase objects,which further proves FPM's potential application in biomedical imaging.

Effect of Optimized Concentrations of Basic Fibroblast Growth Factor and Epidermal Growth Factor on Proliferation of Fibroblasts and Expression of Collagen： Related to Pelvic Floor Tissue Regeneration

Background： Fibroblasts were the main seed cells in the studies of tissue engineering of the pelvic floor ligament. Basic fibroblast growth factor （bFGF） and epidermal growth factor （EGF） were widely studied but at various concentrations. This study aimed to optimize the concentrations of combined bFGF and EGF by evaluating their effects on proliferation and collagen secretion of fibroblasts. Methods： Fibroblasts were differentiated from rat adipose mesenchymal stem cells （ADSCs）. Flow cytometry and immunohistochemistry were used for cell identification. The growth factors were applied at concentrations of 0, 1, 10, and 100 ng/ml as three groups：（1） bFGF alone, （2） EGF alone, and （3） bFGF mixed with EGF. Cell proliferation was evaluated by Cell Counting Kit-8 assays. Expression of Type I and III collagen （Col-I and Col-1II） mRNAs was evaluated by real-time quantitative reverse transcription-polymerase chain reaction. Statistical analysis was performed with SPSS software and GraphPad Prism using one-way analysis of variance and multiple t-test. Results： ADSCs were successfully isolated from rat adipose tissue as identified by expression of typical surface markers CD29, CD44, CD90, and CD45 in flow cytometry. Fibroblasts induced from ADSC, compared with ADSCs, were with higher mRNA expression levels of Col I and Col III （F = 1.29, P = 0.0390）. bFGF, EGF, and the mixture ofbFGF with EGF can enhanced fibroblasts proliferation, and the concentration of 10 ng/ml of the mixture ofbFGF with EGF displayed most effectively （all P 〈 0.05）. The expression levels of Col-I and Col-Ill mRNAs in fibroblasts displayed significant increases in the 10 ng/ml bFGF combined with EGF group （all P 〈 0.05）. Conclusions： The optimal concentration of both bFGF and EGF to promote cell proliferation and collagen expression in fibroblasts was 10 ng/ml at which fibroblasts grew faster and secreted more Type I and III collagens into the extracellular matrix, which might contribute to the stability of the pelvic floor microenvironment.

Efficient and stable expression of foreign genes in cells and transgenic animals is important for gain-of-function studies and the establishment of bioreactors.Safe harbor loci in the animal genome enable consistent overexpression of foreign genes,without side effects.However,relatively few safe harbor loci are available in pigs,a fact which has impeded the development of multi-transgenic pig research.We report a strategy for efficient transgene knock-in in the endogenous collagen type I alpha 1 chain(COL1A1)gene using the clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9(CRISPR/Cas9)system.After the knock-in of a 2A peptide-green fluorescence protein(2A-GFP)transgene in the last codon of COL1A1 in multiple porcine cells,including porcine kidney epithelial(PK15),porcine embryonic fibroblast(PEF)and porcine intestinal epithelial(IPI-2I)cells,quantitative PCR(qPCR),Western blotting,RNA-seq and CCK8 assay were performed to assess the safety of COL1A1 locus.The qPCR results showed that the GFP knock-in had no effect(P=0.29,P=0.66 and P=0.20 for PK15,PEF and IPI-2I cells,respectively)on the mRNA expression of COL1A1 gene.Similarly,no significant differences(P=0.64,P=0.48 and P=0.80 for PK15,PEF and IPI-2I cells,respectively)were found between the GFP knock-in and wild type cells by Western blotting.RNA-seq results revealed that the transcriptome of GFP knock-in PEF cells had a significant positive correlation(P<2.2e–16)with that of the wild type cells,indicating that the GFP knock-in did not alter the global expression of endogenous genes.Furthermore,the CCK8 assay showed that the GFP knock-in events had no adverse effects(P_(24)h=0.31,P_(48)h=0.96,P_(72)h=0.24,P_(96)h=0.17,and P_(120)h=0.38)on cell proliferation of PK15 cells.These results indicate that the COL1A1 locus can be used as a safe harbor for foreign genes knock-in into the pig genome and can be broadly applied to farm animal breeding and biomedical model establishment.

Chitooligosaccharide-zinc(COS·Zn)is a powerful anti-oxidant and anti-aging scavenger,whose anti-oxidative ability immensely exceeds vitamin C.Therefore,this study was aimed to investigate the protective effects of COS·Zn against premature ovarian failure(POF)and potential mechanisms.Female KM adult mice were divided into the following groups:a treatment group(150 mg·kg^(−1)·d^(−1) COS·Zn),a treatment group(300 mg·kg^(−1)·d^(−1) COS·Zn),a prevention group,two control groups and two CY/BUS groups.COS·Zn(150,300 mg·kg^(−1)·d^(−1))and COS·Zn(300 mg·kg^(−1)·d^(−1))were therapeutically and preventatively administered to POF mice in the treatment and prevention studies,respectively.All the groups were administered for 21 days.Fewer primary and secondary follicles were observed in the COS·Zn-treated groups(including the treatment and prevention groups)than those of the control groups.Meanwhile,the ovarian index and the levels of FSH and LH notably increased in the treatment and prevention groups compared with those in the CY/BUS group.The levels of MVH,OCT4 and PCNA in the treatment group(300·kg^(−1)·d^(−1) COS·Zn)and MVH in the prevention group remarkably increased compared with those in the CY/BUS groups.Meanwhile,the levels of P53 and P16 protein were down-regulated in the treatment and prevention groups compared with those in the CY/BUS groups.Additionally,the amounts of Sestrin2(SESN2)and SOD2 protein were obviously higher in the treatment group(150 mg·kg^(−1)·d^(−1) COS·Zn)than those in the CY/BUS groups.Similarly,the amounts of NRF2 and SESN2 protein were up-regulated in the prevention group.Besides,an increased GSH level was observed in the two treatment groups,compared with that in the CY/BUS groups,and the same trend was also present in the prevention group.Taken together,COS·Zn improves the ovarian and follicular development through regulating the SESN2/NRF2 signaling pathway.These results suggest the role of COS·Zn as a novel agent for the treatment and prevention of POF.
