Approach of water-salt regulation using micro-sprinkler irrigation in two coastal saline soils

This study aimed to investigate whether saline silt and sandy loam coastal soils could be reclaimed by micro-sprinkler irrigation.The experiments were run using moderately salt-tolerant tall fescue grass.Micro-sprinkler irrigation in three stages was used to regulate soil matric potential at a 20-cm soil depth.Continued regulation of soil water and salt through micro-sprinkler irrigation consistently resulted in an increasingly large low-salinity region.The application of the three stages of soil wateresalt regulation resulted in an absence of salt accumulation throughout the soil profile and the conversion of highly saline soils into moderately saline soils.There were increases in the plant height,leaf width,leaf length,and tiller numbers of tall fescue throughout the leaching process.The results showed that micro-sprinkler irrigation in three soil water and salt regulation stages can be used to successfully cultivate tall festuca in highly saline coastal soil.This approach achieved better effects in sandy loam soil than in silt soil.Tall fescue showed greater survival rates in sandy loam soil due to the rapid reclamation process,whereas plant growth was higher in silt soil because of effective water conservation.In sandy loam,soil moisture should be maintained during soil reclamation,and in silt soil,soil root-zone environments optimal for the emergence of plants should be quickly established.Micro-sprinkler irrigation can be successfully applied to the cultivation of tall fescue in coastal heavy saline soils under a three-stage soil wateresalt regulation regime.

Preserving GABAergic interneurons in acute brain slices of mice using the N-methyl-D-glucamine-based artificial cerebrospinal fluid method

Defects in the function and development of GABAergic interneurons have been linked to psychiatric disorders, so preservation of these interneurons in brain slices is important for successful electrophysiological recording in various ex vivo methods. However, it is difficult to maintain the activity and morphology of neurons in slices from mice of 〉30 days old. Here we evaluated the N-methyI-D-glucamine （NMDG）- based artificial cerebrospinal fluid （aCSF） method for the preservation of interneurons in slices from mice of up to -6 months old and discussed the steps that may affect their quality during slicing. We found that the NMDG-aCSF method rescued more cells than sucrose-aCSF and successfully preserved different types of interneurons including parvalbumin- and somatostatin-positive interneurons. In addition, both the chemical and electrical synaptic signaling of interneurons were maintained. These results demonstrate that the NMDG-aCSF method is suitable for the preservation of interneurons, especially in studies of gap junctions.

Streptococcus suis(S. suis) is a Gram-positive zoonotic pathogen. S. suis infection in humans commonly causes meningitis, septicemia, arthritis,and streptococcal toxic shock-like syndrome(STSLS). S. suis has 29 serotypes, of which S. suis serotype 2(SS2) has the highest clinical isolation rate and strongest pathogenicity and causes most S. suis human infections.About 1,000 different sequence types(STs)were identified in S.suis based on multilocus sequence typing(MLST).Among these,STs,ST1,and ST7 belong to serotype 2 and are the major hazards of S.suis.in 1998 and 2005,two cases of SS2 human infection outbreak were reported in Jiangsu and Sichuan,China,causing 14 deaths and 38 deaths,respectively.

Facile preparation of methylcellulose/poly(vinyl alcohol)physical complex hydrogels with tunable thermosensitivity

Novel complex hydrogels of methylcellulose （MC） and poly（vinyl alcohol）（PVA） with wide-spectrum thermoresponsivity were prepared via physical and mild process. Thermal phase transition of MC/PVA hydrogels exhibited two forms including sol/sol to gel/sol and sol/gel to gel/gel. The phase transition temperature of MC/PVA solution ranged from 38,7 to 60.6 ℃ and was able to be adjusted by simply changing the feeding ratios of two components. The interior morphology of MC/PVA gels was examined with fluorescence analysis and scanning electron microscopy analysis, which showed that MC was well dispersed in matrix before and after thermally gelling.

Efficacy and Safety of Bortezomib in Multiple Myeloma Patients with Hepatitis B： A Multicenter Retrospective Study

Background： The efficacy and safety evidence of bortezomib in multiple myeloma （MM） patients with hepatitis B is vacant.This study aimed to investigate the efficacy and safety of bortezomib in MM patients with hepatitis B in China.Methods： From 2006 to 2011, 739 newly diagnosed MM patients were screened for serum hepatitis B virus （HBV） biomarkers.HBV-infected patients were followed for HBV reactivation by monitoring of serum alanine transaminase （ALT） and HBV DNA load.The pattern of HBV reactivation in relation to bortezomib was evaluated.Seven hundred thirty-nine MM patients were included in this study.Results： The prevalence of MM patients infected with HBV was 3.4% （n =25）, of which 17 cases were treated with bortezomib.Bortezomib had no significant influence on liver function （ALT before and after treatment： 36.69 ＆#177; 8.90 U/L vs.11.31 ＆#177; 2.74 U/L, P =0.19） and HBV DNA of MM patients with HBV （detectable HBV DNA percentage： 5.9% vs.11.8%, P =0.12）.Conclusions： Bortezomib can be used safely and effectively in MM patients with hepatitis B.HBV prophylaxis and surveillance are recommended during the MM treatment.

In recent years,forest fires and maritime accidents have occurred frequently,which have had a bad impact on human production and life.Thus,the development of seaplanes is an increasingly urgent demand.It is important to study the taxiing process of seaplanes for the development of seaplanes,which is a strong nonlinear fluid-structure interaction problem.In this paper,the smoothed particle hydrodynamics(SPH)method based on the Lagrangian framework is utilized to simulate the taxiing process of seaplanes,and the SPH results are compared with those of the finite volume method(FVM)based on the Eulerian method.The results show that the SPH method can not only give the same accuracy as the FVM but also have a strong ability to capture the splashing waves in the taxiing process,which is quite meaningful for the subsequent study of the effect of a splash on other parts of the seaplane.

Mechanism of Shengmai Injection on Anti-Sepsis and Protective Activities of Intestinal Mucosal Barrier in Mice

Objective:To study the mechanism of Shengmai Injection(SMI)on anti-sepsis and protective activities of intestinal mucosal barrier.Methods:The contents of 11 active components of SMI including ginsenoside Rb1,Rb2,Rb3,Rd,Re,Rf,Rg1,Rg2,ophioposide D,schisandrol A and schisantherin A were determined using ultra-performance liquid chromatography.Fifty mice were randomly divided into the blank,the model,the low-,medium-and high-dose SMI groups(0.375,0.75,1.5 mL/kg,respectively)by random number table,10 mice in each group.On SMI group,SMI was administrated to mice daily via tail vein injection for 3 consecutive days,while the mice in the blank and model groups were given 0.1 mL of normal saline.One hour after the last SMI administration,except the blank group,the mice in other groups were intraperitoneally injected with lipopolysaccharide(LPS)saline solution(2 mL/kg)at a dosage of 5 mL/kg for development of endotoxemia mice model.The mice in the blank group were given the same volume of normal saline.Inflammatory factors including interferon-γ(INF-γ),tumor necrosis factor-α(TNF-α),interleukin(IL)-2 and IL-10 were measured by flow cytometry.Myosin light-chain kinase(MLCK),nuclear factorκB(NF-κB)levels,and change of Occludin proteins in jejunum samples were analyzed by Western blot.Results:The decreasing trends of INF-γ,TNF-α and IL-2 were found in serum of SMI treatment groups.In SMI-treated mice,the content of Occludin increased and MLCK protein decreased compared with the model group(P0.05).Conclusion:SMI may exert its anti-sepsis activity mainly through NF-κB-pro-inflammatory factor-MLCK-TJ cascade.

碳渣缺陷是消失模铸造工艺生产铸铁件时易出现而又较难于解决的问题。为了解决这一缺陷,将传统空腔铸造生产中常用的溢流集渣冒口工艺移植到实型铸造中。生产实践证明,该工艺存在一定的局限性,同时由于集渣块尺寸的不合理,反而会产生缩孔缩陷等缺陷。优化工艺参数,才是解决碳渣缺陷的最有效手段。

Long-term outcomes after hepatectomy of huge hepatocellular carcinoma: A single-center experience in China

Background: Currently, hepatectomy remains the first-line therapy for hepatocellular carcinoma(HCC). However, surgery for patients with huge( > 10 cm) HCCs is controversial. This retrospective study aimed to explore long-term survival after hepatectomy for patients with huge HCC. Methods: The records of 188 patients with pathologically confirmed HCC who underwent curative hepatectomy between 2007 and 2017 were reviewed;patients were divided into three groups according to tumor size: huge( > 10 cm;n = 84), large(5–10 cm;n = 51) and small( 10 cm) had significant effect on OS, and postoperative antiviral therapy and postoperative complications also had significant effects on DFS. Conclusions: Huge HCC is not a contraindication of hepatectomy. Although most of these patients experienced recurrence after surgery, OS and DFS were not significantly different from those of patients with large HCC after resection.

HSP90 and SIRT3 expression in hepatocellular carcinoma and their effect on invasive capability of human hepatocellular carcinoma cells

Objective:To vexplore expression of HSP90,SIRT3 in liver cancer tissue and its effect on liver cancer cell invasion ability.Methods:Moderate expression of HSP90 in SMMC-7721,HepG2,LO2 and Hep-3B cell lines were screened,which was validated by RT-PCR.Overexpression of HSP90 cell line and lentivirus packaging HSP90-RNAi were established,which was validated by RT-PCR and western blot.The level of epithelial-mesenchymal transition(EMT)related gene was detected by western blot.The percentage of cancer stem cells was assayed by flow cytometry.Results:RT-PCR demonstrated the highest expression of HSP90mRNA in SMMC-7721 cells,the lowest expression of HSP90 mRNA in Hep3B and LO2 and the moderate expression of HSP90 mRNA in Hep-G2.Therefore,HepG2 was selected as a follow-up experiment cell lines.Compared with the blank control group,expression of HSP90in HSP overexpression group was increased obviously,and expression of HSP90 in HSP90shRNA group was significantly decreased,which indicated successful establishment of HSP overexpression and shRNA group.The apoptotic cell in hsp-siRNA group was higher than the blank control group,while the HSP overexpression group showed opposite results.Western blot results showed transfection HSP promoted cells EMT transformation,up-regulated the level of E-cadherin,and down-regulated the level of Vimentin;meanwhile,shRNA group showed opposite results.Conclusions:Carcinoma HepG2 cell transfeeted high expression of HSP can promote the transformation of EMT,improve the expression of Vimentin,reduce the expression of E-cadherin,and inhibit apoptosis of cancer stem cells,which improve the invasive ability of cancer of the liver cells.While hsp-siRNA group presents opposite results.In summary,the expression of HSP is closely related to the occurrence,development and invasion of cancer of the liver tissue.
