We propose a nonparametric change point estimator in the distributions of a sequence of independent observations in terms of the test statistics given by Huˇskov′a and Meintanis(2006) that are based on weighted empirical characteristic functions. The weight function ω(t; a) under consideration includes the two weight functions from Huˇskov′a and Meintanis(2006) plus the weight function used by Matteson and James(2014),where a is a tuning parameter. Under the local alternative hypothesis, we establish the consistency, convergence rate, and asymptotic distribution of this change point estimator which is the maxima of a two-side Brownian motion with a drift. Since the performance of the change point estimator depends on a in use, we thus propose an algorithm for choosing an appropriate value of a, denoted by a_s which is also justified. Our simulation study shows that the change point estimate obtained by using a_s has a satisfactory performance. We also apply our method to a real dataset.

Joint observations of the large-scale ULF wave activity from space to ground associated with the solar wind dynamic pressure enhancement

This study reports the rare ultralow-frequency(ULF) wave activity associated with the solar wind dynamic pressure enhancement that was successively observed by the GOES-17(Geostationary Operational Environmental Satellite) in the magnetosphere, the CSES(China Seismo-Electromagnetic Satellite) in the ionosphere, and the THEMIS ground-based observatories(GBO) GAKO and EAGL in the Earth's polar region during the main phase of an intense storm on 4 November 2021. Along with the enhanced-pressure solar wind moving tailward, the geomagnetic field structure experienced a large-scale change. From dawn/dusk sides to midnight, the GAKO, EAGL, and GOES-17 sequentially observed the ULF waves in a frequency range of0.04–0.36 Hz at L shells of ~5.07, 6.29, and 5.67, respectively. CSES also observed the ULF wave event with the same frequency ranges at wide L-shells of 2.52–6.22 in the nightside ionosphere. The analysis results show that the ULF waves at ionospheric altitude were mixed toroidal-poloidal mode waves. Comparing the ULF waves observed in different regions, we infer that the nightside ULF waves were directly or indirectly excited by the solar wind dynamic pressure increase: in the area of L-shells~2.52–6.29, the magnetic field line resonances(FLRs) driven by the solar wind dynamic pressure increase is an essential excitation source;on the other hand, around L~3.29, the ULF waves can also be excited by the outward expansion of the plasmapause owing to the decrease of the magnetospheric convection, and in the region of L-shells ~5.19–6.29, the ULF waves are also likely excited by the ion cyclotron instabilities driven by the solar wind dynamic pressure increase.

Genetic gain-of-function mutations of warm temperature-sensitive transient receptor potential vanilloid 3(TRPV3)channel cause Olmsted syndrome characterized by severe itching and keratoderma,indicating that pharmacological inhibition of TRPV3 may hold promise for therapy of chronic pruritus and skin ***,currently available TRPV3 tool inhibitors are either nonselective or less potent,thus impeding the validation of TRPV3 as therapeutic *** whole-cell patch-clamp and single-channel recordings,we report the identification of two natural dicaffeoylquinic acid isomers isochlorogenic acid A(IAA)and isochlorogenic acid B(IAB)that selectively inhibit TRPV3 currents with IC50 values of 2.7±1.3 and 0.9±0.3μmol/L,respectively,and reduce the channel open probability to 3.7±1.2%and 3.2±1.1%from 26.9±5.5%,*** vivo evaluation confirms that both IAA and IAB significantly reverse the ear swelling of dermatitis and chronic ***,the isomer IAB is able to rescue the keratinocyte death induced by TRPV3 agonist *** docking combined with site-directed mutations reveals two residues T636 and F666 critical for the binding of the two *** together,our identification of isochlorogenic acids A and B that act as specific TRPV3 channel inhibitors and gating modifiers not only provides an essential pharmacological tool for further investigation of the channel pharmacology and pathology,but also holds developmental potential for treatment of dermatitis and chronic pruritus.

The Shanghai Synchrotron Radiation Facility (SSRF) is a low emittance third-generation synchrotron radiation light source under commissioning. The excitation curve calibration for the booster magnets is important to provide the magnet current configurations as reference. Calibration studies give the polynomial coefficients of each type of magnets and provide the magnet current configurations under different beam energies as beam is ramped at speed of 2 Hz. The applications of calibration in booster commissioning which show the accuracy of the magnetic excitation curve calibration are also discussed.

Objective:Family caregivers(FCs)of breast cancer patients play a vital role throughout the treatment *** distress of FCs is common and often ignored.A simple and effective instrument for screening psychological distress would help in selecting those FCs requiring special attention and ***,the validity of distress thermometer(DT)in FCs of Chinese breast cancer patients receiving postoperative chemotherapy was assessed,and the prevalence of anxiety and depression was ***:We recruited 200 FCs of hospitalized breast cancer patients in this cross-sectional descriptive *** the first cycle of adjuvant chemotherapy,the levels of anxiety and depression among FCs were assessed using DT and Hospital Anxiety and Depression Scale(HADS).In total,191 valid cases were *** was used as the diagnostic standard to assess the effectiveness of DT as a screening tool for anxiety and depression as well as to analyze the diagnostic efficiency of DT at various cutoff ***:The definitive prevalence of both anxiety and depression was 8.90%.The mean level of anxiety and depression among FCs was 5.64±3.69 and 5.09±3.85,respectively,both of which were significantly higher than corresponding Chinese norms(P<0.01).The areas under receiver operating characteristic curves of DT for the diagnoses of FCs'anxiety and depression were 0.904 and 0.885,respectively.A cutoff value of 5 produced the best diagnostic effects of DT for anxiety and ***:The levels of both anxiety and depression were higher in the FCs of Chinese breast cancer patients receiving postoperative chemotherapy than the national *** might be an effective tool to initially screen psychological distress among *** process could be integrated into the palliative care of breast cancer patients and warrant further research.

Objective: The mechanism of acquired gene mutation plays a major role in resistance to endocrine therapy in hormone receptor(HR)-positive advanced breast cancer. Circulating tumor DNA(ctDNA) has been allowed for the assessment of the genomic profiles of patients with advanced cancer. We performed this study to search for molecular markers of endocrine therapy efficacy and to explore the clinical value of ctDNA to guide precise endocrine therapy for HR-positive/human epidermal growth factor receptor-2(HER-2)-negative metastatic breast cancer ***: In this open-label, multicohort, prospective study, patients were assigned to four parallel cohorts and matched according to mutations identified in ctDNA: 1) activation of the phosphatidylinositol-3-kinase(PI3K)/AKT/mammalian target of rapamycin(mTOR) signaling pathway preferred mTOR inhibitor combined with endocrine therapy;2) estrogen receptor 1(ESR1) mutation preferred fulvestrant;3) HER-2 mutations preferred pyrotinib;and 4) no actionable mutations received treatment according to the clinical situation. In all cohorts, patients were divided into compliance group and violation group. The primary outcome measure was progression-free survival(PFS), and the secondary outcome measure was overall survival(OS).Results: In all cohorts, the combined median PFS was 4.9 months, and median PFS for the compliance and violation groups was 6.0 and 3.0 months, respectively [P=0.022, hazard ratio(HR)=0.57]. Multivariate Cox regression model showed the risk of disease progression was lower in compliance group than in violation group(P=0.023, HR=0.55). Among the patients with HER-2 mutations, the median PFS was 11.1 months in the compliance group and 2.2 months in the violation group(P=0.011, HR=0.20).
