The peroxisomal matrix proteins involved in many important biological metabolism pathways in eukaryotic cells are encoded by nucleal genes, synthesized in the cytoplasm and then transported into the organelles. Targeting and import of these proteins depend on their two peroxisomal targeting signals （PTS 1 and PTS2） in sequence as we have known so far. The vectors of the fluorescent fusions with PTS, i.e., green fluorescence protein （GFP）-PTS1, GFP-PTS2 and red fluorescence protein （RFP）-PTS1, were constructed and introduced into Magnaporthe oryzae Guy ll cells. Transformants containing these fusions emitted fluorescence in a punctate pattern, and the locations of the red and green fluorescence overlapped exactly in RFP-PTS 1 and GFP-PTS2 co-transformed strains. These data indicated that both PTS1 and PTS2 fusions were imported into peroxisomes. A probable higher efficiency of PTS1 machinery was revealed by comparing the fluorescence backgrotmds in GFP-PTS1 and GFP-PTS2 transformants. By introducing both RFP-PTS1 and GFP-PTS2 into Amgpex6 mutants, the involvement of MGPEX6 gene in both PTS1 and PTS2 pathways was proved. In addition, using these transformants, the inducement ofperoxisomes and the dynamic of peroxisomal number during the pre-penetration processes were investigated as well. In summary, by the localization and co-localization of PTS1 and PTS2, we provided a useful tool to evaluate the biological roles of the peroxisomes and the related genes.

Background Biliary atresia(BA)is a rare fatal liver disease in children,and the aim of this study was to develop a method to diagnose BA *** We determined serum levels of matrix metalloproteinase-7(MMP-7),the results of 13 liver tests,and the levels of 20 bile acids,and integrated computational models were constructed to diagnose *** Our findings demonstrated that MMP-7 expression levels,as well as the results of four liver tests and levels of ten bile acids,were significantly different between 86 BA and 59 non-BA patients(P<0.05).The computational prediction model revealed that MMP-7 levels alone had a higher predictive accuracy[area under the receiver operating characteristic curve(AUC)=0.966,95%confidence interval(CI):0.942,0.989]than liver test results and bile acid *** AUC was 0.890(95%CI 0.837,0.943)for liver test results and 0.825(95%CI 0.758,0.892)for bile acid ***,bile levels had a higher contribution to enhancing the predictive accuracy of MMP-7 levels(AUC=0.976,95%CI 0.953,1.000)than liver test *** AUC was 0.983(95%CI 0.962,1.000)for MMP-7 levels combined with liver test results and bile acid *** addition,we found that MMP-7 levels were highly correlated with gamma-glutamyl transferase levels and the liver fibrosis *** The innovative integrated models based on a large number of indicators provide a noninvasive and cost-effective approach for accurately diagnosing BA in children.

Gravity assistance is a critical factor influencing CO_(2)-Oil mixing and miscible flow during EOR and CO_(2)geological *** on the Navier-Stokes equation,component mass conservation equation,and fluid property-composition relationship,a mathematical model for pore-scale CO_(2) injection in oilsaturated porous media was developed in this *** model can reflect the effects of gravity assistance,component diffusion,fluid density variation,and velocity change on EOR and CO_(2) *** nonhomogeneous porous media,the gravity influence and large density difference help to minimize the velocity difference between the main flow path and the surrounding area,thus improving the oil recovery and CO_(2) *** CO_(2) injection angles and oil-CO_(2) density differences can increase the oil recovery by 22.6% and 4.2%,respectively,and increase CO_(2) storage by 37.9% and 4.7%,*** diffusion facilitates the transportation of the oil components from the low-velocity region to the main flow path,thereby reducing the oil/CO_(2) concentration difference within the porous *** diffusion can increase oil recovery and CO_(2) storage by 5.7% and 6.9%,*** addition,combined with the component diffusion,a low CO_(2) injection rate creates a more uniform spatial distribution of the oil/CO_(2) component,resulting in increases of 9.5% oil recovery and 15.7% CO_(2) storage,*** study provides theoretical support for improving the geological CO_(2) storage and EOR processes.

Background and purpose Recent observational studies have reported that serum total homocysteine(tHcy)is associated with intracranial aneurysms(IAs).However,the causal effect of tHcy on IAs is *** leveraged large-scale genetic association and real-world data to investigate the causal effect of tHcy on IA *** We performed a two-sample Mendelian randomisation(MR)using publicly available genome-wide association studies summary statistics to investigate the causal relationship between tHcy and IAs,following the recommendations of the Strengthening the Reporting of Observational Studies in Epidemiology-MR ***,a propensity score matching(PSM)analysis was conducted to evaluate the detailed effects of tHcy on risk of IA formation by utilizing real-world multicentre data,including 9902 patients with and without IAs(1:1 matched).Further interaction and subgroup analyses were performed to elucidate how tHcy affects risk of IA *** MR analyses indicated that genetically determined tHcy was causally associated with IA risk(OR,1.38,95%CI 1.07 to 1.79;p=0.018).This is consistent with the more conservative weighted median analysis(OR,1.41,95%CI 1.03 to 1.93;p=0.039).Further sensitivity analyses showed no evidence of horizontal pleiotropy or heterogeneity of single nucleotide polymorphisms in causal *** to the PSM study,we found that,compared with low tHcy(≤15μmol/L),moderate tHcy(>15-30μmol/L)(OR 2.13,95%CI 1.93 to 2.36)and high tHcy(>30μmol/L)(OR 3.66,95%CI 2.71 to 4.95)were associated with a higher IA risk(p trenddue to interaction=1.65,95%CI 1.29 to 2.01).Conclusion Both large-scale genetic evidence and multicentre real-world data support a causal as

For sperm cryopreservation,the conventional method,which requires glycerol,has been used for a long *** addition,the permeable cryoprotectant-free vitrification method has been continuously *** the differences of cryopreservation effects between the two methods have being studied,differences in microRNA(miRNA)profiles between them remain *** this study,we investigated the differences in miRNA expression profiles among conventional freezing sperm,droplet vitrification freezing sperm and fresh human *** also analyzed the differences between these methods in terms of differentially expressed miRNAs(DEmiRs)related to early embryonic development and paternal *** results showed no significant differences between the cryopreservation methods in terms of sperm motility ratio,plasma membrane integrity,DNA integrity,mitochondrial membrane potential,acrosome integrity,and ultrastructural ***,sperm miRNA-sequencing showed differences between the two methods in terms of the numbers of DEmiRs(28 and 19 with vitrification using a nonpermeable cryoprotectant and the conventional method,respectively)in postthaw and fresh sperm *** related to early embryonic development and paternal epigenetics mainly included common DEmiRs between the *** results showed that the differences between conventional freezing and droplet vitrification were minimal in terms of miRNA expression related to embryonic development and *** in sperm miRNA expression due to freezing are not always detrimental to embryonic *** study compared differences in miRNA expression profiles before and after cryopreservation between cryopreservation by conventional and vitrification *** offers a new perspective to evaluate various methods of sperm cryopreservation.

BACKGROUND Cell division cyclin 25C(CDC25C)is a protein that plays a critical role in the cell cycle,specifically in the transition from the G2 phase to the M *** research has shown that CDC25C could be a potential therapeutic target for cancers,particularly for hepatocellular carcinoma(HCC).However,the specific regulatory mechanisms underlying the role of CDC25C in HCC tumorigenesis and development remain incompletely *** To explore the impact of CDC25C on cell proliferation and apoptosis,as well as its regulatory mechanisms in HCC *** Hepa1-6 and B16 cells were transduced with a lentiviral vector containing shRNA interference sequences(LV-CDC25C shRNA)to knock down ***,a xenograft mouse model was established by subcutaneously injecting transduced Hepa1-6 cells into C57BL/6 mice to assess the effects of CDC25C knockdown on HCC development in *** proliferation and migration were evaluated using a Cell Counting Kit-8 cell proliferation assays and wound healing assays,*** expression of endoplasmic reticulum(ER)stress-related molecules(glucose-regulated protein 78,X-box binding protein-1,and C/EBP homologous protein)was measured in both cells and subcutaneous xenografts using quantitative real-time PCR(qRT-PCR)and western ***,apoptosis was investigated using flow cytometry,qRT-PCR,and western *** CDC25C was stably suppressed in Hepa1-6 and B16 cells through LV-CDC25C shRNA transduction.A xenograft model with CDC25C knockdown was successfully established and that downregulation of CDC25C expression significantly inhibited HCC growth in ***25C knockdown not only inhibited cell proliferation and migration but also significantly increased the ER stress response,ultimately promoting ER stress-induced apoptosis in HCC *** The regulatory mechanism of CDC25C in HCC development may involve the activation of ER stress and the ER stress-induced apoptosis signali
