Current status and prospects of hematopoietic stem cell transplantation in China

Hematopoietic stem cell transplantation(HSCT)is a highly effective and unique medical procedure for the treatment of most hematological malignancies.The first allogeneic transplantation was performed by E.Donnall Thomas in 1957.Since then,the field has evolved and expanded worldwide.The first successful allogenic HSCT(allo-HSCT)in China was conducted in 1981.Although the development of allo-HSCT in China lagged,China has since made considerable contributions to the process of HSCT worldwide,with more than 10,000 HSCTs performed annually.In particular,haploid HSCT(haplo-HSCT)technology represented in the Beijing Protocol has demonstrated similar efficacy to human leukocyte antigen-matched HSCT and has gradually become the pre-dominant choice for allo-HSCT in China.Currently,the number of haplo-HSCT procedures exceeds 5000 per year,and the Beijing Protocol has been greatly improved by implementing updated individualized strategies for controlling complications,relapse,and infection management.In addition,innovative haplo-HSCT technologies developed by different medical transplantation centers,such as Soochow,Zhejiang,Fujian,Chongqing,and Anhui,have emerged,providing inspiration for the refinement of global practice.This review will focus on the current activity in this field and highlight important trends that are vital in China’s allo-HSCT process,examining the current viewpoint and future directions.

Immune thrombocytopenia(ITP)is an autoimmune bleeding disorder characterized by increased platelet destruction and impaired platelet production.The pathophysiology of ITP is complicated and incompletely understood.It is traditionally recognized that antibody-coated platelets are prematurely cleared via Fc-independent or dependent mechanism in the spleen,liver,or both.Antibody-mediated complement activation and desialylation can also destroy platelets.In addition,antibodies can inhibit megakaryocyte production and maturation.Abnormalities in cellular immunity have also been described,including skewing of T helper(Th)cells toward a type 1 T helper(Th1)and type 17 T helper(Th17)phenotype,reduction of regulatory T-cell activity,and an increase in cytotoxic T cells.

Manganese ion(Mn^(2+))generated from metallurgical,steel making and chemical industries enters sewage treatment plants and affects the sludge activity and flocculation.The effect of Mn^(2+)on the removal of chemical oxygen demand(COD)and total phosphorus(TP)and sludge activity were investigated in anoxic zone of an anaerobic/anoxic/oxic(A2O)process.The compositions and structures of extracellular polymeric substances(EPS)were charac-terized using three-dimensional excitation emission matrix fluorescence spectroscopy(3D-EEM),X-ray photoelectron spectroscopy(XPS)and Fourier transform infrared spectroscopy(FTIR)to reveal the relationship among Mn^(2+),EPS and sludge flocculation.The results showed thatlow concentration of Mn^(2+)(10 mg/L)hindered microbial flocculation and reduced removal efficiencies of the pollutants.When Mn^(2+)was 5 mg/L,removal efficiencies of COD and TP reached 65%and 90%,respectively.Sludge flocculation was the best and SVI was 70.56 mL//g.The changes of Mn^(2+)concentration caused deviation of groups'compositions in LB-EPS and TB-EPS,where the main components were always protein(PN)and polysaccharide(PS).The addition of Mn^(2+)resulted in the degradation of humic acids.However,it did not give rise to significant morphology changes of EPS.

Differing from other subtypes of inhibitory interneuron,chandelier or axo-axonic cells form depolarizing GABAergic synapses exclusively onto the axon initial segment(AIS)of targeted pyramidal cells(PCs).However,the debate whether these AIS-GABAergic inputs produce excitation or inhibition in neuronal processing is not resolved.Using realistic NEURON modeling and electrophysiological recording of cortical layer-5 PCs,we quantitatively demonstrate that the onset-timing of AIS-GABAergic input,relative to dendritic excitatory glutamatergic inputs,determines its bi-directional regulation of the efficacy of synaptic integration and spike generation in a PC.More specifically,AIS-GABAergic inputs promote the boosting effect of voltage-activated Na+channels on summed synaptic excitation when they precede glutamatergic inputs by>15 ms,while for nearly concurrent excitatory inputs,they primarily produce a shunting inhibition at the AIS.Thus,our findings offer an integrative mechanism by which AIS-targeting interneurons exert sophisticated regulation of the input-output function in targeted PCs.

In this paper, we introduce the notion of a generalized weak group-like algebra which is designed to cover finite groupoid algebras and group-like algebras. Then we construct a list of examples and show that a generalized weak group-like algebra can be regarded as a generalized weak bi-Frobenius algebra. Moreover, we discuss when a generalized weak bi-Frobenius algebra is a groupoid algebra. Finally, we give the classifications of low-dimensional generalized weak group-like algebras.

Chromatographic fingerprinting has been perceived as an essential tool for assessing quality and chemical equivalence of traditional Chinese medicine.However,this pattern-oriented approach still has some weak points in terms of chemical coverage and robustness.In this work,we proposed a multiple reaction monitoring(MRM)-based fingerprinting method in which approximately 100 constituents were simultaneously detected for quality assessment.The derivative MRM approach was employed to rapidly design MRM transitions independent of chemical standards,based on which the large-scale fingerprinting method was efficiently established.This approach was exemplified on QiShenYiQi Pill(QSYQ),a traditional Chinese medicine-derived drug product,and its robustness was systematically evaluated by four indices:clustering analysis by principal component analysis,similarity analysis by the congruence coefficient,the number of separated peaks,and the peak area proportion of separated peaks.Compared with conventional ultraviolet-based fingerprints,the MRM fingerprints provided not only better discriminatory capacity for the tested normal/abnormal QSYQ samples,but also higher robustness under different chromatographic conditions(i.e.,flow rate,apparent pH,column temperature,and column).The result also showed for such large-scale fingerprints including a large number of peaks,the angle cosine measure after min-max normalization was more suitable for setting a decision criterion than the unnormalized algorithm.This proof-of-concept application gives evidence that combining MRM technique with proper similarity analysis metrices can provide a highly sensitive,robust and comprehensive analytical approach for quality assessment of traditional Chinese medicine.

The capacity for neurogenesis in the adult mammalian brain is extremely limited and highly restricted to a few regions,which greatly hampers neuronal regeneration and functional restoration after neuronal loss caused by injury or disease.Meanwhile,transplantation of exogenous neuronal stem cells into the brain encounters several serious issues including immune rejection and the risk of tumorigenesis.Recent discoveries of direct reprogramming of endogenous glial cells into functional neurons have provided new opportunities for adult neuro-regeneration.Here,we extensively review the experimental findings of the direct conversion of glial cells to neurons in vitro and in vivo and discuss the remaining issues and challenges related to the glial subtypes and the specificity and efficiency of direct cell-reprograming,as well as the influence of the microenvironment.Although in situ glial cell reprogramming offers great potential for neuronal repair in the injured or diseased brain,it still needs a large amount of research to pave the way to therapeutic application.

Performance and mechanism of La-Fe metal-organic framework as a highly efficient adsorbent for fluoride removal from mine water

Water fluoride pollution has caused non-negligible harm to the environment and humans,and thus it is crucial to find a suitable treatment technology.In this study,La-Fe@PTA adsorbent was synthesized for the defluoridation of mine water.The results showed that the optimum conditions for defluoridation by La-Fe@PTA were p H close to 7.0,the initial F-concentration of 10 mg/L,the dosage of 0.5 g/L and the adsorption time of 240 min.Compared with SO_4^(2-),Cl^(-),NO_(3)^(-),Ca^(2+)and Mg^(2+),CO_(3)^(2-)and HCO_(3)^(-)presented severer inhibition on fluoride uptake by La-Fe@PTA.The adsorption process fits well with the pseudo-second-order kinetic model and Freundlich model,and the maximum adsorption capacity of Langmuir model was 95 mg/g.Fixed-bed adsorption results indicated that fluoride in practical fluorinated mine water could be effectively removed from 3.6 mg/L to less than 1.5 mg/L within130 bed volume(BV)by using 1.5 g La-Fe@PTA.Furthermore,the adsorbent still had good adsorption capacity after regeneration,which confirms the great application potential of La-Fe@PTA as a fluoride ion adsorbent.The mechanism analysis showed that La-Fe@PTA adsorption of fluorine ions is a physicochemical reaction driven by electrostatic attraction and ion exchange.

In this paper, we introduce the notion of the Hom-Leibniz-Rinehart algebra as an algebraic analogue of Hom-Leibniz algebroid, and prove that such an arbitrary split regular Hom-Leibniz-Rinehart algebra L is of the form L=U+∑Iwith U a subspace of a maximal abelian subalgebra H and any I, a well described ideal of L, satisfying [I, I] = 0 if [γ]≠[δ].In the sequel, we develop techniques of connections of roots and weights for split Hom-LeibnizRinehart algebras, respectively. Finally, we study the structures of tight split regular Hom-Leibniz-Rinehart algebras.
