Amphioxus Gli knockout disrupts the development of left–right asymmetry but has limited impact on neural patterning

Cephalochordates,the basal invertebrate chordates within the phylum Chordata,are key to understanding the evolution of developmental mechanisms during the invertebrate-chordate to vertebrate ***,one impediment to amphioxus as a model organism for transgenic research and functional assay of cis-regulatory modules(CRMs),such as enhancers and silencers,has the inability to obtain an extremely high efficient molecular tool such as proximal and basal(or core,minimal) *** this,here,we describe the identification and characterization of the proximal promoter(-745 to -1) and the basal promoter(-129 to -1)(Chinese patent No.201210043564.4) of an amphioxus(Branchiostoma belcheri)β-ACTIN gene(ACTIN-6-2).In the proximal promoter region,typical cis-regulatory elements ofβ-ACTIN gene:TATA, CArG and CAAT were identified at position -56,-130 and -705 *** 745 bp construct can drive reporter gene expression broadly in 91.46%of injected amphioxus *** including the 129 bp alone or plus a random genome fragment show no activity to express reporter gene in injected embryos,however when being integrated a previously reported strong positive enhancer element into the 129 bp construct,as high as 93.82%of injected embryos show reporter gene expression in a tissue-specific ***,we observed these two promoters have significantly high efficiency than the human elongation factor-1-α(EF-1-α) promoter(only have 38.20%efficiency when introducing into amphioxus embryos),which was broadly used in many commercialized mammalian expression ***,these two ACTIN promoters represent the most effective proximal and basal promoters identified so far in amphioxus and will be very useful for future transgenic research and functional assays of CRMs,which will giving an impulse to the process of amphioxus as a model organism. The Gli transcription factors are the primary mediators of Hedgehog(Hh)*** genomes contain multiple Gli paralogues with different functions downstream of Hh signal receipt,in part explaining the complexity of cellular responses to Hh that allow concentration-dependent target gene *** is a chordate that split from the vertebrate lineage early in the evolution of chordates,before the genome duplications that occurred in early vertebrate *** has a single Gli gene whose transcripts can be alternately spliced to yield two protein isoforms called GliS and *** generated two knockout mutations in amphioxus Gli,one that affects the whole gene and a second that only affects *** knockouts showed major morphological and molecular defects in the development of left–right asymmetry,a phenotype that is similar but not identical to that previously found in Hh *** signaling also patterns the amphioxus neural ***,however,knockout of GliL showed no identifiable phenotype,while knockout of the full gene showed only small changes to the expression of one gene family,*** genes that were prominently affected by Hh knockout were not altered in expression in either *** for the differences between Hh and Gli knockouts in the pharynx and neural tube are discussed in the context of the likely different functions of amphioxus Gli isoforms.

Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological *** a broad spectrum of aging biomarkers has been developed,their potential uses and limitations remain poorly *** immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower?This review aims to address this ***,we summarize our current knowledge of biomarkers developed for cellular,organ,and organismal levels of aging,comprising six pillars:physiological characteristics,medical imaging,histological features,cellular alterations,molecular changes,and secretory *** fulfill all these requisites,we propose that aging biomarkers should qualify for being specific,systemic,and clinically relevant.

Mammalian target of rapamycin(m TOR) inhibitor as an attractive drug target with promising antitumor effects has been widely investigated. High quality clinical trial has been conducted in liver transplant(LT) recipients in Western countries. However, the pertinent studies in Eastern world are paucity. Therefore, we designed a clinical trial to test whether sirolimus can improve recurrence-free survival(RFS) in hepatocellular carcinoma(HCC) patients beyond the Milan criteria after LT. This is an open-labeled, single-arm, prospective, multicenter, and real-world study aiming to evaluate the clinical outcomes of early switch to sirolimus-based regimens in HCC patients after LT. Patients with a histologically proven HCC and beyond the Milan criteria will be enrolled. The initial immunosuppressant regimens are center-specifc for the frst 4-6 weeks. The following regimens integrated sirolimus into the regimens as a combination therapy with reduced calcineurin inhibitors based on the condition of patients and centers. The study is planned for 4 years in total with a 2-year enrollment period and a 2-year follow-up. We predict that sirolimus conversion regimen will provide survival benefts for patients particular in the key indicator RFS as well as better quality of life. If the trial is conducted successfully, we will have a continued monitoring over a longer follow-up time to estimate indicator of overall survival. We hope that the outcome will provide better evidence for clinical decision-making and revising treatment guidelines based on Chinese population data.

The Jiangmen Underground Neutrino Observatory(junO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main *** of junO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator *** this paper,we present a comprehensive assessment of junO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/*** ten years of data acquisition,approximately 60,000 signal and 30,000 background events are *** large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation *** m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),junO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith ***,junO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.

厦门翔安大桥施工区域海洋地质环境复杂,而且处于台风高发区域。同时,深海区的23个桥墩基础均采用无栈桥水上平台法进行施工,所有施工资源均依靠水运至墩位,水上施工设备需求数量大,交通组织困难。在考虑国内已建特大型跨海大桥桩基施工经验的基础上,通过现场调研以及收集、整理和分析现有资料,研究了复杂海域条件下钢管精准施沉技术和快速成孔成桩技术,为复杂海域大直径变截面钢管复合桩基础施工提供技术指导。

junO is a multi-purpose neutrino observatory under construction in the south of *** publication presents new sensitivity estimates for the measurement of the △m_(31)^(2),△m_(21)^(2),sin^(2)θ_(12),and sin^(2)θ_(13) oscillation parameters using reactor antineutrinos,which is one of the primary physics goals of the *** sensitivities are obtained using the best knowledge available to date on the location and overburden of the experimental site,the nuclear reactors in the surrounding area and beyond,the detector response uncertainties,and the reactor antineutrino spectral shape constraints expected from the TAO satellite *** is found that the △m_(21)^(2) and sin^(2)θ_(12) oscillation parameters will be determined to 0.5%precision or better in six years of data *** the same period,the △m_(31)^(2) parameter will be determined to about 0.2%precision for each mass ordering *** new precision represents approximately an order of magnitude improvement over existing constraints for these three parameters.

装配式承台的海绵—橡胶止水技术是在综合港珠澳大桥和青岛海湾大桥的止水技术经验基础上,依托厦门第二东通道工程提出的一种新型止水施工工艺。橡胶止水带通过预压、膨胀后产生变形挤密间隙,从而达到环向止水的效果,主要靠海绵条的弹性恢复力与钢管壁之间产生挤压实现竖向止水;速凝砂浆起到二次止水作用。海绵—橡胶止水技术在现场施工中取得了良好的止水效果,有效控制了止水失效的风险,可以高效、经济地实现承台与钢管复合桩之间的止水。
