Lipidomic investigation of the protective effects of Polygonum perfoliatum against chemical liver injury in mice

Objective:Recent investigations have demonstrated that Polygonum perfoliatum L.can protect against chemical liver injury,but the mechanism behind its efficacy is still unclear.Therefore,we studied the pharmacological mechanism at work in P.perfoliatum protection against chemical liver injury.Methods:To evaluate the activity of P.perfoliatum against chemical liver injury,levels of alanine transaminase,lactic dehydrogenase,aspartate transaminase,superoxide dismutase,glutathione peroxidase and malondialdehyde were measured,alongside histological assessments of the liver,heart and kidney tissue.A nontargeted lipidomics strategy based on ultra-performance liquid chromatography quadrupoleorbitrap high-resolution mass spectrometry method was used to obtain the lipid profiles of mice with chemical liver injury and following treatment with P.perfoliatum;these profiles were used to understand the possible mechanisms behind P.perfoliatum's protective activity.Results:Lipidomic studies indicated that P.perfoliatum protected against chemical liver injury,and the results were consistent between histological and physiological analyses.By comparing the profiles of liver lipids in model and control mice,we found that the levels of 89 lipids were significantly changed.In animals receiving P.perfoliatum treatment,the levels of 8 lipids were significantly improved,relative to the model animals.The results showed that P.perfoliatum extract could effectively reverse the chemical liver injury and significantly improve the abnormal liver lipid metabolism of mice with chemical liver injury,especially glycerophospholipid metabolism.Conclusion:Regulation of enzyme activity related to the glycerophospholipid metabolism pathway may be involved in the mechanism of P.perfoliatum's protection against liver injury.

Cryptocurrency, as a typical application scene of blockchain, has attracted broad interests from both industrial and academic communities. With its rapid development, the cryptocurrency transaction network embedding(CTNE) has become a hot topic. It embeds transaction nodes into low-dimensional feature space while effectively maintaining a network structure,thereby discovering desired patterns demonstrating involved users' normal and abnormal behaviors. Based on a wide investigation into the state-of-the-art CTNE, this survey has made the following efforts: 1) categorizing recent progress of CTNE methods, 2) summarizing the publicly available cryptocurrency transaction network datasets, 3) evaluating several widely-adopted methods to show their performance in several typical evaluation protocols, and 4) discussing the future trends of CTNE. By doing so, it strives to provide a systematic and comprehensive overview of existing CTNE methods from static to dynamic perspectives,thereby promoting further research into this emerging and important field.

Synergistic Effect of Dual-Doped Carbon on MO_(2)C Nanocrystals Facilitates Alkaline Hydrogen Evolution

Molybdenum carbide(MO_(2)C)materials are promising electrocatalysts with potential applications in hydrogen evolution reaction(HER)due to low cost and Pt-like electronic structures.Nevertheless,their HER activity is usually hindered by the strong hydrogen binding energy.Moreover,the lack of water-cleaving site's makes it difficult for the catalysts to work in alkaline solutions.Here,we designed and synthesized a B and N dual-doped carbon layer that encapsulated on MO_(2)C nanocrystals(MO_(2)C@BNC)for accelerating HER under alkaline condition.The electronic interactions between the MO_(2)C nanocrystals and the multiple-doped carbon layer endow a near-zero H adsorption Gibbs free energy on the defective C atoms over the carbon shell.Meanwhile,the introduced B atoms afford optimal H_2O adsorption sites for the water-cleaving step.Accordingly,the dual-doped MO_(2)C catalyst with synergistic effect of non-metal sites delivers superior HER performances of a low overpotential(99 mV@10 mA cm^(-2))and a small Tafel slope(58.1 mV dec^(-1))in 1 M KOH solution.Furthermore,it presents a remarkable activity that outperforming the commercial 10%Pt/C catalyst at large current density,demonstrating its applicability in industrial water splitting.This study provides a reasonable design strategy towards noble-metal-free HER catalysts with high activity.

First-principles investigation of β-Ge3N4 loaded with RuO2 cocatalyst for photocatalytic overall water splitting

β-Ge3N4 loaded with nanoparticulate RuO2 as a cocatalyst is the first successful non-oxide photocatalyst for overall water splitting.To get an insight into the working mechanism of this particular photocatalytic system,we have calculated geometrical structures of low-index surfaces forβ-Ge3N4.Analysis of surface energies indicates that the most preferentially exposed surface is(100).The band gap of surface is narrower than that of bulk due to the dangling bonds.Dissociative water adsorption on(100)surface is thermodynamically favorable.The adsorption behavior of(RuO2)n(n=2,3,and 4)clusters on theβ-Ge3N4(100)surface has been explored.It is found that all the clusters bind to(100)surface strongly by forming interfacial bonds so that the adsorptions are exothermic processes.The calculation on density of states forβ-Ge3N4(100)surface loaded with(RuO2)nclusters reveals that photo-induced electrons tend to accumulate on(RuO2)nclusters and holes tend to stay inβ-Ge3N4.Based on the theoretical indication of Type-II staggered band alignment,we have proposed that in photocatalytic water splitting reaction,oxygen evolution reaction is inclined to occur on the surface ofβ-Ge3N4 while hydrogen evolution reaction is apt to occur on(RuO2)nclusters.In a word,loading RuO2 nanoparticles as a reduction cocatalyst benefits the charge separation inβ-Ge3N4.Furthermore,attaching(RuO2)nclusters ontoβ-Ge3N4(100)surface results in the redshift of absorption edge and the increase of absorption intensity.Our calculations have reasonably explained the experimental observation on the decomposition of water into H2 and O2 after loading RuO2 cocatalyst inβ-Ge3N4 photocatalyst.

A recent outbreak of coronavirus disease 2019(COVID-19)caused by the single-stranded enveloped RNA virus severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has developed into a global pandemic,after it was first reported in Wuhan in December 2019.SARS-CoV-2 is an emerging virus,and little is known about the basic characteristics of this pathogen,the underlying mechanism of infection,and the potential treatments.The immune system has been known to be actively involved in viral infections.To facilitate the development of COVID-19 treatments,the understanding of immune regulation by this viral infection is urgently needed.This review describes the mechanisms of immune system involvement in viral infections and provides an overview of the dysregulation of immune responses in COVID-19 patients in recent studies.Furthermore,we emphasize the role of gut microbiota in regulating immunity and summarized the impact of SARS-CoV-2 infection on the composition of the microbiome.Overall,this review provides insights for understanding and developing preventive and therapeutic strategies by regulating the immune system and microbiota.

Adaptive sliding mode control of modular self-reconfigurable spacecraft with time-delay estimation

The reconstruction control of modular self-reconfigurable spacecraft (MSRS) is addressed using an adaptive sliding mode control (ASMC) scheme based on time-delay estimation (TDE) technology. In contrast to the ground, the base of the MSRS is floating when assembled in orbit, resulting in a strong dynamic coupling effect. A TED-based ASMC technique with exponential reaching law is designed to achieve high-precision coordinated control between the spacecraft base and the robotic arm. TDE technology is used by the controller to compensate for coupling terms and uncertainties, while ASMC can augment and improve TDE’s robustness. To suppress TDE errors and eliminate chattering, a new adaptive law is created to modify gain parameters online, ensuring quick dynamic response and high tracking accuracy. The Lyapunov approach shows that the tracking errors are uniformly ultimately bounded (UUB). Finally, the on-orbit assembly process of MSRS is simulated to validate the efficacy of the proposed control scheme. The simulation results show that the proposed control method can accurately complete the target module’s on-orbit assembly, with minimal perturbations to the spacecraft’s attitude. Meanwhile, it has a high level of robustness and can effectively eliminate chattering.

Human gastric cancer is a highly lethal disease,but the underlying multiomic mole cular signatures remain largely uncle ar.Here,we performed multi-regional sampling,parallel single-cell multiomics sequencing and integrated analyses of human gastric cancer.We identified common transcrip tomic alterations of gastric cancer cells,such as aberrant down-regulation of genes associated with normal stomach function and up-regulation of KRT7,PI3,S100A4,etc.Surprisingly,aberrant and prevalent up-regulation of genes highly expressed in normal colorectal epithelial cells were also identified in cancer cells,which maybe partially regulated by promoter chromatin accessibility and DNA methylation levels.We reve aled the single-cell DNA methylome landscape of gastric cancer,and identified candidate DNA methylation biomarkers,such as hypermethylated promoters of TMEM240 and HAGLROS,and hypomethylated promoters of TRPM2-AS and HRH1.Additionally,the relationships between genetic lineages,D NA methylation and transcriptomic clusters were systematically revealed at single-cell level.We showed that DNA methylation heterogeneities were mainly among different genetic lineages of cancer cells.Moreover,we found that D NA methylation levels of cancer cells with poorer differentiation states tend to be higher than those of cancer cells with better differentiation states in the primary tumor within the same patient,although still lower than in normal gastric epithelial cells.Cancer cells with poorer differentiation states also prevalently down-regulated MUC1 expression and immune-related pathways,and had poor infiltration of CD 8+T cells.Our study dissected the mole cular signatures of intratumoral heterogeneities and differentiation states of human gastric cancer using integrative single-cell multiomics analyses.

Invasive fungal infections are life-threatening,and neutrophils are vital cells of the innate immune system that defend against them.The role of LTA4H-LTB_(4)-BLT1 axis in regulation of neutrophil responses to fungal infection remains poorly understood.Here,we demonstrated that the LTA4H-LTB_(4)-BLT1 axis protects the host against Candida albicans and Aspergillus fumigatus,but not Cryptococcus neoformans infection,by regulating the antifungal activity of neutrophils.Our results show that deleting Lta4h or Blt1 substantially impairs the fungal-specific phagocytic capacity of neutrophils.Moreover,defective activation of the spleen tyrosine kinase(Syk)and extracellular signal-related kinase(ERK1/2)pathways in neutrophils accompanies this impairment.Mechanistically,BLT1 regulates CR3-mediated,β-1,3-glucan-induced neutrophil phagocytosis,while a physical interaction with CR3 with slight influence on its dynamics is observed.Our findings thus demonstrate that the LTA4H-LTB_(4)-BLT1 axis is essential for the phagocytic function of neutrophils in host antifungal immune response against Candida albicans and Aspergillus fumigatus.

70 nm: The most unstable grain size in Cu prepared by surface mechanical grinding treatment

The stability of Cu with different average grain sizes prepared by surface mechanical grinding treatment were investigated under the conditions of isothermal annealing and uniaxial tension.In both conditions,experimental results revealed that the stability of the grains decreased with the decrease of grain size when the grain size was above 70–75 nm,while the stability of the grains increased with the decrease of grain size when the grain size was below 70–75 nm.The grains of about 70–75 nm in size showed the worst stability in both thermal and mechanical conditions due to having the highest level of average atom free energy and their large amount of high energy grain boundary with large curvature.This size was very close to the calculated smallest size achievable by severe plastic deformation based on the grain refinement mechanism of dislocation evolution under present processing condition,at which both the highest density of dislocation and highest energy should be produced and induces poor stability.Below 70 nm,the deformation mechanism of nanograined Cu was transformed into a partial dislocation motion,which activated mechanically induced grain boundary(GB)relaxation accompanied by GB flattening and GB energy decrease and resulted in enhanced stability.This discovery offers the potential for developing nanograined metals with high strength and high stability.
