Characterization of Genome-Wide Microsatellites of Saccharina japonica Based on a Preliminary Assembly of Illumina Sequencing Reads

Microsatellites or simple sequence repeats(SSR) function widely and locate dependently in genome. However, their characteristics are often ignored due to the lack of genomic sequences of most species. Kelp(Saccharina japonica), a brown macroalga, is extensively cultured in China. In this study, the genome of S. japonica was surveyed using an Illumina sequencing platform, and its microsatellites were characterized. The preliminarily assembled genome was 469.4 Mb in size, with a scaffold N_(50) of 20529 bp. Among the 128370 identified microsatellites, 90671, 25726 and 11973 were found in intergenic regions, introns and exons, averaging 339.3, 178.8 and 205.4 microsatellites per Mb, respectively. These microsatellites distributed unevenly in S. japonica genome. Mononucleotide motifs were the most abundant in the genome, while trinucleotide ones were the most prevalent in exons. The microsatellite abundance decreased significantly with the increase of motif repeat numbers, and the microsatellites with a small number of repeats accounted for a higher proportion of the exons than those of the intergenic regions and introns. C/G-rich motifs were more common in exons than in intergenic regions and introns. These characteristics of microsatellites in S. japonica genome may associate with their functions, and ultimately their adaptation and evolution. Among the 120140 pairs of designed microsatellite primers, approximately 75% were predicted to be able to amplify S. japonica DNA. These microsatellite markers will be extremely useful for the genetic breeding and population evolution studies of kelp.

Carcinoma-associated fibroblasts(CAFs)are the main cellular components of the tumor microenvironment and promote cancer progression by modifying the extracellular matrix(ECM).The tumor-associated ECM is characterized by collagen crosslinking catalyzed by lysyl oxidase(LOX).Small extracellular vesicles(sEVs)mediate cell-cell ***,the interactions between sEVs and the ECM remain ***,we demonstrated that sEVs released from oral squamous cell carcinoma(OSCC)-derived CAFs induce collagen crosslinking,thereby promoting epithelial-mesenchymal transition(EMT).CAF sEVs preferably bound to the ECM rather than being taken up by fibroblasts and induced collagen crosslinking,and a LOX inhibitor or blocking antibody suppressed this *** LOX(αLOX),but not the LOX precursor,was enriched in CAF sEVs and interacted with periostin,fibronectin,and bone morphogenetic protein-1 on the surface of *** sEV-associated integrinα2β1 mediated the binding of CAF sEVs to collagen I,and blocking integrinα2β1 inhibited collagen crosslinking by interfering with CAF sEV binding to collagen *** sEV-induced collagen crosslinking promoted the EMT of OSCC through FAK/paxillin/YAP *** together,these findings reveal a novel role of CAF sEVs in tumor ECM remodeling,suggesting a critical mechanism for CAF-induced EMT of cancer cells.

目的分析非HIV隐球菌性脑膜炎患者的临床特征及体外药物敏感情况。方法回顾性分析医院2018-2021年收治的138例隐球菌性脑膜炎患者的临床资料,对患者临床表现、基础病情和实验室检查等数据进行分析。结果 138例非HIV隐球菌性脑膜炎患者墨汁染色检测阳性率为73.9%,CrAg检测阳性率为100%,42例患者存在免疫功能抑制疾病,49例伴有其他基础疾病,另外47名患者无基础疾病。患者多伴有头痛、发热、呕吐、四肢乏力和意识障碍等症状。脑脊液生化检查显示糖和氯化物减低,脑脊液蛋白升高。138株隐球菌对5-氟胞嘧啶、两性霉素B、氟康唑、伊曲康唑、伏立康唑的MIC范围分别为≤4~≥16μg/mL、≤0.5~1μg/mL、1~16μg/mL、≤0.125~0.5μg/mL、≤0.06~0.5μg/mL。5种抗真菌药物中5-氟胞嘧啶敏感性为95.60%,氟康唑敏感性为94.93%,伊曲康唑敏感性65.22%,两性霉素B和伏立康唑的敏感性均为100%。结论隐球菌性脑膜炎患者多伴有基础疾病(含免疫功能抑制疾病);CrAg检测阳性率明显高于墨汁染色检测;非HIV隐球菌性脑膜炎患者分离菌株对不同抗真菌药物敏感性有差异,为临床用药提供参考。

疏肝润肠方治疗肝郁气滞型便秘临床研究

目的:观察疏肝润肠方治疗肝郁气滞型便秘的临床疗效。方法:采用随机数字表法将64例肝郁气滞型便秘患者分为对照组和观察组各32例。对照组给予比沙可啶肠溶片口服治疗,观察组给予疏肝润肠方口服治疗。治疗4周后,比较2组完全自主排便次数(CSBM)、便秘症状积分、中医证候积分、便秘患者生活质量量表(PAC-QOL)评分及血清P物质(SP)、血管活性肠肽(VIP)、胃动素(MTL)、胃泌素(GAS)、5-羟色胺(5-HT)水平,并评估2组临床疗效和不良反应发生率。结果:观察组总有效率为93.75%,高于对照组75.00%(P<0.05)。治疗后,2组便秘症状评分、中医证候积分、PAC-QOL评分均较治疗前降低,且观察组低于对照组(P<0.05);2组CSBM较治疗前升高,且观察组CSBM高于对照组(P<0.05)。治疗后,2组5-HT、SP、MTL、GAS水平较较治疗前升高,VIP水平较治疗前降低(P<0.05);且观察组5-HT、SP、MTL、GAS水平高于对照组,VIP水平低于对照组(P<0.05)。结论:疏肝润肠方可有效改善肝郁气滞证便秘患者的便秘症状和中医证候,提高患者生活质量,调节机体胃肠激素水平。

The effect of Fe-Mn minerals and seawater interface and enrich-ment mechanism of ore-forming elements of polymetallic crusts and nodules from the South China Sea

Ferromanganese crusts and nodules are important submarine mineral resources that contain various metal elements with significant economic value. In this study, polymetallic crusts and nodules obtained from the South China Sea （SCS） were determined by using X-ray power diffraction （XRD）, Raman spectroscopy （RS）, Fourier transform infrared spectroscopy （FTIR）, and X-ray photoelectron spectroscopy （XPS） to systematically investigate and analyze the mineralogical and spectral characteristics of the Fe-Mn minerals. XRD measurements revealed that the SCS polymetallic crusts and nodules were composed of vernadite, quartz, and plagioclase. The nodules also contained todorokite. The Fe-phase minerals of the SCS crusts and nodules were composed of amorphous Fe oxide/hydroxide, and the Mn- and Fe-phases minerals exhibited relatively poor degrees of crystallization. FTIR results showed that the Fe-Mn minerals in the crusts and nodules included a large number of surface hydroxyl groups. These surface hydroxyl groups contained protons that could provide reactive sites for complexation of ore-forming elements in seawater. XPS results indicated that the surfaces of the Fe-Mn minerals mainly contained Fe, Mn, and O. Fe was present in the trivalent oxidation state, while Mn, which may contain several bivalent oxidation state, was present in the tetravalent and trivalent oxidation states. The SCS polymetallic crusts and nodules were compared with Pacific seamount crusts, and results showed that the surface hydroxyl （-OH） groups of the SCS crusts and nodules numbered more than the lattice oxygen （O^2-）. But the lattice oxygen of Pacific seamount crusts numbered more than the surface hydroxyl groups. This characteristic indicated that the degree of crystallization of Fe-Mn minerals from the Pacific Ocean was higher than that of minerals from the South China Sea. 普通中空玻璃内加装内置百叶会影响两侧玻璃之间的自然对流换热。本文通过实验测量和数值模拟研究了在一定温差条件下,内置百叶中空玻璃和无百叶中空玻璃系统的热工性能,通过改变空腔间距和百叶倾角来研究其对中空玻璃系统的传热系数的影响情况。研究结果表明,为使百叶起到较好的隔热效果,内置百叶的中空玻璃空腔间距应大于40 mm;百叶倾角为90°时的总传热系数U值最低;内置百叶可使普通中空玻璃的热阻值提高6%~30%。

Perceptual Image Outpainting Assisted by Low-Level Feature Fusion and Multi-Patch Discriminator

Recently,deep learning-based image outpainting has made greatly notable improvements in computer vision ***,due to the lack of fully extracting image information,the existing methods often generate unnatural and blurry outpainting results in most *** solve this issue,we propose a perceptual image outpainting method,which effectively takes the advantage of low-level feature fusion and multi-patch ***,we first fuse the texture information in the low-level feature map of encoder,and simultaneously incorporate these aggregated features reusability with semantic(or structural)information of deep feature map such that we could utilizemore sophisticated texture information to generate more authentic outpainting *** we also introduce a multi-patch discriminator to enhance the generated texture,which effectively judges the generated image from the different level features and concurrently impels our network to produce more natural and clearer outpainting ***,we further introduce perceptual loss and style loss to effectively improve the texture and style of outpainting *** with the existing methods,our method could produce finer outpainting *** results on Places2 and Paris StreetView datasets illustrated the effectiveness of our method for image outpainting.

以多肋藻(Costaria costata)雌雄配子体为材料,研究了蓝光、白光、红光对配子体生长发育的影响。结果表明:光质对多肋藻配子体的生长发育有显著的影响(P<0.05),雌配子体在蓝光条件下相对生长率(5.62%)显著高于红光(3.37%),但与白光下的相对生长率(4.94%)无显著差异;雄配子体在白光下的相对生长率(8.71%)最高,显著高于红光(6.73%),但与蓝光下的相对生长率(7.74%)无显著差异。相同光质条件下,雄配子体相对生长率显著高于雌配子体。整个培养周期过程中,红光条件下,配子体均未发育。相同培养天数下,蓝光组的发育率显著高于白光组。随着培养天数的增加,白光组和蓝光组的发育率均有显著的提高,白光组于第28天达到最大值21.13%,蓝光组于第20天达到最大值67.73%。
