Combination of Recombinant Adenovirus-p53 with Radiochemotherapy in Unresectable Pancreatic Carcinoma

Objective：To assess the safety and efficacy of the combination of recombinant adenovirus-p53 （rAd-p53） with radiochemotherapy for treating unresectable pancreatic ***：The eligible patients received concurrent rAd-p53 intratumoral injection and *** injection of rAd-p53 was guided by B *** consisted of intensity-modulated radiotherapy （IMRT） at two dose levels and intravenous gemcitabine （Gem）.For radiotherapy,gross target volume （GTV） and clinical target volume （CTV） were 55-60 Gy and 45-55 Gy in 25-30 fractions,*** intravenous gemcitabine was administered at 350 mg/m2,weekly,for 6 *** primary end points included toxicity,clinical benefit response （CBR） and disease control rate （DCR）.The secondary end points included progression-free survival （PFS） and overall survival （OS）.Results：Fifteen eligible patients were *** patients （53.3%） were evaluated as CBR and 12 （80%） achieved *** median PFS and OS were 6.7 and 13.8 months,***-year PFS and OS were 40.0% and 51.1%,*** were 8 （53.3%） patients reported grade 3 toxicities including neutropenia （6 patients,40%）,fever （1 patient,6.7%） and fatigue （1 patient,6.7%）.There was no grade 4 toxicity ***：Combination of rAd-p53 in unresectable pancreatic carcinoma showed encouraging efficacious benefit and was well ***-term follow-up is needed to confirm the improvement of PFS and OS.

Background The peritoneum response to peritoneal dialysis can lead to fibrosis. The transforming growth factor β1 （TGF-β1 ） plays a key role in regulating tissue repair and remodelling after injury. Connective tissue growth factor （CTGF）, a downstream mediator of TGF-β1 inducing fibrosis, has been implicated in peritoneal fibrosis. Vascular endothelial growth factor （VEGF） plays a key role in angiogenesis that can hasten peritoneal fibrosis. In this study, we investigated the effect of small interfering RNA （siRNA） of CTGF by pRETRO-SUPER （PRS） retrovirus vector on the expression of CTGF and VEGF in human peritoneal mesothelial cells. Methods Retrovirus producing CTGF siRNA were constructed from the inverted oligonucleotides and transferred into packaging cell line PT67 with lipofectamine, and the virus supernatant was used to infect human peritoneal mesothelial cell （HPMC）. The cells were divided into seven groups： low glucose DMEM, low glucose DMEM ＋ TGF-β1 5 ng/ml, low glucose DMEM ＋ TGF-β1 5 ng/ml ＋ PRS-CTGF-siRNA1-4 and low glucose DMEM ＋ TGF-β1 5 ng/ml ＋ PRS. The expression of CTGF and VEGF were measured by semiquantitative RT-PCR and Western blot. Results Low levels of CTGF and VEGF were detected in confluent HPMCs. Following stimulation with TGF-β1 , the levels of CTGF and VEGF were significantly upregulated （P〈0.01）. Introduction of PRS-CTGF-siRNA1-4 resulted in the significant reduction of CTGF mRNA and protein, and VEGF mRNA （P〈0.01）, especially in groups PRS-CTGF-siRNA, and PRS-CTGF-siRNA4. The introduction of PRS void vector did not have these effects （P〉0.05）. Conclusions The expression of CTGF siRNA mediated by PRS retrovirus vector can effectively reduce the level of CTGF and VEGF induced by TGF-β1 in cultured HPMCs. This study may provide potential therapeutic strategies to prevent the peritoneal fibrosis.

BACKGROUND Aplasia cutis congenita (ACC) in newborns is a condition in which congenital defects or hypoplasia is present in part of the epidermis,dermis and even subcutaneous tissue (including muscle and bones).First reported by Cordon in 1767,ACC is a rare disease with a low incidence of 1/100000 to 3/***,there are 500 cases reported *** can be accompanied by other *** onset mechanism of the disease remains unknown but is thought to be correlated to factors such as genetics,narrow uterus,foetal skin and amniotic membrane adhesion,use of teratogenic drugs in early pregnancy and viral *** SUMMARY In August 2018,we treated a newborn with ACC on the left lower limbs using a combination of ionic silver dressing and moist exposed burn ointment (MEBO) and achieved a satisfactory treatment *** skin defects were observed on the external genitals and on areas from the left foot to 3/4 of the upper left *** tissue and blood vessels were observed in the regions with skin *** following treatments were ***,the wound was rinsed with 0.9% sodium chloride solution followed by disinfection with povidone-iodine *** then MEBO was applied to the wound at a thickness of approximately 1 *** applying ionic silver dressing,the wound was covered with sterile *** wound dressing was replaced every 2-3 *** the 4-mo follow-up,the treatment outcome was *** was minimal scar tissue formation,and limb function was not *** The combination of ionic silver dressing and MEBO to ACC is helpful.

Efficient and precise photon-number-resolving detectors are essential for optical quantum information *** this,very few detectors have been able to distinguish photon numbers with both high fidelity and a large dynamic range,all while maintaining high speed and high timing *** nanostrip-based detectors excel at counting single photons efficiently and rapidly,but face challenges in balancing dynamic range and ***,we have pioneered the demonstration of 10 true photon-number resolution using a superconducting microstrip detector,with readout fidelity reaching an impressive 98%and 90%for 4-photon and 6-photon events,***,our proposed dual-channel timing setup drastically reduces the amount of data acquisition by 3 orders of magnitude,allowing for real-time photon-number *** then demonstrate the utility of our scheme by implementing a quantum random-number generator based on sampling the parity of a coherent state,which guarantees inherent unbiasedness,robustness against experimental imperfections and environmental noise,as well as invulnerability to *** solution boasts high fidelity,a large dynamic range,and real-time characterization for photon-number resolution and simplicity with respect to device structure,fabrication,and readout,which may provide a promising avenue towards optical quantum information science.

Photocatalytic CO_(2) reduction driven by solar light is a green approach that can decrease the greenhouse effect induced by high CO_(2) concentration in the atmosphere and generate carbon-based chemicals/fuels as *** this paper,non-metal co-catalysts ZnO/ZnS type-II hetero-junction nanoparticles with a rough surface were prepared through a hydrothermal *** used as a photocatalyst for CO_(2) reduction,the optimal one showed good cycle stability and a higher yield rate of 27.8µmol·g^(−1)·h^(−1) for CO_(2) conversion into *** outstanding catalytic activity originated from i)the rich interfaces between ZnO and ZnS in the nanoscale could significantly reduce the delivery path of carriers and improve the utilization efficiency of photo-excited electron/hole pairs and ii)enriched surface oxygen defects could supply much more reaction active sites for CO_(2) adsorption.

Quantized feedback control is fundamental to system synthesis with limited communication *** sharp contrast to the existing literature on quantized control which requires an explicit dynamical model,the authors study the quadratic stabilization and performance control problems with logarithmically quantized feedback in a direct data-driven framework,where the system state matrix is not exactly known and instead,belongs to an ambiguity set that is directly constructed from a finite number of noisy system *** this end,the authors firstly establish sufficient and necessary conditions via linear matrix inequalities for the existence of a common quantized controller that achieves our control objectives over the ambiguity ***,the authors provide necessary conditions on the data for the solvability of the LMIs,and determine the coarsest quantization density via semi-definite *** theoretical results are validated through numerical examples.

Rapid detection and discrimination of single photons are pivotal in various applications,such as deep-space laser communication,high-rate quantum key distribution,and optical quantum ***,conventional single-photon detectors(SPDs),including semiconducting and recently developed superconducting detectors,have limited detection speed and photon number resolution(PNR),which pose significant challenges in practical *** this paper,we present an efficient,fast SPD with good PNR,which has 64 paralleled,sandwiched superconducting nanowires fabricated on a distributed Bragg *** detector is operated in a compact Gifford–McMahon cryocooler that supports 64 electrical channels and has a minimum working temperature of 2.3 *** combined detector system shows a functional nanowire yield of 61/64,a system detection efficiency of 90%at 1550 nm,and a maximum count rate of 5.2 ***,it has a maximum PNR of 61,corresponding to the operating *** SPD signifies a substantial improvement in quantum detector technology,with potential applications in deep-space laser communication,high-speed quantum communication,and fundamental quantum optics experiments.

Globally,most organic-rich shales are deposited with volcanic ash *** ash,a source for many sedimentary basins,can affect the sedimentary water environment,alter the primary productivity,and preserve the organic matter(OM)through physical,chemical,and biological *** an increasing number of breakthroughs in shale oil exploration in the Bohai Bay Basin in recent years,less attention has been paid to the crucial role of volcanic impact especially its influence on the OM enrichment and hydrocarbon ***,we studied the petrology,mineralogy,and geochemical characteristics of the organic-rich shale in the upper submember of the fourth member(Es_(4)^(1))and the lower submember of the third member(Es_(3)^(3))of the Shahejie Formation,aiming to better understand the volcanic impact on organic-rich shale *** results show that total organic carbon is higher in the upper shale intervals rich in volcanic ash with enriched light rare earth elements and moderate Eu *** indicates that volcanism promoted OM formation before or after the *** positive correlation between Eu/Eu*and Post-Archean Australian Shale indicates hydrothermal activity before the volcanic *** plane graph of the hydrocarbon-generating intensity(S1+S2)suggests that the heat released by volcanism promoted hydrocarbon ***,the nutrients carried by volcanic ash promoted biological blooms during Es_(4)^(1 )and Es_(3)^(3) deposition,yielding a high primary *** blooms consume large amounts of oxygen and form anoxic environments conducive to the burial and preservation of ***,this study helps to further understand the organic-inorganic interactions caused by typical geological events and provides a guide for the next step of shale oil exploration and development in other lacustrine basins in China.

The Schrodinger cat state produced differently in two directions is anticipated to be a critical quantum resource in quantum information *** exploring the interplay between quantum nonreciprocity and topology in a one-dimensional microcavity array,we obtain the Schrodinger cat state(a pure quantum state)in a chosen direction at the edge cavity,whereas a classical state in the other *** nonreciprocal generation of the cat state originates from the topologically protected chiralitymode excitation in the nontrivial phase,but in the trivial phase,the nonreciprocal generation of cat state ***,our proposal is switchable by tuning the parameters so that a topological phase transition ***,the obtained cat state has nonreciprocal high fidelity,nonclassicality,and quantum coherence,which are sufficient to be used in various one-way quantum technologies,e.g.,invisible quantum sensing,noise-tolerant quantum computing,and chiral quantum *** work provides a general approach to control quantum nonreciprocities with the topological effect,which substantially broadens the fields of nonreciprocal photonics and topological physics.
