Comparison of Cold Tolerance in Three Ground-cover Chrysanthemum Cultivars during Wintering in the Open Field

[Objectives]Among many chrysanthemum types,the ground-cover chrysanthemum cultivar group has the characteristics of dwarf plant,long flowering period,rich color and strong resistance to stress.This study aimed to screen out cultivars with strong cold tolerance in order that they can be used for planting in urban green space and economic forests in northern cold areas,so as to enrich the plant community structures of landscape in cold regions.[Methods]Three ground-cover chrysanthemum cultivars,i.e.,‘Meiaihuang’,‘xinhong’and‘Zixunzhang’,were selected as the experimental materials.During the wintering period in the field in Beijing,they were observed and measured for the changes of the morphological manifestations and the physiological indices,and the relationship between the indices and low temperature was analyzed.The cold tolerance of the three ground-cover chrysanthemum cultivars was also comprehensively evaluated.[Results]During the open-field overwintering period,the root activity of the three ground-cover chrysanthemum cultivars increased slowly with cooling,decreased at the freezing low temperature,and then increased again in early spring.With natural cooling,the membrane injury percentages(MIPs)of the leaves and foot shoots increased significantly,and the contents of malondialdehyde(MDA)increased;the contents of soluble sugar(SS)and proline(Pro)gradually accumulated,and the contents of SS and Pro in foot shoots increased significantly in late winter;and the superoxide dismutase(SOD)activity showed a similar trend of rising slowly at first and then increasing significantly.There were correlations between MIPs in the leaves and foot shoots of the three cultivars and other physiological indices:a significant negative correlation with MDA content,and a significant positive correlation with SS content,Pro content and SOD activity.[Conclusions]Combining the morphological manifestations and physiological indices,the cold tolerance of the three ground-cover chrysanthemum cultivars was evaluated by the subordinate f

MG53 is an essential component of the cell membrane repair machinery,participating in the healing of dermal wounds.Here we develop a novel delivery system using recombinant human MG53(rhMG53)protein and a reactive oxygen species(ROS)-scavenging gel to treat diabetic wounds.Mice with ablation of MG53 display defective hair follicle structure,and topical application of rhMG53 can promote hair growth in the mg53􀀀/􀀀mice.Cell lineage tracing studies reveal a physiological function of MG53 in modulating the proliferation of hair follicle stem cells(HFSCs).We find that rhMG53 protects HFSCs from oxidative stress-induced apoptosis and stimulates differentiation of HSFCs into keratinocytes.The cytoprotective function of MG53 is mediated by STATs and MAPK signaling in HFSCs.The thermosensitive ROS-scavenging gel encapsulated with rhMG53 allows for sustained release of rhMG53 and promotes healing of chronic cutaneous wounds and hair follicle development in the db/db mice.These findings support the potential therapeutic value of using rhMG53 in combination with ROS-scavenging gel to treat diabetic wounds.

Peripheral nerve injury not only affects the site of the injury, but can also induce neuronal apop- tosis at the spinal cord. However, many acupuncture clinicians still focus only on the injury site, selecting acupoints entirely along the injured nerve trunk and neglecting other regions; this may delay onset of treatment efficacy and rehabilitation. Therefore, in the present study, we compared the clinical efficacy of acupuncture at Governor vessel and local meridian acupoints combined （GV/LM group） with acupuncture at local meridian acupoints alone （LM group） in the treatment of patients with peripheral nerve injury. In the GV/LM group （n = 15）, in addition to meridian acupoints at the injury site, the following acupoints on the Governor vessel were stimulated： Baihui （GV20）, Fengfu （GV16）, Dazhui （GV14）, and Shenzhu （GV12）, selected to treat nerve injury of the upper limb, and Jizhong （GV6）, Mingmen （GV4）, Yaoyangguan （GV3）, and Yaoshu （GV2） to treat nerve injury of the lower limb. In the LM group （n = 15）, only me- ridian acupoints along the injured nerve were selected. Both groups had electroacupuncture treatment for 30 minutes, once a day, 5 times per week, for 6 weeks. Two cases dropped out of the LM group. A good or excellent clinical response was obtained in 80% of the patients in the GV/ LM group and 38.5% of the LM group. In a second study, an additional 20 patients underwent acupuncture with the same prescription as the GV/LM group. Electomyographic nerve conduc- tion tests were performed before and after acupuncture to explore the mechanism of action of the treatment. An effective response was observed in 80.0% of the patients, with greater motor nerve conduction velocity and amplitude after treatment, indicating that electroacupuncture on specific Governor vessel acupoints promotes functional motor nerve repair after peripheral nerve injury. AIM:To demonstrate the apoptosis-inducing effect of iidocalne on human corneal stromal(HCS)cells fn vitm,and provide experimental basis for safety anesthetic usage In clinic of ophthalmology.METHODS:In vitro cultured HCS cells were treated with lidocaine at different doses and times,and their morphology was monitored successively with inverted phase contrast microscopy.The membrane permeability of them was detected by acridine orange/ethidium bromide(AO/EB)double staining.The DNA fragmentation of them was examined by agarose gel electrophoresis,and their ultrastructure was observed by transmission electron microscopy(TEM),respectively.RESULTS:Exposure to lidocaine at doses from0.3125g/L to 20g/L induced morphological changes of HCS cells such as cytoplasmic vacuolation,cellular shrinkage,and turning round,and elevated membrane permeability of these cells in AO/EB staining.The change of morphology and membrane permeability was doseand time-dependent,while lidocaine at dose below0.15625g/L could not induce these changes.Furthermore,lidocaine induced DNA fragmentation and ultrastructural changes such as cytoplasmic vacuolation,structural disorganization,chromatin condensation,and apoptotic body appearance of the cells.CONCLUSION:Lidocaine has significant cytotoxicity on human corneal stromal cells in vitro in a dose-and time-dependent manner by inducing apoptosis of these cells.The established experimental model and findingsbased on this model here help provide new insight into the apoptosis-inducing effect of local anesthetics in eye clinic.

为探究裂隙网络含自由面的稳定渗流特性,设计可视化的二维裂隙网络渗流试验装置,通过调节裂隙网络模型的上、下游水头开展一系列裂隙网络渗流试验,依次记录自由面水头高度和渗流流量大小;同时基于有限元软件COMSOL Multiphysics中的Darcy接口,建立等比例二维裂隙网络数值模型,复现试验工况并与试验结果对比分析。对比发现试验结果与模拟结果基本吻合,这验证了试验结果的有效性以及数值模拟的准确性。研究结果表明:影响自由面变化的主要因素为上、下游水头高度及二者高度差:渗流水力梯度越大,自由面位置的下降越快;渗流水力梯度相同的水力条件,上、下游水头越高,形成的自由面水头越高,自由面降落曲线也越陡。其中,若上、下游水头高度不处于近似的水平,二者对于自由面位置的影响存在较大差异。自由面变化的基本规律符合Dupuit公式的表述。稳定渗流的水力梯度与平均流量仅在流量较小时符合线性相关,随着水力梯度的增加,二者关系呈现明显的非线性。

Molecular knowledge of human gastric corpus epithelium remains incomplete.Here,by integrated analyses using single-cell RNA sequencing(scRNA-seq),spatial transcriptomics,and single-cell assay for transposase accessible chromatin sequencing(scATAC-seq)techniques,we uncovered the spatially resolved expression landscape and gene-regulatory network of human gastric corpus epithelium.Specifically,we identified a stem/progenitor cell population in the isthmus of human gastric corpus,where EGF and WNT signaling pathways were activated.Meanwhile,LGR4,but not LGR5,was responsible for the activation of WNT signaling pathway.Importantly,FABP5 and NME1 were identified and validated as crucial for both normal gastric stem/progenitor cells and gastric cancer cells.Finally,we explored the epigenetic regulation of critical genes for gastric corpus epithelium at chromatin state level,and identified several important cell-type-specific transcription factors.In summary,our work provides novel insights to systematically understand the cellular diversity and homeostasis of human gastric corpus epithelium in vivo.

An action-based Markov chain modeling approach for predicting the window operating behavior in office spaces

Reliable energy and performance prediction for building design and planning is important for newly-designed or retrofitted buildings.Window operating behavior has an important influence on the ventilation and energy consumption of these buildings under different realistic scenarios.Therefore,quantitatively describing this behavior and constructing a prediction model are important.In this work,an action-based Markov chain modeling approach for predicting window operating behavior in office spaces was proposed.Two summer measurement data(2016 and 2018)were used to verify the accuracy and validity of the modeling approach.The opening rate,outdoor temperature,time distribution,and on-off curve were proposed as four inspection standards.This study also compared the prediction performance between the action-based Markov chain modeling approach with the state-based Markov chain modeling approach,which is the most popular modeling approach to model occupant window operating behavior.This study proved that the yearly variation of occupants’behavior performed a form of action that remained unchanged during a certain period.Meanwhile,the results also proved that the action-based Markov chain modeling approach can reflect the actual window operating behavior accurately within an open-plan office,which is a beneficial supplement for energy-consumption simulation software in a window-state prediction module.The state-based Markov chain modeling approach showed better stability and accuracy in terms of the opening rate,whereas the action-based Markov chain modeling approach showed good consistency with the measurement data in the on-off curves and in situations with little data.For the on-off curves,the accuracy of action-based modeling approach in the prediction of window open-state is 20%higher.

Epidemiology of Depression and Suicide Ideation in Patients With Psoriasis:A Metaanalysis of Prospective Cohort Studies

Objective:Psoriasis is an immune-mediated inflammatory,chronic,recurrent skin disease associated with a high risk of developing psychiatric disorders,especially depression and suicidal ideation,leading to functional disability and poor quality of life.This study was conducted to comprehensively review and assess the epidemiologic association between psoriasis and the risk ratios(RRs)of depression or suicidal ideation.Methods:Five databases(PubMed,Wanfang Database,CNKI,The Cochrane Library,and EMBASE)were searched for prospective cohort studies on the prevalence of depression and/or suicidal ideation in patients with psoriasis updated to February 2,2023.Two independent reviewers evaluated and extracted the data,which were then pooled into a summary RR with corresponding 95%confidence interval(CI)using random-effects models.Results:Sixteen cohort studies comprising 1,166,840 patients with psoriasis and 3,294,205 controls were eligible for the final analysis.The pooled RR for depression was 1.43(95%CI=1.13-1.81)in patients with psoriasis and1.55(95%CI=1.40-1.71)in patients with psoriatic arthritis.In the subgroup analysis,Asian patients with psoriasis(RR=1.38,95%CI=1.17-1.63)had a lower pooled RR for depression than non-Asian patients(RR=1.45,95%CI=1.07-1.97),and patients with moderate-to-severe psoriasis(RR=1.69,95%CI=1.15-2.50)showed a higher RR for depression than patients with mild psoriasis(RR=1.60,95%CI=1.06-2.42).We also found no increase in the RR for suicidal ideation among people with psoriasis(RR=1.25,95%CI=0.95-1.65).Conclusion:Patients with psoriasis are at an increased risk of depression.Among patients with psoriasis,those with psoriatic arthritis,those who are non-Asian,and those with moderate-to-severe psoriasis are at higher risk for depression.However,the available evidence does not support an association between psoriasis and suicidal ideation.
