Polypyrimidine Tract-Binding Protein Enhances Zika Virus Translation by Binding to the 5'UTR of Internal Ribosomal Entry Site

Objectives To identify the 5'untranslated region of Zika virus(ZIKV 5'UTR)RNA-binding proteins and to investigate the impact of the binding protein on the activity of internal ribosomal entry site(IRES)located in ZIKV 5'UTR and virus *** Interacting proteins in U251 cells were captured using tRSA-tagged ZIKV 5'UTR RNA and tRSA-ZIKV 5'UTR RNA-binding proteins were visualized by SDS-PAGE silver staining,Subsequently,liquid chromatographytandem mass spectrometry(LC-MS/MS),bioinformatics analysis,and Western blot were used to identify the candidate proteins binding to ZIKV 5'*** expression assay and plaque forming assay were performed to analyze the effect of the binding protein on ZIKV IRES activity and ZIKV production,*** tRSA RNA pull-down assay,LC-MS/MS,and Western blot analysis showed that polypyrimidine tractbinding protein(PTB)bound to the ZIKV 5'***,dual luciferase reporter assay revealed that overexpression of PTB significantly enhanced the IRES activity of ZIKV(t=10.220,P<0.001),while PTB knockdown had the opposite effect(t=4.897,P<0.01).Additionally,virus plaque forming assay demonstrated that up-regulation of PTB expression significantly enhanced viral titer(t=6.400,P<0.01),whereas reducing PTB expression level weakened virus infectivity(t=5.055,P<0.01).Conclusion PTB positively interacts with the ZIKV 5'UTR and enhances IRES activity and virus production.

Ribotrap Analysis of Proteins Associated with FHL3 3'Untranslated Region in Glioma Cells

Objective To screen the proteins associated with four-and-a-half LIM domains 3(FHL3) 3' untranslated region(3'UTR) in glioma cells. Methods Western blot was adopted to detect the regulatory effect of poly(C)-binding protein 2(PCBP2) on FHL3. Biotin pull-down and sliver staining were employed to screen and verify the candidate binding proteins of FHL3 3'UTR. Then liquid chromatography-tandem mass spectrometry(LC-MS/MS) and molecule annotation system were used to identify and analyze the candidate binding proteins. Immunoprecipitation was conducted to study the interaction between PCBP2 and polypyrimidine tract-binding protein 1(PTBP1), a binding protein identified by LC-MS/MS. Results PCBP2 could bind to FHL3 mRNA 3'UTR-A and inhibited the expression of FHL3 in T98 G glioms cells. 22 candidate binding proteins were identified. Among them, there were 11 RNA binding proteins, including PCBP2. PTBP1 associated with FHL3 mRNA 3'UTR and interacted with PCBP2 protein. Conclusion PCBP2 and PTBP1 can both associate with FHL3 mRNA 3'UTR through forming a protein complex.

Screening of Substrates of Protein Arginine Methyltransferase 1 in Glioma

Objective To screen the asymmetric dimethyl arginines (ADMA)-containing proteins which could combine with protein arginine methyltransferase 1 (PRMT1). Methods Western blot was adopted to identify the expression of PRMT1 and the proteins with ADMA in glioma cell lines and normal brain tissues, and then to detect the changes of ADMA level after knock-down of PRMT1 with RNAi transfection in U87MG cells. Co-Immunoprecipitation (Co-IP), western blot, and sliver staining were employed to screen the candidate binding proteins of PRMT1. Then liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to identify the binding proteins of PRMT1. Results The expression of PRMT1 and some levels of ADMA were higher in glioma cell lines than in normal brain tissues. After knocking down PRMT1, some ADMA levels were found declined. After screening the binding proteins of PRMT1 with Co-IP and LC-MS/MS, 26 candidate binding proteins were identified. Among them, 6 candidate proteins had higher ions scores (>38) and bioinformation analysis predicted that SEC23-IP, ANKHD1-EIF4EBP3 protein, and 1-phosphatidylinositol-3-phosphate 5-kinase isoform 2 had possible methylated aginine sites. Conclusions The high expression of PRMT1 in glioma may induce the change of ADMA levels. Altogether 26 candidate proteins were identified, which contain ADMA and specifically bind with PRMT1.

APFA:Ameliorated Pathfinder Algorithm for Engineering Applications

Pathfinder algorithm(PFA)is a swarm intelligent optimization algorithm inspired by the collective activity behavior of swarm animals,imitating the leader in the population to guide followers in finding the best food *** algorithm has the characteristics of a simple structure and high ***,PFA faces challenges such as insufficient population diversity and susceptibility to local optima due to its inability to effectively balance the exploration and exploitation *** paper proposes an Ameliorated Pathfinder Algorithm called APFA to solve complex engineering optimization ***,a guidance mechanism based on multiple elite individuals is presented to enhance the global search capability of the ***,to improve the exploration efficiency of the algorithm,the Logistic chaos mapping is introduced to help the algorithm find more high-quality potential solutions while avoiding the worst ***,a comprehensive following strategy is designed to avoid the algorithm falling into local optima and further improve the convergence *** three strategies achieve an effective balance between exploration and exploitation overall,thus improving the optimization performance of the *** performance evaluation,APFA is validated by the CEC2022 benchmark test set and five engineering optimization problems,and compared with the state-of-the-art metaheuristic *** numerical experimental results demonstrated the superiority of APFA.

An Efficient Multi-objective Approach Based on Golden Jackal Search for Dynamic Economic Emission Dispatch

Dynamic Economic Emission Dispatch(DEED)aims to optimize control over fuel cost and pollution emission,two conflicting objectives,by scheduling the output power of various units at specific *** many methods well-performed on the DEED problem,most of them fail to achieve expected results in practice due to a lack of effective trade-off mechanisms between the convergence and diversity of non-dominated optimal dispatching *** address this issue,a new multi-objective solver called Multi-Objective Golden Jackal Optimization(MOGJO)algorithm is proposed to cope with the DEED *** proposed algorithm first stores non-dominated optimal solutions found so far into an ***,it chooses the best dispatching solution from the archive as the leader through a selection mechanism designed based on elite selection strategy and Euclidean distance index *** mechanism can guide the algorithm to search for better dispatching solutions in the direction of reducing fuel costs and pollutant ***,the basic golden jackal optimization algorithm has the drawback of insufficient search,which hinders its ability to effectively discover more Pareto *** this end,a non-linear control parameter based on the cosine function is introduced to enhance global exploration of the dispatching space,thus improving the efficiency of finding the optimal dispatching *** proposed MOGJO is evaluated on the latest CEC benchmark test functions,and its superiority over the state-of-the-art multi-objective optimizers is highlighted by performance ***,empirical results on 5-unit,10-unit,IEEE 30-bus,and 30-unit systems show that the MOGJO can provide competitive compromise scheduling solutions compared to published DEED ***,in the analysis of the Pareto dominance relationship and the Euclidean distance index,the optimal dispatching solutions provided by MOGJO are the closest to the ideal solutions for minimizing fuel costs

Optical detection method of discharge mode transition of inductively coupled plasma in an atmosphere-breathing electric propulsion system

Plasma discharge stability is an important problem in atmosphere-breathing electric propulsion system when maintaining long-term missions at ultra-low earth *** paper designed an inductively coupled plasma source to imitate the ionization *** effect of inflow rate and Radio Frequency(RF)power on the plasma discharge mode transition is experimentally studied.A discharge mode detection method is proposed,which determines the discharge mode by identifying the morphology of the plasma *** using the method,the discharge mode transition is quantified and a control model based on the parameter sensitivity is *** verify the method,the spectra are measured and the electron temperature spatial distribution is *** the method has been proven *** results show that the inductively coupled discharge contains capacitive components affected by the mass flow rate and the radio frequency *** plasma characteristics can be maintained stably by controlling the radio frequency power when the mass flow rate randomly changes in a certain *** is demonstrated that the application of detection method effectively identifies the discharge mode,which is a promising active control method for the plasma discharge mode.

Chiral hybrid metal halides with high photoluminescence quantum yield(PLQY)and superior dissymmetry factor(g_(lum))are promising candidates for circularly polarized luminescence(CPL)emitting ***,it is a great challenge to achieve both high g_(lum)and large ***,we report a new supramolecular strategy to fabricate chiral hybrid Cu(Ⅰ)-halides with nearunity quantum yields and intense CPL *** dissymmetry factor can be dramatically modulated by tuning the hostguest *** to the chirality and variable conformations of the host-guest complexes,a series of chiral[G@L^(R/S)]_(2)Cu_(4)I_(6)have been *** incorporation of different cations within the chiral L^(R/S)hosts leads to variations of packing modes along the c-axis and different extent of distortions in the[Cu_(4)I_(6)]_(2)-*** them,[H_(3)O@L^(R/S)]+cations are stacked based on Van der Waals interactions and hierarchically self-assemble into a right/left-handed helix in[H_(3)O@L^(R/S)]_(2)Cu_(4)I_(6),which results in a high g_(lum)of-7×10^(-3)and a large PLQY of 96.92%.Notably,[G@L^(R/S)]_(2)Cu_(4)I_(6)shows promising applications in single-component white light emitting diodes(WLEDs),exhibiting a maximum dissymmetry factor of-1.2×10^(-3).The effective host-guest complexation and oriented hierarchical self-assembly provide an effective strategy for the development of high-performance CPL materials.
