Correlation between gut microbiota and glucagon-like peptide-1 in patients with gestational diabetes mellitus

BACKGROUND Gestational diabetes mellitus(GDM)places both the mother and offspring at high risk of *** evidence suggests that the gut microbiota plays a role in the pathogenesis of ***,it is still unclear whether the gut microbiota is related to blood biochemical traits,particularly glucagon-like peptide-1(GLP-1),in GDM *** To explore the correlation between the gut microbiota and blood biochemical traits,particularly GLP-1,in GDM *** The V4 region of the 16S ribosomal ribonucleic acid(rRNA)gene was sequenced based on the fecal samples of 35 pregnant women with GDM and was compared to that of 25 pregnant women with normal glucose tolerance(NGT).RESULTS The results showed that Ruminococcaceae_UCG-002,Ruminococcaceae_UCG-005,Clostridium_sensu_stricto_1,and Streptococcus were more abundant in the NGT group than in the GDM *** and Lachnoclostridium were more abundant in the GDM group than in the NGT ***’s correlation analysis was performed to identify the relationships between microbiota genera and blood biochemical ***,Roseburia,Faecalibacterium,and Ruminococcaceae_UCG-002 were significantly negatively correlated with ***_UCG-002 was significantly negatively correlated with hemoglobin *** was significantly positively correlated with ***,Oscillibacter,and Bifidobacterium were significantly positively correlated with GLP-1.A random forest model showed that 20 specific genera plus glucose provided the best discriminatory power,as indicated by the area under the receiver operating characteristic curve(0.94).CONCLUSION The results of this study reveal novel relationships between the gut microbiome,blood biochemical traits,particularly GLP-1,and GDM *** findings suggest that some genera are crucial for controlling blood glucose-related indices and may be beneficial for GDM *** in the microbial composition of the

Multiple synchronous anorectal melanomas with different colors: A case report

BACKGROUND Anorectal melanoma (AM) is an extremely rare malignant tumor originating from anorectal melanocytes with a poor prognosis. AM has been reported to have a much lower incidence than cutaneous or choroid melanoma, accounting for 0.4%-1.6% of all melanomas. CASE SUMMARY We report a 76-year-old female patient diagnosed with anorectal malignant melanoma by colonoscopy and biopsy. Intraoperative examination revealed two distinct anorectal tumors, one melanotic and another amelanotic, as well as two pigmented mucosal zones at the dentate line level. Abdominal perineal resection was performed. A pathological report confirmed all four lesions to be melanomas. Postoperatively, we followed an immunotherapy protocol targeting PD-1 (nivolumab). The patient had 24 mo of disease-free follow-up upon completion of nivolumab treatment. CONCLUSION This is the first reported case presenting coexistence of pigmented and unpigmented AMs in the same patient.

Anterior segment OCT application in quantifying posterior capsule opacification severity with varied intraocular lens designs

Claviceps purpurea produces many pharmacologically important ergot alkaloids(EAS),which are widely used to treat migraine and hypertension and to aid *** an EAS biosynthetic cluster of *** has been discovered more than 20 years ago,the complete biosynthetic pathway of EAS has not been fully characterized until *** main obstacle to elucidating this pathway and strain modification is the lack of efficient genome-editing tools for *** conventional gene manipulation method for *** relies on homologous recombination(HR),although the efficiency of HR in *** is very low(~1-5%).Consequently,the disruption of target genes is laborious and *** CRISPR/Cas9 genome-editing methods based on in vivo Cas9 expression and gRNA transcription have been reported recently,their gene-disruption efficiency is still very ***,we developed an efficient genome-editing system in *** based on in vitro assembled CRISPR/Cas9 gRNA ribonucleoprotein *** proof of principle,three target genes were efficiently knocked out using this CRISPR/Cas9 ribonucleoprotein complex-mediated HR system,with editing efficiencies ranging from 50%to 100%.Inactivation of the three genes,which are closely related to uridine biosynthesis(ura5),hypha morphology(rac),and EAS production(easA),resulted in a uridine auxotrophic mutant,a mutant with a drastically different phenotype in axenic culture,and a mutant that did not produce EAS,*** ribonucleoprotein-based genome-editing system has a great advantage over conventional and in vivo CRISPR/Cas9 methods for genome editing in ***,which will greatly facilitate elucidation of the EAS biosynthetic pathway and other future basic and applied research on ***.

Multivariate Analysis of Prognosis in Patients with Pancreatic Cancer

Objective Pancreatic cancer is a highly malignant digestive system tumor. The prognosis of pancreatic cancer may be affected by many factors. But there is a lack of research on the multivariate analysis of prognostic factors of pancreatic cancer. This study aimed to explore the factors that multivariate influence the prognosis of pancreatic cancer. Methods We conducted a retrospective cohort study using electronic medical record database from The First Hospital of Jilin University from January 2014 to December 2016. Clinical information, such as TNM stage, serum tumor markers, serum albumin and the patient-generated subjective global assessment (PG-SGA), were collected. Kaplan- Meier method was used to calculate survival rate. The survival curve was compared with the Log-rank test. A multivariate analysis was performed for each prognostic factor using the Cox proportional hazards model. The significance level was 0.05. Results 1. The median overall survival time (OS) of 493 patients was 10 months. The 1-year, 2-year, and 3-year cumulative survival rates were 41.5%, 20.6%, and 9.9%, respectively. Univariate survival analysis revealed that TNM stage (P < 0.001), treatment regimen (P < 0.001), CA125 (P < 0.001), the level of serum albumin (P = 0.014), and nutritional status (P = 0.006) were significantly correlated with prognosis of pancreatic cancer. COX proportional hazards regression model showed that TNM stage (P = 0.038), Carbonhydrate antigen 125 (CA125)(P = 0.003) and the level of serum albumin were independent factors affecting the median OS. The prognosis of patients with Ⅳ stage, elevated levels of serum albumin group and elevated levels of CA125 group were poor. 2. Seventy-three patients with pancreatic cancer underwent surgical resection. The median postoperative survival time was 28 months. The 1-year, 2-year, and 3-year cumulative survival rates were 74.4%, 47.7%, and 21.4%, respectively. Univariate analysis revealed that TNM stage (P = 0.032), pathologic

Review of Iron-Based Catalysts for Carbon Dioxide Fischer-Tropsch Synthesis

Capturing and utilizing CO_(2)from the production process is the key to solving the excessive CO_(2)emission problem. CO_(2)hydrogenation with green hydrogen to produce olefins is an effective and promising way to utilize CO_(2)and produce valuable chemicals. The olefins can be produced by CO_(2)hydrogenation through two routes, i.e., CO_(2)-FTS (carbon dioxide Fischer- Tropsch synthesis) and MeOH (methanol-mediated), among which CO_(2)-FTS has significant advantages over MeOH in practical applications due to its relatively high CO_(2)conversion and low energy consumption potentials. However, the CO_(2)-FTS faces challenges of difficult CO_(2)activation and low olefins selectivity. Iron-based catalysts are promising for CO_(2)-FTS due to their dual functionality of catalyzing RWGS and CO-FTS reactions. This review summarizes the recent progress on iron-based catalysts for CO_(2)hydrogenation via the FTS route and analyzes the catalyst optimization from the perspectives of additives, active sites, and reaction mechanisms. Furthermore, we also outline principles and challenges for rational design of high-performance CO_(2)-FTS catalysts.
