A putative 6-transmembrane nitrate transporter OsNRT1.1b plays a key role in rice under low nitrogen

OsNRT1.1a is a low-affinity nitrate（NO_3^-） transporter gene. In this study, another mRNA splicing product, OsNRT1.1b,putatively encoding a protein with six transmembrane domains, was identified based on the rice genomic database and bioinformatics analysis. OsNRT1.1a/OsNRT1.1b expression in Xenopus oocytes showed OsNRT1.1a-expressing oocytes accumulated ^（15）N levels to about half as compared to OsNRT1.1bexpressing oocytes. The electrophysiological recording of OsNRT1.1b-expressing oocytes treated with 0.25 mM NO_3^- confirmed ^（15）N accumulation data. More functional assays were performed to examine the function of OsNRT1.1b in rice. The expression of both OsNRT1.1a and OsNRT1.1b was abundant in roots and downregulated by nitrogen（N） deficiency. The shoot biomass of transgenic rice plants with OsNRT1.1a or OsNRT1.1b overexpression increased under various N supplies under hydroponic conditions compared to wild-type（WT）. The OsNRT1.1a overexpression lines showed increased plant N accumulation compared to the WT in 1.25 mM NH_4NO_3 and 2.5 mM NO_3^- or NH_4~＋ treatments, but not in 0.125 mM NH_4NO_***, OsNRT1.1b overexpression lines increased total N accumulation in all N treatments, including 0.125 m M NH_4NO_3,suggesting that under low N condition, OsNRT1.1b would accumulate more N in plants and improve rice growth, but also that OsNRT1.1a had no such function in rice plants.

Considering the action mechanisms of overpressure on physical changes in skeleton particles of deep reservoir rocks and the differences in physical changes of skeleton particles under overpressure and hydrostatic pressure, the sandstone of the Jurassic Toutunhe Formation in the southern margin of Junggar Basin was taken as an example for physical modeling experiment to analyze the action mechanisms of overpressure on the physical properties of deep reservoirs. (1) In the simulated ultra-deep layer with a burial depth of 6000-8000 m, the mechanical compaction under overpressure reduces the remaining primary pores by about a half that under hydrostatic pressure. Overpressure can effectively suppress the mechanical compaction to allow the preservation of intergranular primary pores. (2) The linear contact length ratio under overpressure is always smaller than the linear contact length ratio under hydrostatic pressure at the same depth. In deep reservoirs, the difference between the mechanical compaction degree under overpressure and hydrostatic pressure shows a decreasing trend, the effect of abnormally high pressure to resist the increase of effective stress is weakened, and the degree of mechanical compaction is gradually close to that under hydrostatic pressure. (3) The microfractures in skeleton particles of deep reservoirs under overpressure are thin and long, while the microfractures in skeleton particles of deep reservoirs under hydrostatic pressure are short and wide. This difference is attributed to the probable presence of tension fractures in the rocks containing abnormally high pressure fluid. (4) The microfractures in skeleton particles under overpressure were mainly formed later than that under hydrostatic pressure, and the development degree and length of microfractures both extend deeper. (5) The development stages of microfractures under overpressure are mainly controlled by the development stages of abnormally high pressure and the magnitude of effect

Nitrate is an important nitrogen source and signaling molecule that regulates plant growth and *** several components of the nitrate signaling pathway have been identified,the detailed mechanisms are still *** previous results showed that OsMADS25 can regulate root development in response to nitrate signals,but the mechanism is still ***,we try to answer two key questions:how does OsMADS25 move from the cytoplasm to the nucleus,and what are the direct target genes activated by OsMADS25 to regulate root growth after it moves to the nucleus in response to nitrate?Our results demonstrated that OsMADS25 moves from the cytoplasm to the nucleus in the presence of nitrate in an *** immunoprecipitation sequencing,chromatin immunoprecipitation qPCR,yeast one-hybrid,and luciferase experiments showed that OsMADS25 directly activates the expression of OsMADS27 and OsARF7,which are reported to be associated with root ***,OsMADS25-RNAi lines,the Osnar2.1 mutant,and OsMADS25-RNAi Osnar2.1 lines exhibited significantly reduced root growth compared with the wild type in response to nitrate supply,and expression of OsMADS27 and OsARF7 was significantly suppressed in these ***,these results reveal a new mechanismby which OsMADS25 interacts with *** interaction is required for nuclear accumulation of OsMADS25,which promotes OsMADS27 and OsARF7 expression and root growth in a nitratedependent manner.

The importance of the nitrate(NO) transporter for yield and nitrogen-use efficiency(NUE) in rice was previously demonstrated using map-based cloning. In this study we enhanced the expression of the OsNRT2.1 gene,which encodes a high-affinity NO transporter,using a ubiquitin(Ubi) promoter and the NO-inducible promoter of the OsNAR2.1 gene to drive OsNRT2.1 expression in transgenic rice plants. Transgenic lines expressing p Ubi:OsNRT2.1 or p OsNAR2.1:OsNRT2.1 constructs exhibited increased total biomass including yields of approximately 21% and 38% compared with wild-type(WT) plants. The agricultural NUE(ANUE) of the p Ubi:OsNRT2.1 lines decreased to 83% of that of WT plants,while the ANUE of the p OsNAR2.1:OsNRT2.1 lines increased to 128% of that of WT plants. The dry matter transfer(DMT) into grain decreased by 68% in the p Ubi:OsNRT2.1 lines and increased by 46% in the pOsNAR2.1:OsNRT2.1 lines relative to the WT. The expression of OsNRT2.1 in shoot culms showed that Ubi enhanced OsNRT2.1 expression by 3 to 20-fold and OsNAR2.1 promoters increased by 33% to 45% higher than the WT. The ratio of OsNRT2.1 to OsNAR2.1 expression was altered in the transgenic lines with ratios of approximately 11.3:1 and 4.7:1 in the p Ubi:OsNRT2.1 and p OsNAR2.1:OsNRT2.1 lines,compared with a ratio of 7.2:1 in WT plants. We show that increased expression of OsNRT2.1,especially in combination with a relative lower expression ratio with its partner gene OsNAR2.1,can improve yield and NUE in rice.

[目的]探讨音乐治疗对大学生社交焦虑的干预效果。[方法]选取太原市某大学理工科专业在校本科生进行社交焦虑测试,有46人存在社交焦虑,采用随机数字表法将其分为试验组及对照组各23人。对照组给予常规干预,试验组在此基础上进行音乐治疗干预,干预10次后比较两组学生的心理症状和社交焦虑情况。[结果]试验组心理症状和社交焦虑评分均低于对照组(P<0.05)。[结论]音乐治疗有助于改善大学生的社交焦虑。

益母草注射液在产后出血患者防治的应用

目的：探讨益母草注射液在产后出血患者防治的应用效果。方法：剖宫产分娩者160例按入院先后依单双数分为观察组80例与对照组80例，2组分别于胎儿娩出后给予益母草注射液与缩宫素治疗。结果：观察组的术后2—24小时与术后24—48小时的出血量明显少于对照组（P〈0．05），而2组手术中出血量对比无明显差异（P〉0．05）。观察组术后的PT、APTT与FIB值明显高于对照组，差异有统计学意义（P〈0．05）。结论：益母草注射液可以减少剖宫产术后出血量，其作用的发挥可能与增强凝血功能有关。
