Expression and location of α-fetoprotein during rat colon development

AIM： To investigate the expression of α-fetoprotein （AFP）, a cancer-associated fetal glycoprotein, and its involvement during rat colon development. METHODS： Colons from Sprague-Dawley rat fetuses, young and adult （8 wk old） animals were used in this study. Expression levels of AFP in colons of different development stage were detected by reversetranscriptase PCR （RT-PCR） and Western blotting. To identify the cell location of AFP in the developing rat colons, double-immunofluorescent staining was performed using antibodies to specific cell markers and AFP, respectively. RESULTS： The highest levels of AFP mRNA were detected in colons of rats at embryonic day 18.5 （e18.5）. Compared to e18.5 d, the AFP expression was significantly decreased during rat development （85% for e20.5, P 〈 0.05, 58% for postnatal day 0.5 （P〈0.5）, P 〈 0.05, 37% for P7, P 〈 0.05, 24% for P14, P 〈 0.05, and 11% for P21, P 〈 0.05） and undetected in adult rats. Only the 72-kDa isoform of AFP was detected by Western blotting, the expression pattern was similar to AFP mRNA and conformed to the results of mRNA expression. The AFP positive staining was identical to different distribution patterns in fetuses, young and adult animals and positive staining for both AFP and vimentin was overlapped in mesenchymal cells at each stage tested. CONCLUSION： This study has for the first time demonstrated that AFP is localized in the mesenchyme of rat colon from the embryo to the weaning stage by immunofluorescence and presents 72-kDa isoform in the developing rat colons by Western blotting. The dynamic expression of AFP in the various developmental stages of the colon indicates that AFP might be involved in many aspects of colon development.

Micro RNA transcriptome of skeletal muscle during yak development reveals that miR-652 regulates myoblasts differentiation and survival by targeting ISL1

The growth and development of skeletal muscle also determine the meat production of yak, ultimately affecting the economic benefits. Hence, improving growth performance is a top priority in the yak industry. Skeletal muscle development is a complex process involving the regulation of several genes, including microRNAs(miRNAs). However,the transcription of miRNAs in yak skeletal muscle during prenatal to postnatal stages is unknown. We used small RNA sequencing(small RNA-Seq) to determine the global miRNAs of longissimus dorsi muscle from yak(the samples were collected from three fetuses and three adults). Totally 264 differently expressed miRNAs(|log2(fold change)|>1and P-value≤0.05) were detected between the two groups. Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) analysis showed that differently expressed miRNAs-targeted genes participated in pathways associated with muscle development, such as MAPK, PI3K-Akt, and Hippo signaling pathways, etc. MiR-652, which was up-regulated in the fetal group, was transfected into C2C12 myoblasts to examine its role. miR-652 promoted(P≤0.05)proliferation and differentiation, but inhibited(P≤0.001) apoptosis at early period. Furthermore, miR-652 reduced(P≤0.001) the proportion of C2C12 myoblasts in the G1 phase while increasing(P≤0.01) the proportion of cells in the S and G2 phases. Dual-luciferase reporter assays indicated that ISL1 served as a target of miR-652. In general, these findings expand our understanding of yak skeletal muscle miRNAs, and suggested that miR-652 probably regulated myogenesis by regulating ISL1.

Sustained Akt signaling in articular chondrocytes causes osteoarthritis via oxidative stress-induced senescence in mice

Osteoarthritis(OA) is an age-related disorder that is strongly associated with chondrocyte senescence. The causal link between disruptive PTEN/Akt signaling and chondrocyte senescence and the underlying mechanism are unclear. In this study, we found activated Akt signaling in human OA cartilage as well as in a mouse OA model with surgical destabilization of the medial *** mouse models mimicking sustained Akt signaling in articular chondrocytes via PTEN deficiency driven by either Col2a1-Cre or Col2a1-Cre^(ERT2) developed OA, whereas restriction of Akt signaling reversed the OA phenotypes in PTEN-deficient ***, prolonged activation of Akt signaling caused an accumulation of reactive oxygen species and triggered chondrocyte senescence as well as a senescence-associated secretory phenotype, whereas chronic administration of the antioxidant N-acetylcysteine suppressed chondrocyte senescence and mitigated OA progression in PTEN-deficient mice. Therefore,inhibition of Akt signaling by PTEN is required for the maintenance of articular cartilage. Disrupted Akt signaling in articular chondrocytes triggers oxidative stress-induced chondrocyte senescence and causes OA.

Truncated Smoothing Newton Method for l_∞ Fitting Rotated Cones

In this paper, the rotated cone fitting problem is considered. In case the measured data are generally accurate and it is needed to fit the surface within expected error bound, it is more appropriate to use l∞ norm than 12 norm. l∞ fitting rotated cones need to minimize, under some bound constraints, the maximum function of some nonsmooth functions involving both absolute value and square root functions. Although this is a low dimensional problem, in some practical application, it is needed to fitting large amount of cones repeatedly, moreover, when large amount of measured data are to be fitted to one rotated cone, the number of components in the maximum function is large. So it is necessary to develop efficient solution methods. To solve such optimization problems efficiently, a truncated smoothing Newton method is presented. At first, combining aggregate smoothing technique to the maximum function as well as the absolute value function and a smoothing function to the square root function, a monotonic and uniform smooth approximation to the objective function is constructed. Using the smooth approximation, a smoothing Newton method can be used to solve the problem. Then, to reduce the computation cost, a truncated aggregate smoothing technique is applied to give the truncated smoothing Newton method, such that only a small subset of component functions are aggregated in each iteration point and hence the computation cost is considerably reduced.

What causes the absence of pulsations in Central Compact Objects in supernova remnants?

Most young neutron stars belonging to the class of Central Compact Objects(CCOs)in supernova remnants do not have known *** investigated seven such CCOs to understand the common reasons for the absence of detected *** use of XMM-Newton,Chandra,and NICER observations,we perform a systematic timing and spectral analysis to derive updated sensitivity limits for both periodic signals and multi-temperature spectral components that could be associated with radiation from hotspots on the neutron star *** on these limits,we then investigated for each target the allowed viewing geometry that could explain the lack of *** find that it is unlikely(<10-6)to attribute that we do not see pulsations to an unfavorable viewing geometry for five considered ***,the carbon atmosphere model,which assumes homogeneous temperature distribution on the surface,describes the spectra equally well and provides a reasonable interpretation for the absence of detected periodicities within current *** unusual properties of CCOs with respect to other young neutron stars could suggest a different evolutionary path,as that proposed for sources experiencing episodes of significant fallback accretion after the supernova event.

Microstructure Evolution and Strain Softening of Carbon Black Filled Natural Rubber Vulcanizates
